Description
で示される4−{3−[cis−ヘキサヒドロシクロペンタ[c]ピロール−2(1H)−イル]プロポキシ}ベンズアミド、又は薬学的に許容しうる酸もしくは塩基とのその付加塩と、NMDA(N−メチル−D−アスパラギン酸)グルタミン酸作動性受容体アンタゴニストとの新規な組み合わせに関する。
◆ 希釈剤として: 乳糖、デキストロース、ショ糖、マンニトール、ソルビトール、セルロース、グリセリン、
◆ 滑沢剤として: シリカ、タルク、ステアリン酸ならびにそのマグネシウム塩及びカルシウム塩、ポリエチレングリコール、
◆ 結合剤として: ケイ酸アルミニウムマグネシウム、デンプン、ゼラチン、トラガカント、メチルセルロース、ナトリウムカルボキシメチルセルロース及びポリビニルピロリドン、
◆ 崩壊剤として: 寒天、アルギン酸及びそのナトリウム塩、発泡性混合物。
1000錠剤の調剤処方; それぞれ、4−{3−[cis−ヘキサヒドロシクロペンタ[c]ピロール−2(1H)−イル]プロポキシ}ベンズアミド塩酸塩5.63mg(塩基当量5mgに相当する)及びメマンチン塩酸塩10mgを含有している:
4−{3−[cis−ヘキサヒドロシクロペンタ[c]ピロール−2(1H)−イル]プロポキシ}ベンズアミド塩酸塩 5.63g
メマンチン塩酸塩 10g
トウモロコシデンプン 20g
マルトデキストリン 7.5g
コロイド状シリカ 0.2g
グリコールデンプンナトリウム 3g
ステアリン酸マグネシウム 1g
乳糖 55g
エピソード記憶のモデルにおける実験、文脈連続識別テスト:
それぞれ単独で又は組み合わせて投与される、4−{3−[cis−ヘキサヒドロシクロペンタ[c]ピロール−2(1H)−イル]プロポキシ}ベンズアミド及びメマンチン(両方とも塩酸塩の形態で)の効果を、中年(14〜15か月齢)C57B16マウス(1群につきn=12匹)で文脈識別テストを使用して検討した(Celerier et al., Learn Mem., 2004, 11(2), 196-204; Tronche et al., Behav. Brain Res., 2010, 215(2): 255-60)。このモデルにおいて、中年マウスは、空間記憶の欠損は無いが、若年マウスに比較して文脈エピソード記憶の特異的機能障害を有している。このモデルは、アルツハイマー病の製剤の効果を評価するために適切であり、なぜならアルツハイマー型認知症を患っている患者は、文脈エピソード記憶の障害も有しているからであり、このことはかなり初期の段階から該当する(Gold and Budson, Expert Rev Neurother., 2008, 8(12): 1879-1891)。
− 正解反応のパーセンテージ(すなわち、白色の床で学習訓練中に餌付けされた穴に頭を下げ入れる%)、
− 妨害反応のパーセンテージ(すなわち、黒色の床(マウスに提示された最後の文脈)で学習訓練の間に餌付けされた穴に頭を下げ入れる%)、
− そして間違いのパーセンテージ(すなわち、白色の床又は黒色床にかかわらず、学習中に餌付けされなかった2つの穴に頭を下げ入れる%(図1を参照のこと)。
Claims (12)
- 4−{3−[cis−ヘキサヒドロシクロペンタ[c]ピロール−2(1H)−イル]プロポキシ}ベンズアミドを、シュウ酸塩又は塩酸塩の形態で用いることを特徴とする、請求項1記載の組み合わせ。
- メマンチンを塩酸塩の形態で用いる、請求項1〜2のいずれか一項に記載の組み合わせ。
- (i)4−{3−[cis−ヘキサヒドロシクロペンタ[c]ピロール−2(1H)−イル]プロポキシ}ベンズアミドを、塩酸塩の形態で2mg、5mg又は20mg(塩基当量で表した)の1日量で投与し、そして(ii)メマンチンが10mg又は20mgの1日量で投与されるメマンチン塩酸塩である、医薬としての使用のための、請求項1〜3のいずれか一項記載の組み合わせ。
- 活性成分として、4−{3−[cis−ヘキサヒドロシクロペンタ[c]ピロール−2(1H)−イル]プロポキシ}ベンズアミド、又は薬学的に許容しうる酸もしくは塩基とのその付加塩を、請求項1〜3のいずれか一項記載のメマンチンと組み合わせて、1つ以上の薬学的に許容しうる賦形剤と組み合わせて含む、医薬組成物。
- 脳の加齢及び神経変性疾患に関連する認知障害の処置における使用のための、請求項5記載の医薬組成物。
- アルツハイマー病に関連する認知障害の処置における、請求項6記載の使用のための医薬組成物。
- 塩酸塩の形態で、4−{3−[cis−ヘキサヒドロシクロペンタ[c]ピロール−2(1H)−イル]プロポキシ}ベンズアミドの2mg、5mg又は20mg(塩基当量で表した)、及びメマンチンとしてメマンチン塩酸塩の10mg又は20mgを含むことを特徴とする、請求項5記載の医薬組成物。
- 脳の加齢及び神経変性疾患に関連する認知障害の処置を目的とする医薬の製造における、請求項1〜4のいずれか一項記載の組み合わせの使用。
- アルツハイマー病に関連する認知障害の処置を目的とする医薬の製造における、請求項1〜4のいずれか一項記載の組み合わせの使用。
- アルツハイマー病に関連する認知障害の処置における使用のための、4−{3−[cis−ヘキサヒドロシクロペンタ[c]ピロール−2(1H)−イル]プロポキシ}ベンズアミド、又は薬学的に許容しうる酸もしくは塩基とのその付加塩と、メマンチンとの、請求項1〜4のいずれか一項記載の組み合わせ。
- アルツハイマー病に関連する認知障害の処置を目的とする医薬の製造における、4−{3−[cis−ヘキサヒドロシクロペンタ[c]ピロール−2(1H)−イル]プロポキシ}ベンズアミド、又は薬学的に許容しうる酸もしくは塩基とのその付加塩と、メマンチンとの、請求項1〜4のいずれか一項記載の組み合わせの使用。
