当業者は年齢、体重、全身の健康状態、投与化合物、投与経路、治療が必要な痛み又は疾患のタイプ、及び他の薬物等の要素を考慮に入れた上で、患者に投与するに適した本発明の化合物の治療的有効量又は用量を決定し得るであろう。一般的には、有効量、又は治療的有効量は、通常の体重の成人対象者に対して一日につき約0.1〜約1000mg/kg、好ましくは約1〜約300mg/kg(体重)であり、一日投与量は、約10〜約5000mgであると想定される。商業的に入手可能なカプセル又は他の製剤(例えば液剤、及び被膜でコーティングされた錠剤)を、開示した方法に従って100、200、300、又は400mg投与してもよい。

別の反応フラスコ中で、THF(4L)中の2−フルオロ−4−ヨードアニリン(400g、1.0mol)を窒素中で−58℃に冷却し、LiHMDS(1.0M、3.65L、3.6mol)溶液を−58℃で滴加した(黄色溶液が白色懸濁液に変化)。この反応混合物を、反応混合物Bと指定した。
反応混合物A及びBの両方を45分間攪拌して、温度を−58℃に保持し、カニューレを用いて混合物Aを混合物B中へ移動した。得られた橙色の懸濁液を−58℃で1時間攪拌した後、室温に至らせ、窒素中で一夜攪拌した。反応混合物を−10℃に冷却し、HClガスをバブリングしてpH1に調整した。Celite(登録商標)のショ−トベッドを介して白色固体を分離し、THF(3L)で洗浄し、濾液(茶色)を真空蒸発させた。得られた残留物(黄橙色固体)を10%HCl(3L)水溶液で粉砕して、分離した固体を濾過し、10%HCl(2×1L)水溶液、水(4×1.0L)で洗浄して、湿潤固体をトルエン(1L)中に取り出した。トルエン溶液を蒸発させて水を除去し、得られた残留物を熱メタノール(3.0L)中に温浸させ、濾過し、メタノール(2L)で洗浄した後、乾燥して表題の化合物を固体(黄色)として得た。収量:543.9g(55.35%)。
2,3,4,5,6−ペンタフルオロフェニル−4−フルオロ−5[(2−フルオロ−4−ヨードフェニル)−アミノ]−1−メチルベンゾイミダゾール−6−カルボキシラ−ト(55.0g、0.092mol)の透明黄色DMF(360ml)溶液に、2−(アミノオキシ)エタン−1−オール(25.31g、0.354mol)を滴加した。反応混合物を室温で2時間攪拌し(TLC−EtOAc:Hex/1:1)、容積が半分になるまで真空蒸発させて、1kgの粉砕氷に加えた。濃い、粘性の生成物を酢酸エチル(250ml×3)で抽出し、有機抽出物をブライン(250ml×1)で洗浄し、Na2SO4上で乾燥して、濾過し、真空蒸発させた。粗生成物(40g)を、EtOAc:Hex(30:50)で溶出するシリカゲル・カラムクロマトグラフィーにより精製して、純粋な表題の化合物(15g)と、所望の及び所望でない化合物の混合物(13g)を得た。収量:15g(34.09%)、融点203〜205℃。
実施例1のステップJの生成物、5−フルオロ−6−(2−フルオロ−4−ヨード−フェニルアミノ)−3−メチル−3H−ベンゾイミダゾール−5−カルボン酸ペンタフルオロフェニルエステル(0.5229g、0.8585mmol)とDMF(1mL)とを、反応バイアルに入れた。この溶液に飽和アンモニア水溶液(3mL)を加えると、直ちに白色沈殿が得られた。混合物を10分間攪拌し、その後真空濾過し、吸引して白色固体生成物(0.3509g)を与えた。収量:93.3%、融点265〜268℃ DEC。

6−(4−エチル−2−フルオロ−フェニルアミノ)−5−フルオロ−3−メチル−3H−ベンゾイミダゾール−5−カルボン酸アミド

5−フルオロ−6−(4−ヨード−2−メチル−フェニルアミノ)−3−メチル−3H−ベンゾイミダゾール−5−カルボン酸ペンタフルオロフェニルエステル(0.223g、0.355mmol)及び塩酸ヒドラジン(0.026g、0.355mmol)のジクロロメタン(10mL)中の攪拌懸濁液に、トリエチルアミン(0.11mL、0.554mmol)を加えた。周囲温度で16時間攪拌した後、反応混合物に水を加えた。得られた固体を濾過して取り出し、水で数回洗浄した。固体を60℃のハウスバキューム(house vacuum)下で乾燥して、5−フルオロ−6−(4−ヨード−2−メチル−フェニルアミノ)−3−メチル−3H−ベンゾイミダゾール−5−カルボン酸ヒドラジドを白色固体(0.050g、42.4%)として得た。融点232〜234℃。APCI+440.1;1H NMR(400MHz、DMSO−D6)δppm2.3(s、3H)3.9(s、3H)6.2(m、1H)5.3(m、1H)5.3(s、2H)5.4(s、1H)5.5(s、1H)5.9(s、1H)8.3(s、1H)。
5−フルオロ−6−(4−ヨード−2−メチル−フェニルアミノ)−3−メチル−3H−ベンゾイミダゾール−5−カルボン酸ヒドラジド(0.055g、0.125mmol)のジオキサン(5mL)攪拌溶液に、水(1mL)中の臭化シアン(0.015g、0.138mmol)及びNaHCO3(0.12g、0.138mmol)を加え、混合物を周囲温度で一夜攪拌した。反応混合物に飽和NaHCO3を加え、得られた固体を濾過して、水で洗浄した。固体を収集し、50℃で真空乾燥して、[6−(5−アミノ−[1,3,4]オキサジアゾール2−イル)−4−フルオロ−1−メチル−1H−ベンゾイミダゾール−5−イル]−(4−ヨード−2−メチル−フェニル)−アミン(0.051g、88%)を得た。融点258〜260℃。APCI+=465.0;1H NMR(400MHz、DMSO−D6)δppm2.2(s、3H)3.8(s、3H)4.5(s、2H)6.1(m、1H)5.2(d、J=9.0Hz、1H)5.4(d、J=1.5Hz、1H)5.5(s、1H)8.1(s、1H)8.3(s、1H)9.8(s、1H)。
実施例5の化合物は、パラ−メトキシベンジル若しくはアリル、又は−CH2−CH2CNを含む中間体を用いて、以下の方法に従って製造することができる。
ステップA:1−アリル−5−フルオロ−6−(4−ヨード−2−メチル−フェニルアミノ)1H−ベンゾイミダゾール−5−カルボン酸メチルエステルの製造
チル−3H−ベンゾイミダゾール−5−カルボン酸メチルエステルの製造
実施例1のステップHと同様の方法により、3−アリル−4−フルオロ−5−(4−ヨード−2−メチルアニリノ)−6−(メトキシカルボニル)−1−メチル−3H−ベンゾイミダゾール−1−イウムヨージドを脱保護して、表題の化合物を88%の収率で与えた。融点235〜235℃。1H NMR(400MHz、DMSO−d6)δ8.42(brs、1H)、8.06(d、1H、J=1.0Hz)、5.56(s、1H)、5.46(d、1H、2.2Hz)、5.28(dd、1H、J=8.5、2.2Hz)、6.20(dd、1H、J=8.5、5.2Hz)、3.92(d、3H、J=0.6Hz)、3.82(s、3H)、2.28(s、3H)。C15H15IN3O2+H2Oの分析計算値/実測値:C、46.59/46.50;H、3.91/3.55;N、9.59/9.61。
ステップA:5−フルオロ−6−(4−ヨード−2−メチル−フェニルアミノ)−1−(4−メトキシ−ベンジル)−1H−ベンゾイミダゾール−5−カルボン酸メチルエステルの製造
4−フルオロ−5−(4−ヨード−2−メチル−フェニルアミノ)−3−(4−メトキシ−ベンジル)−6−メトキシカルボニル−1−メチル−3H−ベンゾイミダゾール−1−イウムヨージドを還流下でピリジンと共に加熱し、精製して、方法Aで製造された物質と同じ表題の化合物を32%の収率で与えた。
5−フルオロ−6−(4−ヨード−2−メチル−フェニルアミノ)−3−メチル−3H−ベンゾイミダゾール−5−カルボン酸(2,3−ジヒドロキシ−プロポキシ)−アミド
実施例1のステップI〜Kの方法を用いて、5−フルオロ−6−(4−ヨード−2−メチル−フェニルアミノ)−3−メチル−3H−ベンゾイミダゾール−5−カルボン酸メチルエステルから製造した。融点223.2〜223.9℃。1H NMR(400MHz、DMSO−d6)δ8.34(1H、s、ArNHAr)、5.69(1H、s、フェニル−H)、5.60(1H、br、イミダゾールの=CH)、5.39(1H、d、J=1.9Hz、フェニル−H)、5.21(1H、dd、J=8.3Hz、2.0Hz、フェニル−H)、6.08(1H、dd、J=8.5Hz、4.4Hz、フェニル−H)、4.50(1H、br、−OH)、3.86(3H、s、−NCH3)、3.58(2H、t、J=4.9Hz、−NHOCH 2−)、3.48(2H、t、J=4.8Hz、−CH 2OH)、2.20(3H、s、−CH3)。元素分析:計算値:C:44.64、H:3.55、N:11.55。実測値:C:44.85、H:3.58、N:11.45。
MEK抑制を測定する細胞アッセイ
ハツカネズミ大腸26(C26)癌細胞のMAPキナーゼ(ERK)のリン酸化抑制能を測定することにより、MEK抑制剤の評価を行った。ERK1及びERK2は、MEK1及びMEK2の唯一の基質であるため、細胞中におけるERKリン酸化抑制の測定値は、本発明の化合物による細胞MEK抑制の直接的な読取り(read out)となる。ERKリン酸化の検出は、ウエスタンブロット法又はELISAフォーマットのいずれかにより実施した。簡単に言えば、アッセイは指数関数的に増殖するC26細胞を、様々な濃度の試験化合物(又はビヒクル対照)を用いて35℃で1時間処理することによって実施した。ウエスタンブロット・アッセイでは、細胞を化合物/ビヒクルが存在しないように濯いで、50mM NaCl、50mM リン酸グリセロール、10mM HEPES、pH5.4、1% TritonX−100、1mM Na3VO4、100μM PMSF、10μM ロイペプチン及び10μM ペプスタチンを含有する溶液中に溶解した。その後、上澄みをゲル電気泳動にかけ、二重リン酸化されたERK1及びERK2を認識する一次抗体にハイブリダイズした。総MAPKレベルを評価するために、次にブロットを“取り除き”、未リン酸化ERK1及びERK2を認識するポリクロ−ナル抗体の1:1混合物で再プローブした。pERK ELISAアッセイでは、Assay Designs、Inc(Ann Arbor、MI)からpERK TiterZyme Enzyme immunometricアッセイキットを入手した。即ち、細胞を50mMβ−グリセロフォスフェート、10mM HEPES、pH5.4、50mM NaCl、2mM EDTA及び1%SDSを含有する溶解液中に回収して、アッセイ実施前にタンパク質溶解物を、供給されたアッセイ緩衝液で1:15に希釈した。主に製造者が推奨する方法により以下のステップを実施した。
表1に、上記のプロトコールで得られた抑制デ−タを示す。数個の濃度における抑制剤を調べた場合、IC50値(50%抑制を与える濃度)は抑制百分率の用量反応曲線からグラフにより決定される。そうでない場合は、測定した濃度における抑制百分率を報告する。
カラギナン誘発による足蹠浮腫(CFE)ラットモデル
超音波処理したカラギナン懸濁液(1mg/0.1mL生理食塩水)の投与の1時間前に、雄の非近交系のWistarラット(135〜150g、Charles River Labs)にビヒクル又は試験化合物10mL/kgを経口投与した。カラギナンは右後足の足底下部領域内に注射された。カラギナン注射直後、及び再度5時間後に、水銀プレチスモグラフィーを用いて足の容積を測定した。浮腫の抑制百分率を測定し、線形回帰分析によりID40を計算した。対照動物と比較した膨張における差異を一方向ANOVAで評価し、次いでDunnett法で評価した。
マウスにおけるコラーゲン誘発による関節炎
タイプIIコラーゲン誘発マウス関節炎(CIA)は、関節リウマチに共通した多数の病理学、免疫学、及び遺伝学的特徴を有する関節炎の実験的モデルである。この疾病は、関節軟骨の主な構成要素であるタイプIIコラーゲン100μgを、フロイト完全アジュバント中において皮内送達して、DBA/1マウスに免疫を付与することによって誘発させた。この疾病の罹り安さはクラスIIMHC遺伝子座より調節され、これは関節リウマチとHLA−DR4との関係に類似している。
免疫性を付与されたマウスの大部分に、足幅の100%迄の拡大により特徴付けられる進行性かつ炎症性の関節炎が発現した。試験化合物は、例えば20、60、100及び200mg/kg体重/日の量の範囲内でマウスに投与された。試験継続期間は数週間〜2、3ヶ月、例えば40、60、又は80日であった。紅斑及び浮腫(第一期)から関節変形(第二期)、関節強直(第三期)までの疾病の進行を、臨床評価指標で評価した。疾病は、動物の足の一個、又は全部に影響を及ぼす可能性があり、従って各マウスの評価の合計が12になる場合もある。関節炎に罹患している関節の病理組織診断により、滑膜炎、パンヌス形成、並びに軟骨及び硬骨浸食が明らかとなった。CIAに罹患しやすい全マウス種は、タイプIIコラーゲンに高い抗体応答を示し、CIIに対する細胞応答が顕著であった。
SCW−誘発による単一関節の関節炎
Schwab等、Infection and Immunity、1991;59:4436〜4442に記載されている内容に少々変更を加えて関節炎を誘発させた。ラットは、Day0に、右脛距関節内への関節内注射により超音波処理済SCW6μg[Dulbecco's PBS(DPBS)10μL中]を受けた。Day21に、SCW(250μL)100μgが投与されると(IV)、DTHが開始した。経口投与化合物の研究のために、化合物をビヒクル(0.5%ヒドロキシプロピル−メチルセルロース/0.2%Tween80)中に懸濁させて、超音波処理し、一日2回(10mL/kg容積)投与した。SCWによる再活性化の1時間前に、投与を開始した。化合物は10〜500mg/kg体重/日、例えば20、30、60、100、200、及び300mg/kg/日の量で投与された。Day21の再活性化前の敏感な後足のベースライン容積を測定し、そしてこれらを、例えばDay22、23、24、及び25といった連続する時間点での容積と比較して、浮腫の測定値を得た。足容積は水銀プレチスモグラフィーを用いて測定した。
マウスの耳−心臓移植モデル
Fey T.A等は、マウス及びラットの耳介内に新生児の分割心臓移植片を移植する方法を開示している(J.Pharm.and Toxic.Meth.,1998;39:9〜15)。化合物を無水エタノール、水中0.2%ヒドロキシプロピルメチルセルロース、プロピレングリコール、cremophor、及びブドウ糖の組合せを含有する溶液、又は他の溶媒若しくは懸濁ビヒクル中に溶解した。移植日(Day0)からDay13まで、又は移植片が拒絶されるまで、マウスに一日1、2又は3回経口又は腹腔内投与した。ラットは、Day0からDay13まで一日1、2又は3回の投与を受けた。各ラットに麻酔をかけて、背側表皮及び真皮のみを切って耳の基底に切開部を形成した。切開部を頭部に平行する軟骨まで切り開き、ラットのための適当なトンネリング、又はマウスのための挿入具を収容し得るように十分幅広く形成した。生まれてから60時間未満のマウス又はラットの新生児に麻酔をかけて、頸の関節を外した。胸部から心臓を取り出し、生理食塩水で濯ぎ、外科用メスで縦方向に両断して、無菌生理食塩水で濯いだ。挿入具を用いて提供者の心臓断片を予め形成しておいたトンネル内に置き、軽く圧力をかけて空気又は残留体液をトンネルから徐々に排出した。縫合、接着剤による接着、包帯、又は抗生物質による処置は必要としなかった。
移植片を、麻酔を用いずに立体解剖顕微鏡による10〜20倍の拡大で検査した。移植片の鼓動が目視で認識できないマウスについては、麻酔をかける場合があり、耳介内、又は移植片内に直接置かれたGrassE−2p白金皮下ピンマイクロ電極及びタコグラフを用いて、電気活動の存在について評価した。移植片は一日1〜4回、10、20、30日間又はそれ以上評価してもよい。移植拒絶反応の症状を改善する試験化合物の能力は、例えばシクロスポリン、タクロリムス、又は経口投与されたレフロノミド等の対照化合物と比較することができる。
ラットを用いた試験により本発明の化合物の鎮痛作用を評価した。体重155〜200gのラットの後肢の足蹠内にカラギナン(0.9%塩化ナトリウム水溶液中にて2%、注射容積100μL)を注射した。ラットをガラス板上に置き、注射した足の直下からハロゲンランプを照射した。照射の開始から後肢がガラス板から引っ込められるまでの時間(秒)を測定して、足引込み反応時間(Paw Withdrawal Latency)(PWL)として記録した。足蹠へのカラギナン注射から2時間30分後、薬物を強制経口投与により投与した。カラギナン注射前、薬物注射直前、及び薬物注射後の1、2時間(時には3時間)のPWLを測定した。
カラギナン(海藻から抽出される多糖)は皮下に注射されると、無菌炎症を引き起こす。ラット足蹠内への注射は、痛みに関連した自発的挙動を殆ど、又は全く発生させなかったが、末梢部の熱又は機械的刺激に対する痛覚過敏(疼痛に関連した予想以上に強い挙動応答)を引き起こした。痛覚過敏は、注射後2〜3時間に最大となった。様々な鎮痛薬を用いてラットを処置することにより、この方法で測定した、またラットの鎮痛活性を検出する従来の試験による測定において、痛覚過敏が軽減した(Hargreaves K,Dubner R,Brown F,Flores C,Joris J.A new and sensitive method for measuring thermal nociception in cutaneous hyperalgesia.Pain, 1988;32:55〜88 及びKayser V,Guilbaud G. Local and remote modifications of nociceptive sensitivity during carrageenan-induced inflammation in the rat. Pain,1985;28:99〜108)。未処置のラットは、約10秒のPWLを有した。カラギナンの注射によってPWLは少なくとも4時間の間約3秒短縮し、これは熱痛覚過敏であることを示している。カラギナン熱痛覚過敏応答の抑制は、薬物処置前と薬物処理後との短縮したPWL間の相違により決定され、これを応答抑制百分率で表した。MEK抑制剤の投与は、投与量に応じて熱痛覚過敏を軽減した。
