Description
本明細書において、以下の用語は次のように定義される。用語「組織(tissue)」とは、多細胞生物において、構成細胞が分化し同一の機能、形態を持った細胞集団をいう。動物の組織は形態的、機能的、又は発生学的根拠に基づいて上皮組織、結合組織、筋肉組織、神経組織に大別できる。組織の成分は細胞のみではなく細胞間を満たす物質である基質(マトリックス)がある。複数の組織や細胞が集まって器官(organ)を構成する。動物の器官は臓器とも呼ばれる。本発明において、「組織」とは「器官」又は「臓器」と同義に用いてもよく、例えば、皮膚、骨格筋、心筋、神経等の他、肝臓、腎臓、すい臓、脾臓、精巣、卵巣、副腎、脳、甲状腺等を含む。
本発明の凍結保存方法は、最初に生体から採取した組織を断片化する。夫々の組織によって好適は断片化方法を用いることができ、例えば、トリプシンやコラゲナーゼ等の酵素処理による化学的な方法でも、手術用ハサミやメスにより細断する物理的な方法であっても何れでもよい。哺乳動物の皮膚のように比較的薄くて脆弱な組織の場合は、物理的な方法による断片化が好ましい。断片化された組織片の大きさは、用いる組織によって異なるが、例えば皮膚組織の場合、最大径で5mm以下であることが好ましく、さらに好ましくは最大径で1mm〜0.5mmである。
凍結された組織片懸濁物は、約1〜数秒以内で解凍するように高速に昇温することにより解凍する。例えば、組織片懸濁物を凍結するチューブを水浴にて加熱するか、又は誘導加熱してもよい。好ましくは、チューブ内で凍結された組織片懸濁物を37℃の水浴に浸漬することにより解凍する。

- 哺乳動物より採取された皮膚を断片化し、断片化された真皮層を有する皮膚片を血清含有最小必須培地中で線維芽細胞が皮膚片の辺縁に遊走するために必要な期間培養し、培養された皮膚片を回収して凍結保護溶液中に懸濁し、そして当該皮膚片懸濁物を−70℃以下の温度で凍結させることを特徴とする真皮由来の線維芽細胞の凍結保存方法。
- 前記断片化された皮膚片の最大径が5mm以下であることを特徴とする請求項1に記載の方法。
- 前記断片化された皮膚片の最大径が1mm〜0.5mmであることを特徴とする請求項1に記載の方法。
- 前記皮膚片の培養は、37℃の温度にて5〜10日間行うことを特徴とする請求項1〜3何れか一項に記載の方法。
- 前記皮膚片の培養は、37℃の温度にて7日間行うことを特徴とする請求項1〜3何れか一項に記載の方法。
