抽出溶媒としては、メタノール、エタノール、ブタノール、ヘキサン、ヘプタン、シクロヘキサン、酢酸エチル、アセトン、モノテルペン類などの一般に用いられる有機溶媒、グリセリン、1,3−ブチレングリコール、ジプロピレングリコール、プロピレングリコールなどのグリコール類および水などを挙げることができ、これらの1種を単独で又は2種以上を混合して使用することができる。これらの溶媒の中では抽出効率の点から、特にエタノール、水、1,3−ブチレングリコールおよびこれらの混合溶媒が望ましい。
なお抽出処理は、冷浸、温浸、加熱還流、パーコレーション法などの常法によって行うことができる。溶媒抽出のほかに、水蒸気蒸留、炭酸ガスを超臨界状態にして行う超臨界抽出によって得たエキスも同様に利用できる。超臨界抽出では、抽出助剤としてヘキサン、エタノール等を用いることもできる。
また、抽出物の分離精製は、抽出物を活性炭処理、液液分配、カラムクロマトグラフィー、液体クロマトグラフィーなどで行うことができる。
その他の抽出条件、例えば、抽出温度、抽出pHなどは、抽出溶媒や抽出処理の種類に応じて、適切な条件を設定すればよい。
さらに、例えば、公知のソフォラフラバノンGを出発物質としてナトリウムメトキシド等による公知のエーテル化法によりメチルエーテル化し、高速液体クロマトグラフィー等により分離精製を行い、化学合成物を得ることもできる。
クララの根40gにエタノール800mlを加えて加温抽出し、冷却後、減圧濃縮して蒸留水を添加し、50%エタノール抽出溶液を得た。得られた50%エタノール抽出溶液をろ過して固形物を除き、ろ液を減圧下で濃縮乾固後、乾固形物を固形物換算で20%となるようにメタノールに溶解し、シリカゲルカラム(和光純薬工業社製:ワコーゲル C−200)にてクロロホルム:メタノール=90:10により溶出して溶出液を得た。得られた溶出液を減圧下で濃縮乾固し、乾固形物を固形物換算で10%となるように50%エタノールに溶解し、高速液体クロマトグラフィー(島津社製:Tskgel ODS−80Ts)にて蒸留水:アセトニトリル:酢酸=50:50:1により溶出して溶出液を得た。次に減圧下で濃縮乾固し、ソフォラフラバノンG−5−メチルエーテル及びソフォラフラバノンG−4",5"−ジヒドロ−5"−ヒドロキシ−5−メチルエーテルを得た。
体臭原因菌として、Corynebacterium xerosis菌を用いて、本発明の抗菌剤の殺菌試験及び防臭試験を実施した。
TSB培地(DIFCO製)でCorynebacterium xerosis菌を37℃,24時間前培養した。655nmにおける吸光度が1.3以上あることを確認後、3000rpmで5分間遠心分離処理を行い、菌体(沈殿物)を回収した。菌体を生理食塩水に懸濁させ、再び同様の操作を行い、得られた菌体をもとの菌液の1/2量のリン酸緩衝液pH6.0に懸濁させ、供試菌液とした。
試験菌として、Corynebacterium xerosis菌、Staphylococcus epidermidis菌およびStaphylococcus aureus菌を用いて、最小発育阻止濃度測定試験を実施した。
以下、本発明のソフォラフラバノンG−5−メチルエーテル及びソフォラフラバノンG−4",5"−ジヒドロ−5"−ヒドロキシ−5−メチルエーテルを所要の組成物に配合した処方例を示す。
Claims (6)
- コリネバクテリウム属の微生物による臭気の発生を抑制する作用を有するソフォラフラバノンG−4",5"−ジヒドロ−5"−ヒドロキシ−5−メチルエーテルを有効成分として含む防臭剤。
- 植物抽出液を精製処理することにより取得され、ソフォラフラバノンG−4",5"−ジヒドロ−5"−ヒドロキシ−5−メチルエーテルを含有する画分として、前記有効成分を含むことを特徴とする請求項1に記載の防臭剤。
- 前記植物抽出液がクララ抽出液であることを特徴とする請求項2に記載の防臭剤。
- 請求項1〜3のいずれかに記載の防臭剤を含有することを特徴とする防臭用組成物。
- 請求項1〜3のいずれかに記載の防臭剤を含有することを特徴とする化粧料組成物。
- 請求項1〜3のいずれかに記載の防臭剤を含有することを特徴とする洗浄用組成物。
