Description
本発明は、薬剤とナノ粒子を連結することができるリンカーに関する。特に、本発明のリンカーは、pH感受性であり、ナノ粒子と同種または異なる種の薬剤を同時に結合することができる。
治療化合物および診断化合物を細胞、特にサイトゾルに特異的且つ効率的に送達させることは、多くの製薬会社の主な目的である。特異性および取り込みを増大させるために多くの異なるアプローチが利用されてきた。例えば、薬物送達および癌治療のための新規のストラテジーの開発においてナノテクノロジーが広く使用されている。薬物放出が酸性腫瘍環境によって特異的に誘発されるPH感受性ナノシステムが開発されており、かかるシステムは、癌処置の効率を改善することができる。Feng Wnangらは、ポリ(エチレングリコール)空間を使用して酸不安定性連結を介してAuNP表面上にドキソルビシンを係留することによる薬物送達系を開発している(American Chemical Society,2011,Vol.5,No.5,pp.3679−3692)。Tian−Meng Sunらは、ヒドラゾン結合を介して金ナノ粒子に抱合したドキソルビシンを使用したがん幹細胞治療を開示している(Biomaterials 35,2014,pp.836−845)。米国特許出願公開第2013/0331764号は、癌細胞を区別するためのpH感受性金属ナノ粒子に抗癌薬を結合することによって抗癌薬を癌細胞内に送達させる方法に関する。WO2013/139942号は、金属ナノ粒子および少なくとも1つのリンカーを含むナノ粒子を提供している。

本発明者らは、9種のリンカーの細胞傷害性を試験し、各リンカーのIC50値を計算した。ヒト乳腺腺癌(brast adenocarcinoma)MCF7、MDA−MB−453、およびMDA−MB−231ならびにヒト乳房上皮細胞H184B5F5/M10を、細胞傷害性試験のために選択した。細胞を6ウェルプレートに播種し、リンカーI−1(リンカーI−アミド、m=2)、リンカーI−2(リンカーI−アミド、m=1)、リンカーI−3(リンカーI−アミド、m=3)、リンカーI−4(リンカーI−アミド、m=4)、リンカーI−5(リンカーI−アミド、m=5)、リンカーII−1(リンカーII−アミド、z=1)、リンカーII−2(リンカーII−アミド、z=2)、リンカーII−3(リンカーII−アミド、z=3)、およびリンカーII−4(リンカーII−アミド、z=4)で処置し、37℃で72時間さらにインキュベートした。写真撮影後、血球計算器を使用して細胞を計数した。成長阻害を無処置対照と比較してリンカーの50%成長阻害濃度(IC50)を見出した。IC50のまとめを表1に示す。リンカーI−1〜リンカーI−5およびリンカーII−1におけるIC50値は300〜700μMであった。リンカーII−2〜II−4のIC50値は800μMを超える。これらのデータは、薬物送達のために薬物と金ナノ粒子を抱合するためのリンカーとしての細胞毒性が低いことを示唆している。

Au/LKI−1/Tras複合体の製剤を、UV−Vis分光光度計(Beckman,DU 800)で分析した。記録された表面プラズモン共鳴スペクトルは、抗体抱合金ナノ粒子のプラズモンピーク(λmax)の8nmレッドシフトを示した(図3)。本発明者らは、Au/LKI−1/Tras複合体を試験した。Au/LKI−1/Tras複合体のフローサイトメトリー分析により、ヒト乳腺腺癌細胞に対する標的親和性が証明された。Au/LKI−1/Tras複合体は、トラスツズマブと比較して類似の標的能を示した(表3)。
実施例22 Au/リンカー(LK)/Dox複合体の細胞傷害性アッセイ
本発明者らは、Au/LKs/Dox複合体の細胞傷害性を試験し、IC50値を計算した。ヒト乳腺腺癌(brast adenocarcinoma)MCF7、MDA−MB−453、およびMDA−MB−231、ならびにヒト乳房上皮細胞H184B5F5/M10を、細胞傷害性試験のために選択した。細胞を96ウェルプレートに播種し、2μMのドキソルビシン(Dox)、以下の表中に示した種々のリンカーを使用してドキソルビシンと抱合し、SH−PEGでキャップした20〜50nmの金ナノ粒子(Au/LK/Dox、2μMドキソルビシン含有)で処置し、37℃で72時間さらにインキュベートした。CellTiter 96(登録商標)AQueous One Solution細胞増殖アッセイ(MTS)試薬をPromega(Madison,WI,USA)から購入し、製造者の説明書にしたがってアッセイを行なった。マイクロプレートリーダー(Bio−tek,Powerwave X340,Winooski,VT,USA)を使用して490nmの吸光度を測定した。成長阻害を無処置対照と比較してAu/LKs/Dox複合体の50%成長阻害濃度(IC50)を見出した。
1mLのAu/LKI−1/Dox複合体溶液を、14000rpmで20分間遠心分離した。上清を破棄し、pH5.5の緩衝液またはpH7.4の緩衝液を添加した。異なる時点で、複合体溶液を14000rpmで20分間遠心分離し、次いで、上清を取り出して557.6nmでの蛍光スキャン(F−4500FL分光光度計)に供してOD値を得た。複合体からのDoxの放出量を、Dox検量線からの内挿によって得ることができる(X時間Dox放出量)。

BALB/c−nu/nuマウスを、BioLASCO Taiwanから購入した。8週齢の雄マウスに、1.0×107個のMDA−MB−231乳房腫瘍細胞を含む200μLの50:50 Matrigel/Leibovitz’s(L−15)を、皮下注射によって背側の片側に注射した。(1)ビヒクル−PBS(ネガティブコントロール)、(2)ドキソルビシン(ポジティブコントロール)(5mg/kg)、(3)Au/PEG、(4)Au/LKI−1/Dox複合体(Doxは5mg/kg)。臨床症状によって屠殺を必要とするまで処置を継続した。マウスの体重を最低2回/週測定し、臨床症状によって屠殺を必要とするまで腫瘍サイズをモニタリングした(図6A、6Bを参照のこと)。異種移植片モデルにおけるAu/LKI−1/Dox複合体で処置したMDA−MB−231乳房腫瘍細胞のTEM画像。図中の矢印は、Au/LKI−1/Dox複合体がMDA−MB−231乳房腫瘍細胞内に存在することを示す(図6Cを参照のこと)。異種移植片モデルにおけるAu/LKI−1/Dox複合体で処置したMDA−MB−231腫瘍細胞のH&E染色(図6Dを参照のこと)。

- 前記金属ナノ粒子複合体が、異なる分子長を有する複数のリンカーを含む、請求項21に記載の金属ナノ粒子複合体。
- 本発明で使用される前記PEGの分子量が約2000〜20,000Daの範囲である、請求項21に記載の金属ナノ粒子複合体。
- 前記金属ナノ粒子のサイズが約80nm未満である、請求項21に記載の金属ナノ粒子複合体。
- 前記金属ナノ粒子複合体が、1つまたは複数の同一の異なる治療薬または診断薬とさらに連結している、請求項21に記載の金属ナノ粒子複合体。
- 前記治療薬が抗腫瘍薬または抗体であり;好ましくは、前記抗体が腫瘍細胞などの特定の細胞を標的にする抗体または抗腫瘍抗体である、請求項27に記載の金属ナノ粒子複合体。
- 前記抗腫瘍薬が抗癌薬である、請求項28に記載の金属ナノ粒子複合体。
- 前記抗癌薬が、アルキル化剤、スルホン酸アルキル、アジリジン、プリンアナログ、ピリミジンアナログ、カンプトセシン、またはドキソルビシン、シスプラチンである、請求項29に記載の金属ナノ粒子複合体。
- 前記抗癌物質の量が前記金属ナノ粒子複合体の約1%〜約50%(重量/重量)を占める、請求項29に記載の金属ナノ粒子複合体。
- 1つまたは複数の同一の異なる治療薬または診断薬および薬学的に許容され得るキャリアと連結した1つまたは複数の金属ナノ粒子複合体を含む薬学的組成物。
- 治療薬または診断薬を被験体に送達させる方法であって、請求項27に記載の金属ナノ粒子複合体を前記被験体に投与する工程を含む、方法。
- 1つまたは複数の同一の異なる治療薬または診断薬を連結した1つまたは複数の請求項1〜20のいずれか1項に記載の金属ナノ粒子複合体を含む薬物送達系。
