Description
アレルギー性鼻炎の被験者ボランティア3名に、20mg/kgのALA塩酸塩を経口投与し、約6時間後に蛍光観察可能な気管支鏡(PENTAX社製 SAFE−3000)を用いて鼻腔に408nmの紫色光を照射し、ALA−PDDを行った。その結果、PpIXが取り込まれた鼻粘膜炎症部位が赤色蛍光として検出された(図1)。かかる鼻粘膜炎症部位に、635nmの波長を有する赤色LED(Light Emitting Diode)(図2)の光を赤色蛍光が肉眼的に観察できなくなるまで照射した(図3)。より具体的には、635nmの波長を有する赤色LED光を一定時間照射後にかかるPDT照射を中断し、照射装置と入れ替えで気管支鏡(PENTAX社製 SAFE−3000)を鼻腔内に挿入し、赤色光を観察した後、再度PDT照射を行った。その結果、照射後数日までは、激しい鼻水やくしゃみの症状が被験者3名に認められたが、かかる症状はその後数週間にわたり軽減した。これらの結果は、炎症を起こした鼻粘膜に蓄積したPpIXが励起され、活性酸素が生成された結果、かかる活性酸素が鼻粘膜上皮を変性することによりアレルギー反応性の低下をもたらしたことを示している。
次に投与するALA塩酸塩が、塗布による液体タイプでもアレルギー性鼻炎が治療できるかどうかを検討した。成人男性4名の花粉症被験者に対し、5重量%ALA塩酸塩生食溶解液を十分浸した綿で鼻腔内に塗布し、1時間留置させた後にかかる浸漬綿を除去した。3名の被験者には浸漬綿を除去した直後に、残りの1名の被験者には浸漬綿を除去した1時間後に、それぞれ635nmの波長を有する赤色半導体レーザー光100Jを鼻腔内に6分間照射した。その結果、4名の花粉症による激しい鼻水やくしゃみの症状は、1時間後に照射した1名では消失し、直後に照射した3名では軽減した。この結果は、投与するALA塩酸塩が、塗布による液体タイプでもアレルギー性鼻炎が治療できることを示している。さらに、かかる液体タイプの塗布による治療は、鼻粘膜炎症部位周辺に直接ALA塩酸塩溶液を塗布することにより、経口投与よりも投与するALA塩酸塩の量を少なく抑えることができ、また、短時間でPpIXが鼻粘膜炎症部位に蓄積されることから、費用対効果や時間対効果の点から考慮すると、経口投与よりも優れた治療であることを示している。
Claims (5)
- 5−アミノレブリン酸(ALA)若しくはそのエステル誘導体、又はそれらの塩を有効成分として含有し、400nm〜700nmの波長の光を照射する5−アミノレブリン酸−光線力学的療法(ALA−PDT)のためのアレルギー性鼻炎の治療剤。
- 625nm〜640nmの波長の光を照射することを特徴とする請求項1記載のアレルギー性鼻炎の治療剤。
- 経口投与することを特徴とする請求項1又は2記載のアレルギー性鼻炎の治療剤。
- 鼻腔内に経粘膜投与することを特徴とする請求項1又は2記載のアレルギー性鼻炎の治療剤。
- 鼻腔内に経粘膜投与する治療剤の剤型が、塗布される溶液タイプ、水溶性軟膏溶解タイプ、又はゼリー剤溶解タイプであることを特徴とする請求項4記載のアレルギー性鼻炎の治療剤。
