Description
本出願は、その内容が全体にわたって引用することにより本明細書の一部をなすものとする、2015年10月16日出願の米国仮特許出願第62/242,611号、および2016年10月6日出願の第62/404,931号による優先権の利益を主張するものである。
(b)哺乳動物肺線維芽細胞を含む間質細胞層、および
(c)哺乳動物内皮細胞(例えば毛細血管内皮細胞)を含む内皮細胞層
を含む人工哺乳動物肺オルガノイドが本明細書で提供される。
多孔質膜の第1の側面に、哺乳動物内皮細胞を含む内皮細胞層を堆積させるステップ、
多孔質膜の第1の側面の反対側である多孔質膜の第2の側面に、哺乳動物肺線維芽細胞を含む間質細胞層を堆積させるステップ、および
間質細胞層に直接、哺乳動物肺上皮細胞を含む上皮細胞層を堆積させるステップ
を含んでいてよい。
哺乳動物肺線維芽細胞を含むヒドロゲルを用意するステップ、
ヒドロゲルの第1の側面に、哺乳動物内皮細胞を含む内皮細胞層を堆積させるステップ、および
ヒドロゲルの第1の側面の反対側であるヒドロゲルの第2の側面に、哺乳動物肺上皮細胞を含む上皮細胞層を堆積させるステップ
を含んでいてよい。
(a)哺乳動物肺上皮細胞を含む上皮細胞層、(b)哺乳動物肺線維芽細胞を含む間質細胞層、および(c)哺乳動物内皮細胞を含む内皮細胞層を含む、人工哺乳動物肺オルガノイドが本明細書で提供される。一部の実施形態で、上皮細胞層は初代哺乳動物肺上皮細胞を含んでいてよい。一部の実施形態で、間質細胞層は初代哺乳動物肺線維芽細胞、または幹細胞もしくは細胞株から分化した哺乳動物肺線維芽細胞を含んでいてよく、内皮細胞層は初代哺乳動物内皮細胞、または幹細胞もしくは細胞株から分化した哺乳動物内皮細胞を含んでいてよい。一部の実施形態で、内皮細胞層は毛細血管内皮細胞を含んでいてもよい。一部の実施形態で、内皮細胞層はヒト臍帯静脈内皮細胞(HUVEC)を含んでいてよい。
膜(例えば多孔質膜)の第1の側面に生哺乳動物内皮細胞(例えば毛細血管内皮細胞)を含む内皮細胞層を堆積させ、
膜の第1の側面の反対側である膜の第2の側面に生哺乳動物肺線維芽細胞を含む間質細胞層を堆積させ、
間質細胞層に直接、生哺乳動物肺上皮細胞を含む上皮細胞層を堆積させる
ことによって調製することができる。
間質細胞(例えば生哺乳動物肺線維芽細胞)をそこに含むヒドロゲルを用意し、
ヒドロゲルの第1の側面に、生哺乳動物内皮細胞(例えば毛細血管内皮細胞)を含む内皮細胞層を堆積させ、
ヒドロゲルの第1の側面の反対側であるヒドロゲルの第2の側面に、生哺乳動物肺上皮細胞を含む上皮細胞層を堆積させる
ことによって調製することができる。
一部の実施形態で、肺オルガノイドはマイクロ流体デバイスとして提供されうる。オルガノイドを支持するのに有用な各種マイクロ流体デバイス構成が、当技術分野で既知となっている。例えば、Ingberらの米国特許出願公開第2014/0038279号;Bhatia and Ingber, “Microfluidic organs-on-chips,” Nature Biotechnology 32:760-772 (2014)を参照のこと。
本明細書に記載される肺オルガノイドは、化合物もしくはワクチンスクリーニング(例えば、有効性、毒性、またはその他の代謝もしくは生理的活性についてのスクリーニング)、または肺感染症もしくは疾患(例えば、慢性閉塞性肺疾患(COPD))の治療(治療に対する抵抗性を含む)についての、生きた動物による実験の代替として使用可能である。急性治療試験では、化合物またはワクチンを例えば数時間に1回適用することができる。慢性治療試験では、化合物またはワクチンを例えば数日~1週間適用することができる。このような試験は、本明細書に記載される肺オルガノイド製品を、その製品の構成細胞を生きたまま維持する条件下(例えば、酸素添加した培養培地)で用意し、試験する化合物(例えば薬物候補)を、(例えば、上皮層への局所適用または気体適用により)肺オルガノイドに適用し、次いで生理応答(例えば、損傷、瘢痕組織形成、感染症、細胞増加、熱傷、細胞死、ヒスタミン放出、サイトカイン放出などのマーカー放出、遺伝子発現変化など)を検出することによって実施可能であり、このような生理応答の存在によって、前記化合物またはワクチンが、哺乳動物対象の肺に吸い込まれるかその他の方法で送達された場合に治療有効性、毒性、またはその他の代謝もしくは生理的活性を有することが示される。対照化合物(例えば、生理食塩水、化合物溶媒または担体)が適用されうる肺オルガノイドの対照試料を同様の条件下で維持することができ、その結果比較結果を得るか、過去データとの比較または薄いレベルの試験化合物の適用により得られるデータとの比較などに基づいて損傷を決定することができる。
一旦産生されると、上記のデバイスは速やかに使用するか、保存および/または輸送用に調製することができる。
ポリエステル膜(Corning(登録商標)Costar(登録商標)Snapwell(商標)細胞培養インサート、12mmで孔0.4μm、孔密度4x106孔/cm2、ポリエステル膜、TC処理済み、滅菌済み)の各側面をコラーゲンIV150μl(Sigma C7521)でコーティングし、バイオセーフティフードで一晩(フード開放および送風機オン)放置した。翌朝、膜を30分間UV滅菌した。
細胞の特性決定:初代肺線維芽細胞は継代の間、線維芽細胞特異的なマーカーの発現を維持し、NHBE細胞は培養中、基底細胞、杯細胞、線毛細胞およびクララ細胞に対するマーカーの発現を維持した。
複数の初代気道細胞型を組み合わせて機能性の上気道オルガノイドを生成した。この生物工学によるオルガノイド系は、ヒト疾患についての研究、毒性研究ならびに薬物およびワクチン開発の実施に有用である。
Claims (6)
- (a)哺乳動物肺上皮細胞を含む上皮細胞層、
(b)哺乳動物肺線維芽細胞を含む間質細胞層、および
(c)哺乳動物内皮細胞を含む内皮細胞層
を含む人工哺乳動物肺オルガノイドであって、
任意選択的に、前記オルガノイドが前記上皮細胞層と前記間質細胞層の間および/または前記間質細胞層と前記内皮細胞層の間に多孔質膜をさらに含み、
前記多孔質膜の少なくとも1つの側面が、肺組織由来の細胞外マトリックス組成物でコーティングされている、人工哺乳動物肺オルガノイドを調製する方法であって、
前記多孔質膜の第1の側面に、前記哺乳動物内皮細胞を含む前記内皮細胞層を堆積させるステップ、
前記多孔質膜の前記第1の側面の反対側である前記多孔質膜の第2の側面に、前記哺乳動物肺線維芽細胞を含む前記間質細胞層を堆積させるステップ、および
前記間質細胞層に直接、前記哺乳動物肺上皮細胞を含む前記上皮細胞層を堆積させるステップ
を含む、方法。 - 前記堆積させるステップが、広げる、塗る、コーティングする、プリントする、バイオプリントする、および/または噴霧することにより実施される、請求項1に記載の方法。
- (a)哺乳動物肺上皮細胞を含む上皮細胞層、
(b)哺乳動物肺線維芽細胞を含む間質細胞層、および
(c)哺乳動物内皮細胞を含む内皮細胞層
を含む人工哺乳動物肺オルガノイドであって、
任意選択的に、前記オルガノイドが前記上皮細胞層と前記間質細胞層の間および/または前記間質細胞層と前記内皮細胞層の間に多孔質膜をさらに含み、
前記多孔質膜の少なくとも1つの側面が、肺組織由来の細胞外マトリックス組成物でコーティングされている、人工哺乳動物肺オルガノイドを調製する方法であって、
前記哺乳動物肺線維芽細胞を含むヒドロゲルを用意するステップ、
前記ヒドロゲルの第1の側面に、前記哺乳動物内皮細胞を含む前記内皮細胞層を堆積させるステップ、および
前記ヒドロゲルの前記第1の側面の反対側である前記ヒドロゲルの第2の側面に、前記哺乳動物肺上皮細胞を含む前記上皮細胞層を堆積させるステップ
を含む、方法。 - 前記堆積させるステップが、広げる、塗る、コーティングする、プリントする、バイオプリントする、および/または噴霧することにより実施される、請求項3に記載の方法。
- 前記ヒドロゲルが架橋結合している、請求項3または4に記載の方法。
- 前記ヒドロゲルが、ゼラチン、フィブリノーゲン、ジェランガム、プルロニック(ポロクサマー)、アルギン酸、キトサン、ヒアルロン酸、セルロースおよび/またはコラーゲンを含む、請求項3から5のいずれか1項に記載の方法。
