Description
本出願は、米国特許法第119条(e)の下で、2014年2月19日に出願された米国出願第61/941,896号の優先権を主張し、この仮出願は、その全体が参照により組み込まれる。

技術分野
本発明は、p97(メラノトランスフェリン)及びイズロン酸−2−スルファターゼ(IDS)の融合タンパク質、ならびにその使用に関連する組成物及び方法に関し、例えば、血液脳関門(BBB)を越えるIDSの送達を促進し、かつ/またはCNS及び末梢組織におけるその組織浸透を向上させ、それによって中枢神経系(CNS)構成要素を有するものを含む、ハンター症候群(ムコ多糖症II型、MPS II型)及び関連するリソソーム蓄積症を治療及び/または診断する方法に関する。
リソソーム蓄積症(LSD)は、細胞のリソソーム内の特定の酵素またはタンパク質の欠如またはそれらの低下した活性に起因する。細胞内において、欠けている酵素活性の影響は、細胞内リソソーム内の分解されていない「蓄積材料」の蓄積として見出すことができる。この蓄積によってリソソームは膨潤し、かつ機能不全を起こし、細胞損傷及び組織損傷をもたらす。リソソーム蓄積症は典型的には遺伝的な病因をもつため、多くの組織が問題の酵素を欠くことになる。しかしながら、異なる組織は、同一の酵素活性の欠如を異なる形で被る。組織がどれだけ不都合に影響を受けるかは、その組織が欠けている酵素の基質を生成する度合いにある程度依存して決定される。蓄積によって最も負担を受ける組織の種類は、ひいては、薬物がどのように患者に投与されるべきかを規定することになる。
本発明は、例えば以下の項目を提供する。

融合タンパク質のインビトロ活性
ヒトp97(メラノトランスフェリン、MTf)及びヒトイズロン酸−2−スルファターゼ(duronate−2−sulfatase)(IDS)の融合タンパク質を調製し、インビトロの酵素活性について試験した。表E1はアミノ酸配列を提供し、表E2は調製及び試験した融合タンパク質の対応するポリヌクレオチドコード配列を提供する。
脳内でのI2S−MTf及びMTfpep−I2S融合物のインビボ分散
マウスにおけるI2S−MTf及びMTfpep−I2S融合タンパク質の脳生体内分布を、定量的共焦点顕微鏡イメージングで評価した。I2S−MTf及びMTfpep−I2Sの治療量等価物を、尾静脈注射でマウスに100μL投与した。安楽死させる前に、マウスには、脳の血管系を染色するために、10分間、トマトレクチン−FITC(40μg)を注射した(静脈内)。血液は、10mlのヘパリン添加した食塩水を毎分1mlの速度で心臓内灌流させることで取り除いた。脳を切除し、OCT内で凍結させ、−80℃で保管した。Tissue Tekに脳を載置し、−20℃のクリオスタットで薄片に切り分けた。薄片をSuperfrost Plus顕微鏡スライド上に載置し、室温で10分間、低温のアセトン/MeOH(1:1)中で固定し、PBSで洗浄した。Prolong Gold褪色防止用封入剤を、DAPI(分子プローブ、P36931)と共に使用して、カバーガラスを薄片上に載置した。三次元(3D)共焦点顕微鏡検査及び定量的分析を行った。
MPS II型のマウスモデルにおけるI2S−MTf及びMTfpep−I2S融合物のインビボ活性
I2S−MTf及びMTfpep−I2S融合タンパク質の治療効果を、ハンター症候群の治療に対して示されるイズルスルファーゼ(Elaprase(登録商標))と比較して、ハンター症候群またはムコ多糖症II型(MPS II型)のマウスモデルにおいて評価する。これらの研究は、ムコ多糖症II型(MPS II型)のノックアウトマウスモデルにおける脳病理に対する、これらの融合タンパク質の静脈内(IV)及び腹腔内(IP)投与の効果を評価するように設計される。

CA−630、2mMのPefabloc SC)中で組織を均質化することによって調製する。次いで、破砕物を、エタノール/乾燥氷浴及び37℃の水浴を用いる5回の凍結融解サイクルに供する。組織破片を室温、2000gでの12分間の遠心分離によって2度ペレット化し、上清を収集し、ビシンコニン酸(bicinchonic acid)(BCA)アッセイ(Pierce,Rockford,IL)で、総タンパク質濃度(mg/mL)についてアッセイする。

- p97ポリペプチドフラグメントのN末端に融合したイズロン酸−2−スルファターゼ(IDS)ポリペプチドと、それらの間の任意選択の異種ペプチドリンカー(L)とを含むp97(メラノトランスフェリン)融合タンパク質であって、前記p97ポリペプチドフラグメントは、1つ、2つまたは3つの追加の末端アミノ酸残基を伴うアミノ酸配列DSSHAFTLDELR(配列番号14)からなり、かつ、輸送活性を有する、p97(メラノトランスフェリン)融合タンパク質。
- 前記p97ポリペプチドフラグメントが、2つの追加の末端アミノ酸残基を伴うアミノ酸配列DSSHAFTLDELR(配列番号14)からなり、かつ輸送活性を有する、請求項1に記載のp97(メラノトランスフェリン)融合タンパク質。
- 前記p97ポリペプチドフラグメントが、1つの追加の末端アミノ酸残基を伴うアミノ酸配列DSSHAFTLDELR(配列番号14)からなり、かつ輸送活性を有する、請求項1に記載のp97(メラノトランスフェリン)融合タンパク質。
- 前記追加の末端アミノ酸残基の各々がシステイン残基またはチロシン残基である、請求項1に記載のp97(メラノトランスフェリン)融合タンパク質。
- 前記追加の末端アミノ酸残基の各々がチロシン残基である、請求項1に記載のp97(メラノトランスフェリン)融合タンパク質。
- 前記追加の末端アミノ酸残基の各々がシステイン残基である、請求項1に記載のp97(メラノトランスフェリン)融合タンパク質。

- 前記ペプチドリンカーが、剛性リンカー、可動性リンカー、及び酵素的に切断可能なリンカーのうちの1つ以上から選択される、請求項10に記載のp97(メラノトランスフェリン)融合タンパク質。
- 前記リンカーが、アミノ酸配列(EAAAK)1〜3(配列番号36〜38)を有する、請求項1に記載のp97(メラノトランスフェリン)融合タンパク質。
- 前記リンカーが、アミノ酸配列(EAAAK)3(配列番号38)を有する、請求項12に記載のp97(メラノトランスフェリン)融合タンパク質。
