A液およびB液を混合した混合液において、ナトリウムイオン濃度が140mEq/L、カリウムイオン濃度が3.7mEq/L、カルシウムイオン濃度が2.6mEq/L、マグネシウムイオン濃度が1.1mEq/L、塩素イオン濃度が115〜120mEq/L、無機リン濃度が3.7mg/dL、炭酸水素イオン濃度が30mEq/L、ブドウ糖濃度が110〜180mg/dL、好ましくは150mg/dLである血液ろ過用補充液、

[4]A液およびB液の容量がいずれも1000mLである上記[1]〜[3]のいずれかに記載の血液ろ過用補充液、
[5]連通可能な隔離手段で区画された少なくとも2室を有する複室容器製剤であって、上記[1]〜[4]のいずれかに記載の血液ろ過用補充液のA液およびB液をそれぞれ第1室および第2室に収容してなる複室容器製剤、
[6]複室容器がプラスチック製ダブルバッグである上記[5]記載の複室容器製剤、
[7]複室容器を脱酸素剤と共にガスバリア性フィルムによりさらに包装した上記[5]または[6]記載の複室容器製剤、および
[8]さらに複室容器外側かつガスバリア性フィルム内側に酸素検知剤を含む上記[7]記載の複室容器製剤
に関する。
表1の組成にしたがい、A液の成分を注射用水に混合・溶解し、pHを調整し、注射用水で全量1000mLとした。同様に、表2の組成にしたがい、B液の有効成分を注射用水に溶解したのち、pHを調整し、最終的に注射用水で全量を1000mLとした。
実施例1および2で得られたダブルバッグ(n=2)をフィルム包装の外袋から取り出し、下室を両手で握るように圧縮し、隔壁を開通した。A液(下室)側およびB液(上室)側を交互に押して良く混合し、その後室温に静置した。安定性の測定は、混合直後、4、12、24および36時間で、性状、pH、純度試験(糖の分解産物である5−ヒドロキシメチルフルフラール(5−HMF)を測定)、不溶性異物、不溶性微粒子の各項目について実施した。各項目の試験方法は、いずれも日本薬局方に規定された試験方法に従うものであり、目標とする規格と共に表3に示す。
実施例1および2で得られたダブルバッグを外装のままさらに二次包装として紙箱に収納し、温度40℃±1℃、湿度75%PH±5%RHで保存した。貯蔵安定性の測定は、保存開始時、保存開始から1、2、3および6ヵ月で、A液およびB液についてそれぞれ表5に示す評価項目について実施した(n=3)。各項目の試験方法は、いずれも日本薬局方に規定された試験方法に従うものであり、目標とする規格と共に表5に示す。
試験例1および2を通じて、5−HMF、炭酸水素ナトリウム、ブドウ糖、カルシウムおよびマグネシウムの定量は、つぎの条件で行った。
検出器:紫外可視分光光度計((株)島津製作所製のUV−2700、測定波長:284nm)
(2)炭酸水素ナトリウム
検出器:自動滴定装置(平沼産業(株)製のCOM−1700S)
(3)ブドウ糖
検出器:高速自動旋光計((株)堀場製作所製のSEPA−200)
(4)カルシウム
検出器:高速液体クロマトグラフィー((株)日立ハイテクノロジーズのL−7000)
(5)マグネシウム
検出器:高速液体クロマトグラフィー((株)日立ハイテクノロジーズのL−7000)
Claims (8)
- 塩化ナトリウム、塩化カリウムおよび炭酸水素ナトリウムを含むA液、ならびに塩化ナトリウム、塩化カリウム、塩化カルシウム、塩化マグネシウムおよびブドウ糖を含むB液からなる用時混合型血液ろ過用補充液であり、A液またはB液のいずれか一方にリン酸イオンを含有しかつ酢酸イオンをいずれにも含有せず、A液およびB液が同容量である血液ろ過用補充液であって、
A液およびB液を混合した混合液において、ナトリウムイオン濃度が140mEq/L、カリウムイオン濃度が3.7mEq/L、カルシウムイオン濃度が2.6mEq/L、マグネシウムイオン濃度が1.1mEq/L、塩素イオン濃度が115〜120mEq/L、無機リン濃度が3.7mg/dL、炭酸水素イオン濃度が30mEq/L、ブドウ糖濃度が110〜180mg/dLである血液ろ過用補充液。 - A液が、塩化ナトリウム4.98g/L、塩化カリウム275.8mg/L、炭酸水素ナトリウム5.04g/Lおよびリン酸水素二ナトリウム十二水和物859.6mg/Lを含有する水溶液であり、
B液が、塩化ナトリウム7.60g/L、塩化カリウム275.8mg/L、塩化カルシウム二水和物382.2mg/L、塩化マグネシウム六水和物223.6mg/Lおよびブドウ糖2.20〜3.60g/Lを含有する水溶液である
請求項1記載の血液ろ過用補充液。 - A液が、塩化ナトリウム5.26g/L、塩化カリウム283.3mg/Lおよび炭酸水素ナトリウム5.04g/Lを含有する水溶液であり、
B液が、塩化ナトリウム7.46g/L、塩化カリウム268.4mg/L、塩化カルシウム二水和物382.2mg/L、塩化マグネシウム六水和物223.6mg/L、リン酸二水素ナトリウム二水和物374.4mg/Lおよびブドウ糖2.20〜3.60g/Lを含有する水溶液である
請求項1記載の血液ろ過用補充液。 - A液およびB液の容量がいずれも1000mLである請求項1〜3のいずれか1項に記載の血液ろ過用補充液。
- 連通可能な隔離手段で区画された少なくとも2室を有する複室容器製剤であって、請求項1〜4のいずれか1項に記載の血液ろ過用補充液のA液およびB液をそれぞれ第1室および第2室に収容してなる複室容器製剤。
- 複室容器がプラスチック製ダブルバッグである請求項5記載の複室容器製剤。
- 複室容器を脱酸素剤と共にガスバリア性フィルムによりさらに包装した請求項5または6記載の複室容器製剤。
- さらに複室容器外側かつガスバリア性フィルム内側に酸素検知剤を含む請求項7記載の複室容器製剤。
