Description
(1)
支持体の少なくとも片面に粘着剤層を備える貼付製剤であって、該粘着剤層は、粘着剤層総重量基準で5〜50重量%の粘度平均分子量2,500,000〜5,500,000の第一の合成ゴム、粘着剤層総重量基準で5〜50重量%の粘度平均分子量40,000〜85,000の第二の合成ゴム、粘着剤層総重量基準で0.1〜10重量%のlogPowが0.5〜5.5である薬物、粘着剤層総重量基準で20重量%より多く、50重量%より少ない有機液状成分および粘着剤層の総重量基準で5〜50重量%のタッキファイヤーを含む、貼付製剤であって、該有機液状成分は、2種以上の脂肪酸アルキルエステルを含む、貼付製剤;
(2)
有機液状成分が、炭素数16〜22の長鎖アルコールをさらに含む、(1)記載の貼付製剤;
(3)
長鎖アルコールが、粘着剤層総重量基準で0重量%より多く、7.5重量%より少なく粘着剤層に含まれる、(2)記載の貼付製剤;
(4)
該薬物は固体薬物である、(1)〜(3)いずれかに記載の貼付製剤;
を提供する。
一方、粘度平均分子量が6,500,000を超える合成ゴムを含む粘着剤層は、皮膚接着力やタックが低下する虞がある。
薬物:フェンタニル
分配係数logPow=3.78、分子量336.47、融点83〜84℃
A:粘度平均分子量4,000,000の2−メチルプロペン−ポリマー
B:粘度平均分子量55,000の2−メチルプロペン−ポリマー
C:粘度平均分子量800,000の2−メチルプロペン−ポリマー
D:粘度平均分子量2,600,000の2−メチルプロペン−ポリマー
E:粘度平均分子量2,120,000の2−メチルプロペン−ポリマー
F:粘度平均分子量1,660,000の2−メチルプロペン−ポリマー
G:粘度平均分子量1,250,000の2−メチルプロペン−ポリマー
H:粘度平均分子量900,000の2−メチルプロペン−ポリマー
タッキファイヤー: ポリブテン
IPM:ミリスチン酸イソプロピル
IPP:パルミチン酸イソプロピル
ODO:オクチルドデカノール
(予備試験例1〜2、実施例1〜12および比較例1〜7)
表1に記載の配合割合にしたがって粘着剤層形成用組成物のトルエン溶液を調製し、得られた溶液をシリコーン剥離処理を施したポリエチレンテレフタレート(PET)製ライナー(厚さ75μm)上に、乾燥後に表1に示した厚みとなるように塗布し、これを熱風循環式乾燥機中で100℃、5分間乾燥して粘着剤層を形成した。この粘着剤層をPET製支持体に貼り合わせ、シート状の貼付製剤を得た。
予備試験例1〜2、実施例1〜12および比較例1〜7で得られた貼付製剤において、糊はみだしと結晶を観察し、保持力を測定した。観察結果を表1に示す。
(1)糊はみだし
室温で3ヶ月保存した製剤を包材より取り出すときに次の指標で評価した。
○:糊はみだしがほとんど見られず、包材からべたつかずに容易に取り出すことができた。
×:糊はみだしが見られ、包材にべたつき取り出すことが困難であった。
(2)結晶
室温で3ヶ月保存した貼付製剤を目視と顕微鏡で観察し次の指標で評価した。
○:薬物の結晶は析出していなかった。
×:薬物の結晶が析出していた。
(3)保持力
23±2℃、50±10%RH(相対湿度)の室内において、清浄にしたフェノール樹脂板を試験片とし、幅10mmの試験片の粘着面を軽く貼り、試験片の上から850gの圧着ローラーを1往復させることにより圧着させた。圧着面積を200mm2とした。40±2℃の環境にされた保持力試験機内にて30分放置した後、試験板の一端を止めて、試験片が垂直に垂れ下がるようにし、その下端を150gの荷重をかけて、落下するまでの時間(分)を求めた。また、破壊について、今回測定した全ての試験片は凝集破壊モードであったことを確認した。
第一の合成ゴムとしてAを用い、有機液状成分の合計割合が20重量%以上である実施例1〜12は、糊はみだしが発生せず、また薬物の結晶も析出しなかった。しかし、Aの代わりにCを用いた比較例の場合、有機液状成分の合計割合が20重量%より少ないにもかかわらず、糊はみだしが発生し(比較例1および2)、薬物の結晶が析出した(比較例2〜5)。Aの代わりにGまたはHを使用した比較例6および7は、糊はみ出しが発生し、保持力も低かった。有機液状成分を23.5重量%含む実施例7〜10および比較例6〜7において、第一の合成ゴム成分の粘度平均分子量が1,600,000以上、特に2,000,000以上のものが保持力が十分であり、一方合成ゴム成分の粘度平均分子量が1,600,000未満である比較例6〜7は保持力が不十分であった。また、実施例5〜7および11〜12において、いずれも保持力は高かったので、これらの粘着剤層はいずれも、きわめて良好な保形性を有しており、貼付製剤保管中の糊はみ出しや、貼付製剤の皮膚からの剥離時における皮膚面の糊残りの虞がなかった。
<試験方法>
直径12mmφの円形状に切断した上記貼付製剤をヒト皮膚の角質面に貼り付け、フランツ型拡散セルに真皮層側を装着し、レセプター液に32℃の生理食塩液(0.9重量%NaCl+0.01重量%NaN3)を用いて試験した。一定時間毎にレセプター液をサンプリングし、サンプリング液中の薬物重量をHPLC法にて定量し、48時間目までの最大フラックスを算出した。最大フラックスは比較例1を100としたときの相対値を示した。
試験結果を表2に示す。
<試験方法>
実施例5および7で得られた貼付製剤を、ポリエチレンテレフタレート/アルミニウム/アクリロニトリル系樹脂包材にて包装し、50℃で2ヶ月間保存した。次いで保存期間後の固体薬物重量をHPLC法にて定量し、初期の薬物重量に対する保存後の薬物重量の割合を百分率で算出した。結果を表3に示す。

- 支持体の少なくとも片面に粘着剤層を備える貼付製剤であって、該粘着剤層は、粘着剤層総重量基準で5〜50重量%の粘度平均分子量2,500,000〜5,500,000の第一の合成ゴム、粘着剤層総重量基準で5〜50重量%の粘度平均分子量40,000〜85,000の第二の合成ゴム、粘着剤層総重量基準で0.1〜10重量%のlogPowが0.5〜5.5である薬物、粘着剤層総重量基準で20重量%より多く、50重量%より少ない有機液状成分および粘着剤層の総重量基準で5〜50重量%のタッキファイヤーを含む、貼付製剤であって、該有機液状成分は、2種以上の脂肪酸アルキルエステルを含む、貼付製剤。
- 有機液状成分が、炭素数16〜22の長鎖アルコールをさらに含む、請求項1記載の貼付製剤。
- 長鎖アルコールが、粘着剤層総重量基準で0重量%より多く、7.5重量%より少なく粘着剤層に含まれる、請求項2記載の貼付製剤。
- 該薬物は固体薬物である、請求項1〜3いずれかに記載の貼付製剤。
