【発明の属する技術分野】本発明は、ボルジン誘導体を含有することを特徴とする美白剤に関する。
【０００２】
【従来の技術】従来の美白化粧料には、メラニンの生成を抑制する事を目的として、アルブチン、コウジ酸、エラグ酸、ビタミンＣ、カミツレエキス、プラセンタエキス等が配合されてきた。これらの成分は、チロシナーゼの遺伝子発現阻害、チロシナーゼ活性阻害、ケラチノサイトからメラノサイトへの情報伝達物質の生成抑制、メラニン合成中間体の還元等により美白効果を示すものであるが、これらの美白剤は、美白効果が十分でなかったり、安全性が十分でなかったり、高価であるといった問題があった。

【発明が解決しようとする課題】そこで、本発明は、十分な美白効果があり、皮膚に対する刺激性がなく人体に安全である、新規の美白化粧料を提供することを目的とする。

【課題を解決するための手段】本発明者は、前記課題を解決すべく鋭意検討した結果、ボルジン誘導体を美白剤として化粧料に添加することにより前記課題を解決できることを見出し、本発明に至った。即ち、本発明は、ボルジン誘導体を単独または少なくとも１種以上の他の美白有効成分に加えることを特徴とする、優れた美白効果を有する化粧料あるいは皮膚外用剤に関するものである。
【０００５】従来の細胞活性化の仕組みでは、外的または内的刺激により、サイトカイン・ホルモンが分泌され、これらがターゲット細胞に結合し、次に内部（第２次）メッセンジャーが放出され、これが細胞核に結合し、ＲＮＡ合成・タンパク合成、細胞分裂、タンパク活性、などを経て更なる経路へ続いてゆくと考えられている。本発明では、メラニン形成過程において、カルシウムイオンが第２次メッセンジャーとして関与していることを見出し、そのカルシウムイオン量を制御することにより、メラニン合成が抑制できることを見出した。
【０００６】ＵＶ照射による細胞内のカルシウム量の変化を測定したところ、ＵＶ照射した細胞内のカルシウム量が、ＵＶ照射していない細胞内のカルシウム量に比べ、明らかに増加していることを確認した。
また、細胞内カルシウムを７０μＭＣａまたは１０００μＭＣａに調整し、ＵＶ照射していないもの、ＵＶ照射したもの、アシルグリセロールを添加したもので、メラニン量を測定したところ、細胞内カルシウムが多いほど、ＵＶまたはアシルグリセロール誘発性のメラニン合成が増加する事を確認した。
【０００７】さらにボルジン誘導体が細胞のアドレナリンアンタゴニストとして作用すると、カルシウムイオンの流入阻害がおこり、その結果、細胞内のタンパクキナーゼＣの活性阻害がおこり、これによりチロシナーゼの活性が阻害されることを見出し、従来の美白剤とは異なる作用機構により美白効果を発揮する美白剤を見出した。ボルジン誘導体は、従来の美白剤とは異なる作用機構で効果を示すので、従来の美白剤と併用することにより相乗効果を発揮することもわかった。

【発明の実施の形態】以下で、本発明を実施例等を用いてさらに具体的に説明するが、本発明はこれら実施例等により何ら限定されるものではない。本発明の実施例等で用いる手段などは以下の通りである。
【０００９】本発明に使用されるボルジン誘導体は、ジメチル６−メチル−ノル−アポルフィン、別名ボルジンと呼ばれるようなノル−アポルフィン誘導体である。本発明のボルジン誘導体を単独または少なくとも１種以上の他の美白有効成分に加えることを特徴とし、化粧料におけるボルジン誘導体の含有量は、０．０００１重量％から５重量％、好ましくは０．００４重量％から１重量％である。
【００１０】ボルジン誘導体に併用する他の美白剤としては、アルブチン、コウジ酸、アスコルビルリン酸ナトリウム（商品名ＳＴＡＹ−Ｃ５０、ロシュ・ビタミン・ジャパン製）などのアスコルビン酸およびその誘導体、チンピエキス（商品名メラスロー、クローダジャパン製）、ウワウルシエキス、ミトラカーバススケーバーエキス（商品名エチオリン、クローダジャパン製）、ジチアオクタンジオール、グルコン酸、β−カロチン（商品名メラクリア、クローダジャパン製）、グルタチオン、カミツレエキス、甘草エキス、チョウジエキス、茶抽出物、アスタキサンチン、牛胎盤エキス、トコフェロールおよびその誘導体、トラネキサム酸およびその塩、アズレン、γ−ヒドロキシ酪酸等がある。これら美白剤の含有量は、０．００１重量％から５重量％、好ましくは０．００４重量％から１重量％である。
【００１１】本発明の美白化粧料とは、外皮に適用されるすべての化粧品を意味し、その形態も水溶液系、可溶化系、乳化系、粉末系など幅広い形態をとりうる。本発明の美白化粧料には、その効用を損なわない範囲において、通常、化粧料に用いられる任意の成分を配合することができる。
