B−231、SKBR−3、T−47D)、ヒト肺癌細胞株(A549、H1975、PC9)、ヒト大腸癌細胞株(HCT−116、HT29、WiDr)及びヒト前立腺癌細胞株(DU−145、PC−3、LNCaP)が挙げられる。
図1に示した態様の分離構造体1を使用して、がん細胞の分離および培養を行なった。分離構造体1は、詳細には、筒状部材2は内径Φ18mm、縦70mm、容量15mLの円筒状のポリプロピレン製部材である。また筒状部材2の先細り形状7の部分の傾斜角度は30°であり、筒状部材2との連通開口6は内径Φ2mmである。筒状部材3は内径Φ10mm、縦54mm、容量3mLのポリプロピレン製部材である。
実施例1と同様に、生物試料として、健常者血液に約100個のヒト肺がん細胞(PC9)を混合して得られた血液試料、生理食塩水及び結合剤の混合液を密度勾配溶液の上へ重層し、遠心分離操作を実施した。実施例2では、前記生物試料を重層し、5分静置した後に遠心分離を実施した。なお、密度差分離における遠心条件は2000×g、5分間、室温にて実施した。なお、がん細胞とともに混入する白血球数は50万個前後であった。
実施例1と同様に、生物試料として、健常者血液に約100個のヒト肺がん細胞(PC9)を混合して得られた血液試料、生理食塩水及び結合剤の混合液を密度勾配溶液の上へ重層し、遠心分離操作を実施した。なお、密度差分離における遠心は2000×g、5分間、回収における遠心は300×g、10分間室温にて実施した。比較例1では、上部筒状部材2からの回収試料に混入した赤血球の破砕処理を、塩化アンモニウム溶液を用いて実施した。
実施例2と同様に、生物試料として、健常者血液に約100個のヒト肺がん細胞(PC9)を混合して得られた血液試料、生理食塩水及び結合剤の混合液を密度勾配溶液の上へ重層し5分静置後、遠心分離操作を実施し、滅菌した純水を用いて赤血球の破砕処理をすることで、細胞懸濁液を回収した。実施例3では、回収した1mLの細胞懸濁液は、以下の(A)から(G)に示す、がん細胞の相互作用に関わる因子(タンパク質)のいずれかで被覆した細胞培養用のマルチウェルプレートに播種し、37℃、5% CO2濃度のインキュベーター内にて培養し、5日毎に培地交換を実施した。なおこれら因子は免疫グロブリンのFc領域との融合体であり、当該Fc領域を介して前記基材固相表面に固定されている。

また培養に用いた、がん細胞の相互作用に関わる因子で被覆したプレートの作製は、前記因子のPBS希釈溶液(10μg/mL)を、無処理ポリスチレン製培養マルチウェルプレートに注ぎ、37℃で一晩コーティング処理し、洗浄後、細胞の非特異的接着を抑えるため、細胞を播種する前に、0.1%(w/v)BSAを含むPBSで1時間のブロッキング処理を行ない、作製した。
実施例2と同様に、生物試料として、健常者血液に約100個のヒト肺がん細胞(PC9)を混合して得られた血液試料、生理食塩水及び結合剤の混合液を密度勾配溶液の上へ重層し5分静置後、遠心分離操作を実施し、滅菌した純水を用いて赤血球の破砕処理をすることで、細胞懸濁液を回収した。比較例2では、回収した1mLの細胞懸濁液は、無処理ポリスチレン製培養マルチウェルプレートに播種し、37℃、5% CO2濃度のインキュベーター内にて培養し、5日毎に培地交換を実施した。
実施例2と同様に、生物試料として、健常者血液に約100個のヒト肺がん細胞(A549)を混合して得られた血液試料、生理食塩水及び結合剤の混合液を密度勾配溶液の上へ重層し5分静置後、遠心分離操作を実施し、滅菌した純水を用いて赤血球の破砕処理をすることで、細胞懸濁液を回収した。当該がん細胞は、細胞密度が約2×105個/cm2になるように静置し、5ng/mLのTGF−βを添加して培養後、0.25%トリプシン/1mM EDTAにより細胞をディッシュから剥離し、限界希釈により調整したものである。実施例4では、回収した1mLの細胞懸濁液は、以下の(H)および(I)に示す、がん細胞の相互作用に関わる因子(タンパク質)のいずれかで被覆したマルチウェルプレートに播種し、37℃、5% CO2濃度のインキュベーター内にて5ng/mLのTGF−βを添加して培養し、5日毎に培地交換を実施した。

実施例1と同様にして、生物試料として、健常者血液に約1×106個のヒト肺がん細胞(PC9)を混合して得られた血液試料、生理食塩水及び結合剤の混合液を密度勾配溶液の上へ重層後、遠心分離操作を実施し、滅菌した純水による赤血球の破砕処理後、Hanks’溶液を添加して遠心分離した。遠心分離後、ペレットの頂部の液体をピペットで取り出し、最終液量を25μLとした。前記溶液に、マトリゲル25μLを添加した細胞懸濁液を、ヌードマウスであるKSN/Slcマウス(6週齢、オス、日本エスエルシー社製)の皮下に全量接種した。接種したがん細胞の増殖は、腫瘍形成部の体積(長径×長径×短径÷2)を基に評価した。
実施例5に記載の赤血球の破砕処理を、塩化アンモニウム溶液を用いて実施した他は、実施例5と同様な方法で、マウスに接種したがん細胞の増殖を評価した。

Claims (4)
- 血液試料中の腫瘍細胞の分離方法であって、血液試料から腫瘍細胞を濃縮して濃縮液を得る第一工程と、前記第一工程で得られた濃縮液を純水で処理する第二工程と、を備えることを特徴とする、前記分離方法。
- 前記第一工程が、腫瘍細胞と腫瘍細胞以外の成分との間の比重差を用いて濃縮する工程であることを特徴とする、請求項1に記載の分離方法。
- 前記第二工程の後、腫瘍細胞を培養することを特徴とする、請求項1又は2に記載の分離方法で分離した腫瘍細胞の培養方法。
- 腫瘍細胞を実験動物に移植して培養することを特徴とする、請求項3に記載の培養方法。
