Claims (11)
- 培養される細胞を含む液体培地を収容する培養槽と、
矩形断面を有する湾曲流路を有し、前記湾曲流路を流れることによって生じる渦流れを利用して、前記培養槽から供給される液体培地を、相対的に細胞密度が高い富細胞画分と、相対的に細胞密度が低い貧細胞画分に分離する流体力分離装置と、
前記流体力分離装置によって分離される貧細胞画分から細胞を除去して液体培地を回収するフィルタを有するろ過分離装置と
前記流体力分離装置と前記培養槽とを接続し、前記富細胞画分を前記培養槽へ供給する第1返送路と、
前記ろ過分離装置と前記培養槽とを接続し、前記貧細胞画分から除去される細胞を前記培養槽へ供給する第2返送路と、
を有する細胞培養システム。 - 更に、細胞を含む液体培地の一部をブリードとして外部へ排出する排出路を有する請求項1に記載の細胞培養システム。
- 前記排出路は、前記第1返送路から分岐して設けられ、前記富細胞画分の一部または全部を廃棄する請求項2に記載の細胞培養システム。
- 前記ろ過分離装置のフィルタは、デプスフィルタであり、
更に、前記富細胞画分をブリードとして外部へ廃棄する排出路を有する請求項1に記載の細胞培養システム。 - 更に、
新たな液体培地を前記培養槽に供給する培地補給路と、
前記培養槽から供給される液体培地から細胞を除去して液体培地を回収するフィルタを有する追加のろ過分離装置と
を有する請求項4に記載の細胞培養システム。 - 培養される細胞を含む液体培地を収容する培養槽と、
矩形断面を有する湾曲流路を有し、前記湾曲流路を流れることによって生じる渦流れを利用して、前記培養槽から供給される液体培地を、相対的に細胞密度が高い富細胞画分と、相対的に細胞密度が低い貧細胞画分に分離する流体力分離装置と、
前記流体力分離装置と前記培養槽とを接続し、前記貧細胞画分を前記培養槽へ供給することによって、前記培養槽の液体培地の細胞密度を低下させる培地返送路と、
前記培養槽から供給される液体培地から細胞を除去して液体培地を回収するフィルタを有するろ過分離装置と
を有する細胞培養システム。 - 更に、
前記ろ過分離装置と前記培養槽とを接続し、前記ろ過分離装置によって除去された細胞を含む残部の液体培地を前記培養槽へ供給する返送路
を有する請求項6に記載の細胞培養システム。 - 前記流体力分離装置は、前記液体培地を取り入れる単一の導入口と、分離した液体培地を排出する少なくとも2つの導出口とを有し、前記導出口の1つから前記富細胞画分が排出され、もう1つの導出口から前記貧細胞画分が排出される請求項1~7の何れか一項に記載の細胞培養システム。
- 更に、
前記流体力分離装置へ液体培地を供給するための流動圧を液体培地に付勢する付勢装置と、
前記流体力分離装置へ導入される液体培地と、前記流体力分離装置から導出される液体培地との圧力差が所定値以下になるように圧力環境を制御する圧力制御機構と
を有する請求項1~8の何れか一項に記載の細胞培養システム。 - 液体培地で細胞を培養する細胞培養と、
矩形断面を有する湾曲流路を流れることによって生じる渦流れを利用して、前記細胞培養から供給される液体培地を、相対的に細胞密度が高い富細胞画分と、相対的に細胞密度が低い貧細胞画分に分離する流体力分離と、
前記流体力分離によって分離される貧細胞画分から、フィルタを用いて細胞を除去して液体培地を回収するろ過分離と、
前記流体力分離で得られた前記富細胞画分の前記細胞培養への供給と、
前記貧細胞画分から除去される細胞の前記細胞培養への供給と、
を有する細胞培養方法。 - 液体培地で細胞を培養する細胞培養と、
矩形断面を有する湾曲流路を流れることによって生じる渦流れを利用して、前記細胞培養から供給される液体培地を、相対的に細胞密度が高い富細胞画分と、相対的に細胞密度が低い貧細胞画分に分離する流体力分離と、
前記貧細胞画分を前記細胞培養へ供給することによって、前記細胞培養の液体培地の細胞密度を低下させる培地返送と、
前記細胞培養から供給される液体培地から、フィルタを用いて細胞を除去して液体培地を回収するろ過分離と
を有する細胞培養方法。
