Abstract
【解決手段】 システイン含有率が7〜20w/w%であるシステインリッチペプチドの混合物を含む調製物であって、該混合物が、タンパク質源のタンパク質をペプチドに開裂する工程;上記工程で得たペプチドを少なくとも1種類のエキソペプチダーゼで消化して(この作用は、少なくとも、該ペプチド中のシステインまたはシステインおよびグリシンの位置で弱められる)、末端システインまたはシステインおよびグリシンを有する消化されたペプチドを形成する工程;消化されたペプチドを精製する工程を含むことを特徴とする、システインまたはシステインおよびグリシン含有タンパク質を含むタンパク質源からシステイン含有率またはシステインおよびグリシン総含有率7〜20w/w%のペプチド混合物を調製する方法に従ってシステイン含有タンパク質の酵素的開裂により得られるものである、調製物。

Description
a)タンパク質源のタンパク質をペプチドに開裂する工程;
b)工程a)で得たペプチドを少なくとも1種類のエキソペプチダーゼで消化して(ここで、この作用は、少なくとも、該ペプチド中のシステインの位置で弱められる)、末端システインを有する消化されたペプチドを形成する工程;
c)消化されたペプチドを精製する工程
を含むことを特徴としている。
圧力: 30バール
温度: 50〜55℃
初期流速: 58リットル/m2/時
最終流速: 23リットル/m2/時
プロセス: 30リットルまで濃縮、次いで、200%ダイアフィルトレーション
逆相HPLC法(RPC)を、ペプチド混合物中のシステイン含有ペプチドを同定および定量化できるように設定した。まず、システイン残基を蛍光標識(SBD−F;7−フルオロベンゾ−2−オキサ−1,3−ジアゾール−4−スルホン酸;Sigma F−4383)で標識化した。この標識は、システイン残基に特異的に結合する。
図2は、蛍光を測定することによる同加水分解産物からの特異的システイン含有ペプチドの検出を示す。
Claims (8)
- システイン含有率が7〜20w/w%であるシステインリッチペプチドの混合物を含む調製物であって、該混合物が、
a)システイン含有タンパク質を含むタンパク質源のタンパク質をペプチドに開裂する工程;
b)工程a)で得たペプチドを少なくとも1種類のエキソペプチダーゼで消化して(ここで、この作用は、少なくとも、該ペプチド中のシステインの位置で弱められる)、末端システインを有する消化されたペプチドを形成する工程;
c)消化されたペプチドを精製する工程
を含む、システイン含有タンパク質を含むタンパク質源からシステイン含有率7〜20w/w%のペプチド混合物を調製する方法に従ってシステイン含有タンパク質の酵素的開裂により得られるものである、調製物。 - a)システインおよびグリシン含有タンパク質をペプチドに開裂する工程;
b)工程a)で得たペプチドを少なくとも1種類のエキソペプチダーゼで消化して(ここで、この作用は、少なくとも、該ペプチド中のシステインおよびグリシンの位置で弱められる)、末端システインまたはグリシンを有する消化されたペプチドを形成する工程;
c)消化されたペプチドを精製する工程
を含む、システインおよびグリシン含有タンパク質を含むタンパク質源からシステインおよびグリシン総含有率7〜20w/w%のペプチド混合物を調製する方法に従って乳漿タンパク質、アルブミン、小麦グルテン、トウモロコシタンパク質単離体、卵タンパク質、およびシスタチンから選択される1種類またはそれ以上のタンパク質の酵素的開裂により得られるシステインおよびグリシンリッチペプチドの混合物を含む調製物であって、該ペプチドがシステインおよびグリシンを7〜20w/w%含む、調製物。 - システイン含有タンパク質が乳漿タンパク質を含む、請求項1記載の調製物。
- ペプチドの分子量が200Da〜8000Daである、請求項1または2記載の調製物。
- 薬物の製造のための請求項1または2記載の調製物の使用。
- 酸化的障害により媒介される症状の治療用の薬物の製造のための請求項1または2記載の調製物の使用。
- 人体または動物体において細胞内グルタチオンレベルを上昇させる薬物の製造のための請求項1または2記載の調製物の使用。
- 乳児用処方物の製造のための請求項1または2記載の調製物の使用。
