Description
(1) 清潔な環境で外装を開封し、アルミパックから滅菌紙包装を取り出す。
(2) 無菌的に滅菌紙から本体を取り出し、無菌容器に入れておく。
(3) 処置をする患部の大きさにあうだけのビトリゲルの大きさに見合った個数を準備する。
(1) 患部のデブリードマンを十分に施したあと、コラーゲンビトリゲル乾燥膜側の剥離紙をはぎ、その面を患部に当て、患部を包み込むように合わせる。
(2) 患部がすべて覆われた状態で、動かないように押えたまま粘着性フィルムの上部にあるポリプロピレンフィルムを無菌的に剥がし、しっかり押さえる。
(3) 患部全体がコラーゲンビトリゲル乾燥膜に覆われた状態を確認し、湿潤環境ができていることも確認する。
(4) 本発明人工皮膚と創傷面との間に気泡がないことを確認する。もし気泡が存在したら指で気泡を外に誘導する。
(5) 湿潤液があふれ、粘着性フィルムがつかない場合は、止血とデブリードマンをやり直し、再度コラーゲンビトリゲル乾燥膜を施す。
(6) 1週間〜2週間、患部の状態を観察しながら剥がすタイミングを探る。患部に残すのはコラーゲンビトリゲル薄膜のみであり、押える役割の額縁状保持シートや、接着防止シート、粘着性フィルムはすべて除去する。
広範囲皮膚欠損治療効果:
野生型マウス(C57BL6J)の背部に、直径1.5cmの広範囲皮膚全層欠損を作製し、皮膚欠損創傷モデルとした。その創傷部に本発明の人工皮膚または被覆材を貼付し、創傷部での瘢痕収縮抑制効果の検証を行った。

- 少なくとも粘着性フィルム、接着防止シートおよびコラーゲンビトリゲル膜乾燥体をこの順で含み、前記接着防止シートは、前記コラーゲンビトリゲル膜乾燥体が粘着性フィルムの粘着層に付着することを防止するのに十分な大きさで、コラーゲンビトリゲル膜乾燥体の位置と対応する前記粘着性フィルムの粘着剤層に貼付されてなり、前記コラーゲンビトリゲル膜乾燥体は、貼付時は、前記接着防止シート面に固定、保持されながら、前記粘着フィルムの剥離時には前記接着防止シートから脱着容易とされた貼付型人工皮膚製剤であって、貼付時の前記コラーゲンビトリゲル膜乾燥体の前記接着防止シート面への固定、保持が、次の(1)または(2)のいずれかの手段、
(1)前記コラーゲンビトリゲル膜乾燥体の下に設けられ、その外周部で前記粘着性フィ
ルムの粘着剤層と接着された、窓部を有する額縁状保持シートの窓部の内周部もしく
はこれに設けられた突起による固定、保持、
(2)前記コラーゲンビトリゲル膜乾燥体の、弱い接着力により前記接着防止シートに貼
付することによる固定、保持。 - 接着防止シートが片面に剥離処理を施したプラスチックシートである請求項1に記載の貼付型人工皮膚製剤。
- 剥離処理を施したプラスチックシートが、シリコーンコーティングを施したポリエチレンテレフタレートシートである請求項2記載の貼付型人工皮膚製剤。
- コラーゲンビトリゲル膜乾燥体が、アテロコラーゲンビトリゲル膜乾燥体である請求項1ないし3の何れかの項記載の貼付型人工皮膚製剤。
- 皮膚欠損部に容易に貼りつけることが可能であり、貼り換えの際にもコラーゲンビトリゲル膜が粘着性フィルムおよび接着防止シートに接着することなく創傷部に遺残することで、二次損傷を防止する機能を付与することを特徴とする請求項1ないし4の何れかの項記載の貼付型人工皮膚製剤。
- 創傷部の湿潤環境を粘着性フィルムが保持しつつ、コラーゲンビトリゲル膜乾燥体に含まれるコラーゲン成分が表皮細胞へ足場を供給しつつ、筋線維芽細胞の出現を抑制することで瘢痕形成を抑制して早期の治療を達成することを特徴とする請求項1ないし5の何れかの項記載の貼付型人工皮膚製剤。
- コラーゲンビトリゲル膜乾燥体が、3型コラーゲンを含むアテロコラーゲンより作成されたものであり、コラーゲンビトリゲル膜が創傷部へ移行後も長時間遺残し、かつ異物反応を惹起しないことを特徴とする請求項1ないし6の何れかの項記載の貼付型人工皮膚製剤。
- コラーゲンビトリゲルが含有する3型コラーゲンの作用より、創傷部周辺の線維芽細胞におけるTGF−βとCTGFの発現を抑制することにより、線維芽細胞から筋線維芽細胞への分化転換を抑制することを特徴とする請求項7記載の貼付型人工皮膚製剤。
