Description
以下の合成例及び実施例において、各種の分析等は以下の条件で行った。すなわち、まず、核磁気共鳴(NMR)スペクトルは、日本電子社製のJNM−GSX−400を用いて測定した。ガスクロマトグラフ−質量分析(GC−MS)は、島津社製のQP−5050を用い、電子衝撃法により行った。高分解質量分析(HRMS)は、日本電子社製のJMS−DX−303を用いて行った。ガスクロマトグラフ(GC)分析は、島津社製のGC−8Aにジーエルサイエンス社製のシリコンOV−17充填ガラスカラム(内径2.6mm、長さ1.5m)を装着して用いた。カラムクロマトグラフィー分離におけるシリカゲルは、和光純薬工業社製のワコーゲルC−200を用いた。
まず、出発原料である3,3’−ジブロモ−2,2’−ビチオフェンを、参考文献(Hong M., Wei H., J. Org. Chem., 2000, 65, 3895)の記載を参照して合成した。そして、これを用いてハロゲン交換反応を行い、3,3’−ジヨード−2,2’−ビチオフェンを合成した。すなわち、まず、300mLの三口フラスコに3,3’−ジブロモ−2,2’−ビチオフェン(2.7g(7mmol))を入れ、これをジエチルエーテル(70mL)に溶かした。次に、反応容器内を窒素置換し、−78℃に冷却した。続いて、ブチルリチウム(1.5Mへキサン溶液、10.3mL(15.4mmol))を加え、1時間撹拌した。さらに、ジエチルエーテルに溶かしたヨウ素(3.9g(15.4mmol))を加え、室温下で1時間撹拌して反応させた。
[縮合環化合物の製造]
20mLの二口フラスコに、3,3’−ジヨード−2,2’−ビチオフェン(84mg(0.2mmol))、酢酸パラジウム(II)(Pd(OAc)2、4.5mg(0.02mmol))、4−オクチン(66mg(0.6mmol))、N,N−ジシクロヘキシルメチルアミン(Cy2MeN、117mg(0.6mmol))、N,N−ジメチルホルムアミド(2.5mL)を加え、反応容器内を窒素置換して100℃で加熱、撹拌して反応させた。4時間後、GC及びGC−MS分析により、反応混合物中に4,5−ジ(n−プロピル)ベンゾ[2,1−b:3,4−b’]ジチオフェンがほぼ定量的(収量;99%以上)に生成していることが確認された。
1H−NMR(400MHz,CDCl3,ppm):δ7.45(d,J=5.5Hz,2H),7.35(d,J=5.5Hz,2H),3.01(m,4H),1.74−1.64(m,4H),1.07(t,J=7.3Hz,6H).
HRMS(EI):m/z 274.0847(C16H18S2で測定して得られた値は274.0850であった).
N,N−ジシクロヘキシルメチルアミンに代えてトリブチルアミン(Bu3N、111mg(0.6mmol))を用いたこと、及び、反応時間を8時間としたこと以外は、実施例1と同様の反応を行った。反応後、GC及びGC−MS分析により、反応混合物中に目的物がほぼ定量的(収量;99%以上)に生成していることが確認された。また、上記一般式(5d)で表される化合物は生じていなかった。
N,N−ジシクロヘキシルメチルアミンに代えてシクロヘキシルジメチルアミン(CyMe2N、76mg(0.6mmol))を用いたこと、及び、反応時間を8時間としたこと以外は、実施例1と同様の反応を行った。反応後、GC及びGC−MS分析により、反応混合物中に目的物が92%の収量で生成していることが確認された。また、上記一般式(5d)で表される化合物は生じていなかった。
N,N−ジシクロヘキシルメチルアミンに代えてジイソプロピルエチルアミン(i−Pr2EtN、78mg(0.6mmol))を用いたこと、及び、反応時間を6時間としたこと以外は、実施例1と同様の反応を行った。反応後、GC及びGC−MS分析により、反応混合物中に目的物が88%の収量で生成していることが確認された。また、上記一般式(5d)で表される化合物は生じていなかった。
20mLの二口フラスコに3,3’−ジヨード−2,2’−ビチオフェン(84mg,0.2mmol)、Pd(OAc)2(2.2mg,0.01mmol)、6−ドデシン(40mg,0.24mmol)、N,N−ジシクロヘキシルメチルアミン(94mg,0.48mmol)、N,N−ジメチルホルムアミド(2.5mL)を加え、反応容器内を窒素置換して130℃で加熱、撹拌して反応させた。
1H−NMR(400MHz,CDCl3,ppm):δ7.46(d,J=5.5Hz,2H),7.37(d,J=5.5Hz,2H),3.02(m,4H),1.70−1.62(m,4H),1.53〜1.37(m,8H)、0.93(t,J=7.3Hz,6H).
HRMS(EI):m/z 330.1469(C20H26S2で測定して得られた値は330.1476であった。).
6−ドデシンに代えて、8−ヘキサデシン(53mg(0.24mmol))を用いたこと以外は、実施例5と同様にして、上記一般式(5c)においてn=5である目的物を油状物質(58mg、単離収率:74%)の状態で得た。なお、反応後の溶液をGCにより分析したときの生成物の収量は99%以上であった。また、上記一般式(5d)で表される化合物は生じていなかった。
1H−NMR(400MHz,CDCl3,ppm):δ7.46(d,J=5.5Hz,2H),7.36(d,J=5.5Hz,2H),3.01(m,4H),1.70−1.61(m,4H),1.52〜1.45(m,4H)、1.41〜1.25(m,12H),0.90(t,J=7.0Hz,6H).
HRMS(EI):m/z 386.2168(C24H34S2で測定して得られた値は386.2102であった。).
N,N−ジシクロヘキシルメチルアミン(Cy2MeN)に代えて、アミン以外の塩基である炭酸カリウム(K2CO3、83mg(0.6mmol))を用いたこと、及び、反応時間を8時間としたこと以外は、実施例1と同様の反応を行った。反応後、GC及びGC−MS分析により、反応混合物中に上記一般式(5c)においてn=1である目的物が54%の収量で生成していることが確認された。また、上記一般式(5d)においてn=1である副生成物が11%の収量で生成していることが確認された。
1H−NMR(400MHz,CDCl3,ppm):δ7.54(d,J=5.5Hz,1H),7.47(d,J=5.5Hz,1H),7.38(d,J=5.5Hz,1H),7.32(d,J=5.5Ha,1H),6.82−6.76(m,1H),5.98(dq,J=15.8,6.6Hz,1H),3.04−2.90(m,2H),2.01(dd,J=6.4,1.8Hz,3H),1.72−1.64(m,2H),1.03(t,J=7.3Hz,3H).
HRMS(EI):m/z 272.0690(C16H16S2で測定して得られた値は272.0693であった。).
Claims (3)
- トリアルキルアミン及びパラジウム錯体の存在下で、下記一般式(2a)で表される化合物と、下記一般式(2b)で表される化合物と、を、反応させて、下記一般式(2c)で表される縮合環化合物を得る、ことを特徴とする縮合環化合物の製造方法。
- 前記トリアルキルアミンを、前記一般式(2a)で表される化合物に対し、200〜400モル%用いる、ことを特徴とする請求項1記載の縮合環化合物の製造方法。
- 前記パラジウム錯体を、前記一般式(2a)で表される化合物に対し、0.01〜50モル%用いる、ことを特徴とする請求項1又は2記載の縮合環化合物の製造方法。
