Abstract
Description
乳液鋳型法を用いてインスリン・ナノ粒子を調製した。80 mgのインスリン、および20 mgのラクトースを、1 mlの0.1M HClに溶解した。別に、2 gのレシチン(フォスファチジルコリン界面活性剤)を秤量し、7 mlのクロロフォルムに溶解し、油相を形成した。水相を、低速(10,000 rpm)で、次いで高速(24,000 rpm)で5分ホモジェナイズしながら、この油相に滴下した。直ちに液体窒素を用いて、形成された乳液を急冷し、分散相を不動とした。この凍結乳液を、真空密栓ガラスジャー(Girovac Ltd., North Walsham, Norfolk, UK)に移し、-110℃に設定された凍結乾燥機(Drywinner 110, Heto-Holten A/S, Gydevang, Denmark)に接続した。凍結乾燥は、最小12時間行い、凍結した微小水性小滴から水分を除去した。
SXを秤量し、既知量の冷却シトラールに溶解した。SXは、66-81 μg/mlの水性溶解度(Tong, HY, et al, Pharm. Res. (2001) 18, 852-858)を持つ、すなわち、本明細書の観点からは「水溶性」ではない。この溶液(量/濃度については下記参照)を、低速(10,000 rpm)でホモジェナイズしながら、10 mlの冷却HFA 134aに滴下した。直ちに、63 μlの作動計量バルブ(Valois DF60 MK42; Valois, フランス)を、pMDIキャニスターに嵌めこんだ(手動瓶クリンパー3000、Aero-Tech Laboratory Equipment Company, 米国、を用いて)。使用したシトラールの量は、全体処方剤に基づき、2から33% v/vまで変動させ、一方、処方におけるSX濃度は、0.05から0.9% w/vまで変動させた。pMDIバイアルは約1分間超音波処理(XB6 Grant Instrument Ltd., UK)し、懸濁の安定性を目視によって評価した。0.05, 0.1, 0.2 および0.9% w/vのSX、並びに、2, 9 および23% v/vのシトラールを含む処方を、60L/分に設定した2段階(ツイン)噴射機(段階間の実効カットオフ直径= 6.4 μm)を用いて、その噴霧特性を評価した。このツイン噴射機の上部および下部段階におけるSXの量を、表1に掲げる条件に設定した高速液体クロマトグラフィー(HPLC)を用いて評価した。

Claims (27)
- 加圧型用量計量吸引器であって、薬学的に受容可能なヒドロフルオロアルカン液体噴霧剤、該ヒドロフルオロアルカン液に対して不溶で、粒状において肺に送達される薬剤、および、室温において該ヒドロフルオロアルカン液と混和可能な、薬学的に受容可能な精油である懸濁安定剤を含む組成物を含む、吸引器。
- 薬剤を含む前記粒子が、固形保護剤、好ましくは分解保護剤、より好ましくは糖、もっとも好ましくはラクトースおよびトレハロースから選ばれる糖をさらに含むことを特徴とする、請求項1に記載の吸引器。
- 前記薬剤が、水溶性であり、0.1 mg/mlを超える水溶性を有する化合物であることを特徴とする、請求項1または請求項2に記載の吸引器。
- 前記薬剤が、ペプチドまたはタンパク質、好ましくは酵素またはホルモン、より好ましくはインスリンであることを特徴とする、請求項3に記載の吸引器。
- 前記粒子が、0.01から20 μmの範囲の平均直径を有することを特徴とする、請求項3または請求項4に記載の吸引器。
- 前記薬剤が、0.1 mg/ml未満(室温)の水溶性を有し、安定剤に対し少なくとも0.5 mg/ml(室温)の溶解度を有し、ヒドロフルオロアルカンに対し0.1 mg/ml未満(室温)の溶解度を有することを特徴とする、請求項1に記載の吸引器。
- 前記薬剤が、ステロイド、コルチコステロイド、気管支拡張剤、ベータ2作用剤、抗生物質、抗菌剤、抗ウィルス剤、および抗ヒスタミン剤から選ばれることを特徴とする、請求項6に記載の吸引器。
- 前記薬剤が、サルメテロール、テルブタリン、クロモリン、ベクロメタゾン、ブデソニド、モメタゾン、シクレソニド、トリアムシノロン、フルチカゾン、ロフレポニド、サルブタモール、フォルモテロール、および、それらの薬学的に受容可能な塩およびエステルから選ばれることを特徴とする、請求項7に記載の吸引器。
- 前記油が両親媒性を有し、該油において、疎水性部分は、少なくとも一つのC6-24-アルキル鎖を含み、親水性部分は、好ましくはケトン様またはアルデヒド様カルボニル基を含むことを特徴とする、請求項1〜8のいずれか1項に記載の吸引器。
- 前記油が、シトラール、メントール、ユーカリ油、シナムアルデヒド、およびシネオールから選ばれることを特徴とする、請求項9に記載の吸引器。
- 前記ヒドロフルオロアルカンが、HFA 134a、またはHFA 227であることを特徴とする、請求項1〜10のいずれか1項に記載の吸引器。
- 安定剤の、ヒドロフルオロアルカンに対する容量比が、1:10,000から10,000:1の範囲、好ましくは1:100から1:2の範囲にあることを特徴とする、請求項1〜11のいずれか1項に記載の吸引器。
- 全体組成物に対し、0.001から10% w/wの範囲の薬剤濃度を有する、請求項1〜12のいずれか1項に記載の吸引器。
- 作動毎に、0.001から10 mg薬剤の範囲の量として薬剤を含む組成物用量を送達するように適応される、請求項1〜13のいずれか1項に記載の吸引器。
- 請求項1に記載の吸引器を生産する方法であって、薬剤は、懸濁安定剤において分散されて分散体を形成し、該分散体は、ヒドロフルオロアルカンに加えられて懸濁液を形成し、該懸濁液は容器に納められ、該容器は、内容物が室温で加圧されるように封鎖されることを特徴とする方法。
- 前記薬剤が、0.1 mg/ml以下の程度まで懸濁安定剤に可溶であり、該懸濁安定剤において粒状に分散して、該懸濁安定剤において該粒子の懸濁液を形成し、該懸濁液が、ヒドロフルオロアルカンと組み合わされることを特徴とする、請求項15に記載の方法。
- 前記粒子が、請求項2に定義する保護剤を含むことを特徴とする、請求項16に記載の方法。
- 予備的工程、すなわち:薬剤および保護剤の水溶液を形成する工程;連続油相において、薬剤および保護剤水溶液の油中水乳液を形成する工程;および、該乳液を乾燥して粒子を形成する工程、を含む、請求項17に記載の方法。
- 前記乳液が、スプレイ乾燥、または凍結乾燥のプロセスによって乾燥されることを特徴とする、請求項18に記載の方法。
- 前記薬剤は、少なくとも0.5 mg/mlレベルにおいて前記懸濁安定剤に可溶であり、該薬剤は、該懸濁安定剤に溶解して溶液を形成し、該溶液は、前記ヒドロフルオロアルカンと組み合わされて、薬剤沈殿物を形成することを特徴とする、請求項15に記載の方法。
- 前記分散体および前記ヒドロフルオロアルカンが、容器において加圧下に混ぜ合わされ、次いで、該容器が封鎖されて、加圧型用量計量吸引器を形成することを特徴とする、請求項15に記載の方法。
- 請求項2〜13のいずれかに定義されるさらに別の特性を1つ以上有することを特徴とする、請求項15に記載の方法。
- MDIにおいて、薬学的に受容可能なヒドロフルオロアルカン液における粒状薬剤の懸濁液を安定化するための、薬学的に受容可能な精油の使用方法であって、該ヒドロフルオロアルカン液に対し、該薬剤は不溶であり、該精油は混和可能であることを特徴とする、使用方法。
- 吸引を通じて動物対象の肺に投与するための組成物の製造における、薬学的に受容可能なヒドロフルオロアルカン液、薬剤、および、薬学的に受容可能な精油の組み合わせの使用方法であって、該薬剤は、該組成物において粒状となり、該薬剤の粒子は、該ヒドロフルオロアルカン液に懸濁され、該精油は、該ヒドロフルオロアルカン対し混和可能であり、かつ、該懸濁液は、該精油によって安定化されることを特徴とする、使用方法。
- 請求項2〜13のいずれかに定義されるさらに別の特性を有することを特徴とする、請求項23または24に記載の使用方法。
- 薬学的に受容可能なヒドロフルオロアルカン液体噴霧剤、懸濁粒子状のインスリン、およびシネオールを含む、吸引用加圧組成物。
- 薬学的に受容可能なフルオロアルカン液体噴霧剤、肺に送達されるステロイドまたはコルチコステロイド、およびシトラールを含む、吸引用加圧組成物。
