Claims (11)
- 圧電素子に一末端部位が固定化された対象核酸分子を、遊離末端側から一核酸分子単位で切断する第1工程と、
第1工程前の核酸分子の質量(M 1 )と切断後の核酸分子の質量(M 2 )との質量差分(Δm=M 1 −M 2 )に対応する前記圧電素子の振動数変化量(ΔF)を測定する第2工程と、
前記第2工程で求めた振動数変化量(ΔF)を、塩基種ごとのリファレンスデータと比較又は照合して、前記第1工程で切断された一核酸分子の塩基種を特定する第3工程と、
を少なくとも行う核酸分子の塩基配列決定方法。 - 前記第1工程から第3工程を繰り返すことによって、前記対象核酸分子の全塩基配列を決定することを特徴とする請求項1記載の塩基配列決定方法。
- 前記第1工程を、核酸分解酵素(nuclease)を少なくとも用いて行うことを特徴とする請求項1記載の塩基配列決定方法。
- 前記核酸分解酵素は、エキソヌクレアーゼ(exonuclease)であることを特徴とする請求項3記載の塩基配列決定方法。
- 前記対象核酸分子は、一本鎖核酸分子であることを特徴とする請求項1記載の塩基配列決定方法。
- 前記対象核酸分子を断片化した後に、前記第1工程へ移行することを特徴とする請求項1記載の塩基配列決定方法。
- 断片化核酸分子の各塩基配列を、同時並行的に決定することを特徴とする請求項6記載の塩基配列決定方法。
- 予め、同一の塩基種のみからなるオリゴ核酸分子又はポリ核酸分子を対象として前記第1工程及び第2工程を行って、各塩基種に対応する一核酸分子単位の質量変化に相関する振動数変化を求めておき、これを塩基種ごとのリファレンスデータとして用いるとを特徴とする請求項1記載の塩基配列決定方法。
- 前記圧電素子は、水晶発振子であることを特徴とする請求項1記載の塩基配列決定方法。
- 圧電素子の反応領域中に固定化された対象核酸分子を一核酸分子単位で切断する物質を、前記反応領域へ供給する物質供給部と、
切断前の核酸分子の質量(M 1 )と切断後の核酸分子の質量(M 2 )との質量差分(Δm=M 1 −M 2 )に対応する圧電素子の振動数変化量(ΔF)を検出するための検出部と、
予め記憶されている塩基種ごとのリファレンスデータと、前記検出部で検出された振動数変化量(ΔF)とを比較又は照合して、前記対象核酸分子から切断された一核酸分子の塩基種を特定するデータ解析部と、
前記解析部の解析結果を出力する出力部と、
を少なくとも備える塩基配列決定装置。 - 前記圧電素子は、前記対象核酸分子を固定化可能な端面構成を備えるカンチレバーであり、前記検出部は、該カンチレバーの振動振幅を検出することを特徴とする請求項10記載の塩基配列決定装置。
