(I)リンデラノリドB
(II)スバモリドA
およびその塩類、エステル類、または溶媒和物を含む。個体は動物またはヒトを含む。
(I)リンデラノリドB
(II)スバモリドA
およびその塩類、エステル類、または溶媒和物を含む。
キナモムム・スバベニウム(Cinnamomum subavenium)は、ニッケイ属(Cinnamomum)に属し、台湾原生の常緑高木の1種である。主に海抜500〜1500メートルの間に分布している。その樹皮はニッケイの香りを有し、香料として使用される。葉から抽出した葉油は、食品、タバコの香料とされ、または殺菌剤として使用される。
キナモムム・スバベニウム(Cinnamomum subavenium)の茎から2種の化合物、リンデラノリドBおよびスバモリドAを分離した。以下化合物(I)および(II)と略す。

(II)スバモリドA
本実施例は、ヒトの皮膚のメラニン細胞を用いて、2つの化合物がチロシナーゼを抑制する効果を測定した。
初代ヒト新生児メラニン形成細胞(neonatal foreskin primary human epidermal melanocytes、HEMn−MP)は、Cascade Biologics(登録商標)から購入し、培地254(Cascade Biologics(登録商標))で培養した。ヒトメラニン細胞増殖添加剤(human melanocyte growth supplement、HMGS、cat.No.S−002−5)を添加し、37℃の湿潤環境(5%二酸化炭素)下、初代ヒト新生児メラニン形成細胞(HEMn−MP)を、24ウェル培養プレートを用いて、細胞密度は105個/ウェルで培養した。
異なる濃度の化合物(I)および(II)(0.01μM、0.1μM、1μM、5μMおよび10μM)、コウジ酸(kojic acid)100μM、N−フェニルチオ尿素phenylthiourea(PTU)100μM、メラニン細胞刺激ホルモンα−Melanocyte−Stimulating Hormone(MSH)0.1μMを細胞培養プレートに添加し、MTSアッセイ(Mosmann T.、Rapid colorimetric assay for cellular growth and survival:application to proliferation and cytotoxicity assays、J Immunol Methods、1983年、16、65(1〜2)、55〜63ページ)によって、細胞毒性試験を行った。コウジ酸およびN−フェニルチオ尿素が、メラニン阻害剤であることはすでに知られており、メラニン細胞刺激ホルモンはメラニン生成を刺激する。ここで同時に実験を行い、化合物(I)および(II)の効果との比較に用いた。実験結果は、表1および図2Aに示すとおりである。コントロール群のデータを100%とし、結果はすべて、3回以上の独立した実験によって得られた。
Yoshimuraが行った方法(Yoshimura他、Effects of all−trans retinoic acid on melanogenesis in pigmented skin equivalents and monolayer culture of melanocytes、2001年、J.Dermatol.Sci.、27 Suppl1、S68〜75ページ)に基づき、ドパクロム(dopachrome)生成速度を利用して、チロシナーゼ活性を測定した。HEMn−MP細胞(105個/ウェル)を24ウェル培養プレートで培養し、すべてのウェルに500μlDMEM(Dulbecco’s modified Eagle’s medium)培養液を加え、実験する化合物(I)、(II)、コウジ酸、N−フェニルチオ尿素およびメラニン細胞刺激ホルモンをそれぞれ加えて、3日間培養した。
Linの方法によって、メラニンを測定した(Lin他、Constituents from the Formosan apple reduce tyrosinase activity in human epidermal melanocytes、2007年、Phytochemistry、68巻、8号、1189〜1199ページ)。細胞を1N水酸化ナトリウムおよびジメチルスルホキシド(Dimethylsulfoxide、DMSO)に溶解し、1時間80℃で加熱した。10,000rpmで10分間遠心し、上澄液を吸光度405ナノメートルで測定した。結果は表3および図2Cのとおりである。コントロール群のデータを100%とし、結果はすべて3回以上の独立した実験により得られた。化合物(I)および(II)濃度の増加に伴い、細胞のメラニン含量が低下した。コウジ酸およびN−フェニルチオ尿素も、メラニン含量を減少させる効果を示したが、メラニン細胞刺激ホルモンはメラニン含量を増加させた。
本実施例は、ゼブラフィッシュの生体試験を利用して、2つの化合物の美白効果を、またその投与量がゼブラフィッシュに対して有害性または致死性がないことを検査した。異なる濃度の2つの化合物(1μM、5μMおよび10μM)、N−フェニルチオ尿素10μM、アルブチン(arbutin)50μMを、それぞれゼブラフィッシュ胎児の培養環境に加えた。すべての実験群は4〜9個の胎児を含む。図4からわかるように、10μMの2つの化合物はゼブラフィッシュの色素を20%〜30%減少させた。またゼブラフィッシュに対して毒性を示さなかったが、その効果は濃度が比較的高いN−フェニルチオ尿素およびアルブチン(arbutin)にわずかに劣った。

Claims (4)
- 色素沈着を抑制する組成物であって、以下の化学構造(I)または(II)の化合物、
を有効成分として含むことを特徴とする、組成物。 - チロシナーゼ活性を抑制する作用を有することを特徴とする、請求項1に記載の組成物。
- 飲料、化粧品または医薬品に添加されることを特徴とする、請求項1に記載の組成物。
- 該飲料が各種ジュース、茶類、麺類、乳製品、または調味料を含み、該化粧品が化粧水、ゲル、ローション、クリーム、軟膏、乳液、乳液状もしくはクリーム状のファンデーション、口紅、おしろい、洗顔フォーム、ヘアローションなどの液体、固体、ゾル体を含み、該医薬品が粉、カプセル、噴霧剤、ゲル、軟膏、タブレット、座薬、顆粒、パッチ、丸薬、トローチ、注射薬、点滴薬、懸濁剤、静脈内乳剤、徐放製剤、放出制御製剤を含むことを特徴とする、請求項3に記載の組成物。
