Description
酸化ストレス誘発剤として鉄ニトリロ三酢酸(Fe−NTA)を用いたラット酸化ストレスモデルを用いて、各被験物質を単回強制経口投与した際の血清中過酸化脂質量をチオバルビツール酸反応物(TBARS)量として定量することで各被験物質の抗酸化作用を調べた。
被験物質として、黒生姜エキス及び/又はビタミンEを表1に示す配合比で混合し被験物質溶液を調製した。調製方法は、以下のとおりである。なお、本件等で使用される黒生姜エキス(黒生姜抽出物の精製粉末)及びビタミンEは市販品をそのまま使用した。
(試験前日から)絶食したラットを試験日の体重値が各群でほぼ均一となるように各群に分け、試験に供した。試験日の体重値に基づき各被験物質溶液を単回強制経口投与した。対照例1と対照例2では、溶媒である1%Tween80水溶液を単回強制経口投与した。投与液量は、10mL/kgとした。
蒸留水を溶媒として、硝酸鉄(III)九水和物とニトリロ三酢酸二ナトリウム塩を、それぞれモル比として1:4となるように溶解し、炭酸水素ナトリウムを用いてpH7.4になるよう調整してFe−NTA溶液とした。
被験物質の投与1時間後に、試験日の体重値に基づきFe−NTA溶液を9mgFe/kg(10mL/kg)にて腹腔内投与した。対照例1は、Fe−NTA溶液の代わりに、生理食塩水を10mL/kgにて腹腔内投与した。
Fe−NTA投与2時間後に採血し、室温で30分間以上放置した後、遠心分離を行い、血清を採取した。採取した血清は−30℃下にて凍結保存を行った。
得られた血清を用いて、TBARS Assay Kit(CaymanChemical Company)にて測定を行った。
さらに、上記の黒生姜エキス粉末及びビタミンE、並びに下記要領で得た黒生姜粉砕物を用い、下記表の処方で飲料、顆粒、ソフトカプセルを作成した。
黒生姜の根茎を洗浄後、1〜10mm程度にスライスし、1日天日干しにした。その後、40〜100℃に設定したオーブン乾燥機で4〜6時間乾燥し、粗粉砕後、130〜200℃で5〜20秒間殺菌を行った。殺菌した粗粉砕物を粉砕機によって粉砕し、黒生姜粉砕物を得た。
表2の配合割合で各成分を配合し、液剤とした。
表3に従い、スクラロース0.3kg、クエン酸3kg、還元麦芽糖水飴36.9kg、難消化性デキストリン48kg、カンゾウ抽出物0.3kg、黒生姜粉砕物0.5kg、ビタミンE3kg、シクロデキストリン5kg、ショウヤク3kgをフローコーターNFLO−200型流動層造粒機(フロイント産業(株)製)に投入し、数分間気流で混合する。これに、水60Lを1分間に2000mL噴霧することにより造粒を行った。つづいて、得られた造粒物を30メッシュの篩いにて篩別し顆粒剤とした。
実施例3と同様にして、表4の成分を配合し顆粒剤とした。
表5の配合割合で配合した内容液を調整し、表6の配合割合で配合したカプセル皮膜に充填することでソフトカプセルとした。カプセル化は、カプセル皮膜液を流延しフィルム化すると共に、内部に内容液を充填しヒートシールし、成形されたソフトカプセルを乾燥させて行った。
表7の配合割合で配合した内容液を調整し、表8の配合割合で配合したカプセル皮膜に充填し、実施例5と同様にしてソフトカプセルとした。
