Description
分野: 本発明は一般的に薬剤学的調剤(pharmaceutical formulation)の分野に関する。より具体的には、本発明はT細胞(PHA−芽細胞)を選択的に活性化可能でありそして、非常に好ましくは、ナチュラルキラー(「NK」)細胞の減じられた活性化を示すIL−2ムテインを含む安定化させた治療上活性なインターロイキン−2調剤に関する。好ましい性質を有する安定化させた組成物は以下に記載されているIL−2の変種を含む。
本発明は、ヒスチジンで安定化させたIL−2またはその変種(ムテイン類)の薬剤学的組成物または調剤である。好ましくは、組成物は低いイオン強度(例えば<0.1)の溶液を生ずる混合物を含んでなりそして他の安定剤、例えば糖類およびアミノ酸類、好ましくはスクロースおよびグリシン、を含む。調剤は0〜0.9重量%のNaClを含むことができる。組成物はアルブミンを含まずそして凍結乾燥形態での調剤は水を用いて急速(<1分間)に再形成することができる。組成物は生理学的に許容可能なpH条件下で、好ましくは約5.0〜約6.5の範囲のpHにおいて、界面活性剤、例えばドデシル硫酸ナトリウムを使用せずに溶解する。再形成された溶液はほぼ等張性であり且つ皮下および静脈内の両方で投与することができる。非常に好ましい態様では、組成物のIL−2は単一アミノ酸置換を有するムテイン、好ましくはWO 99/60128に記載された変種、である。
IL−2 0.1−0.5重量%
ヒスチジン 0.08−1.6重量%
NaCl 0−0.9重量%
スクロース 1−10重量%および
グリシン 0−3重量%
を水性形態で(重量/重量基準で)5〜6.5のpHにおいて含んでなる。
ここで使用される用語IL−2は、活性な野生型IL−2およびその生物学的に活性な変種またはムテイン類、例えばWO 99/60128に記載されているもの、の両方を包含する。以下の実施例では、我々はアミノ酸蛋白質88の位置でアルギニン(R)に突然変異したアスパラギン(N)を有するヒトIL−2の組み換えムテインであるIL−2(N88R)として知られるIL−2を使用した。このムテインはチャイニーズ・ハムスターの卵巣(CHO)細胞から発現されそしてグリコシル化されたおよびグリコシル化されていない両方の形態の混合物を含んでなっていた。それは以上で関連出願として引用されたWO 99/60128に記載されている。
種々のpHにおける液体IL−2(N88R)の安定性を40℃において試験した。結果は、IL−2(N88R)の沈澱速度がpH5.0〜6.5の間で最低であったことを示した(図1)。従って、pH5.5が液体状態における最適な調剤pHとしてそして凍結乾燥調剤の製造用に選択された。
我々はIL−2(N88R)の安定性に対する種々の緩衝剤の可能な影響を試験した。我々が試験した緩衝剤はクエン酸塩、酢酸塩、およびヒスチジンを含んでなっていた。これらの緩衝剤は1mg/mlのIL−2(N88R)および150mM(0.9重量%)のNaClを含有するIL−2(N88R)溶液中20mMで使用された。これらの安定性試料を、UV/VIS分光光度法により監視しながら、25℃から95℃に毎分1℃で加熱した。ヒスチジンは酢酸塩およびクエン酸塩と比べて、沈澱温度を高めることによりそして沈澱速度を減ずることによりIL−2(N88R)を有意に安定化させた(図2)。表1は、これらの3種の緩衝剤の存在下におけるIL−2(N88R)の開始沈澱温度を示す。開始沈澱温度は350nmにおける光学密度(OD350)がある水準(OD350の場合には0.2および1.0)に達する温度として任意に設定された。ヒスチジンの存在下におけるIL−2(N88R)の沈澱温度は他の2種の緩衝剤の存在下におけるものより数度ほど高かった。この実施例は、ヒスチジンが液体状態のIL−2(N88R)用の緩衝剤として使用されることに加えて特異的な安定剤でありえることを示した。
ヒスチジンの安定化効果をさらに評価する試みでは、凍結乾燥されたIL−2(N88R)を種々の水性調剤から製造した(表2参照)。調剤のほとんどは2重量%のグリシンを増量剤としてそして1重量%のスクロースを安定剤として含有した。5重量%のマンニトールを調剤中でIL−2の商業製品であるプロロイキンRに対する比較剤として使用した。両方とも0.1重量%の2種の界面活性剤であるツィーン80およびプルロニック(Pluronic)F68を蛋白質表面吸着及び凝集の防止に関して評価した。全ての調剤はヒスチジンまたはクエン酸塩を緩衝剤として5.5に調節されたpHで含有していた。pH5.5におけるヒスチジンの安定化効果をクエン酸塩のものと区別するためにクエン酸塩が含まれた。これらの凍結乾燥調剤を40℃において貯蔵しそしてUV/VIS分光光度法、SEC−HPLC、およびRP−HPLCを包含する多くの分析方法により分析した。
野生型IL−2も調剤Aと同じ組成の水性調剤から凍結乾燥した。野生型IL−2に関する40℃における安定性データはIL−2(N88R)のものに匹敵した(表4)。
ヒトIL−2は6の螺旋構造(A−F)を形成する133個のアミノ酸類を有する。これらの螺旋の4個は四螺旋束モチーフ(tetra-helix bundle motif)と称するものを形成する。cys58およびcys105の間の分子内ジスルフィド結合は螺旋間の伸びたループ上に位置する。遊離cys125はアミノ酸類117−133を組み入れる螺旋F上に位置する。
Claims (1)
- ヒスチジン、グリシンおよびスクロースで安定化させたヒトインターロイキン−2(IL−2)ムテインを含んでなる安定な凍結乾燥医薬組成物であって、該IL−2ムテインが単一アミノ酸置換N88Rを有し、マンニトール、クエン酸ナトリウム、ツィーン80およびプルロニックF68を含まない、上記組成物。
