【発明の属する技術分野】
本発明は、ウリ科植物果実搾汁液を濃縮する工程で搾汁液より分離して得られる液体(以下、この液体をウリ科植物果実水と言うことがある)を有効成分とする老化防止剤又は細胞賦活剤、及びこれを含有する皮膚外用剤に関する。

【従来の技術】
従来より、乳液、クリーム、化粧水、パック、洗浄料、分散液、軟膏、液剤、エアゾール、貼付剤、パップ剤、リニメント剤等の皮膚外用剤には、これらに所定の薬効を付与することを目的として薬効剤が加えられている。そして、皮膚の老化防止、すなわち加齢、紫外線曝露等により生じる皮膚のしわやたるみ、張りや弾力性の低下を予防あるいは改善するために、例えばビタミンAや大豆抽出物、海藻抽出物等の細胞賦活剤が加えられている。
しかしながら、これらの細胞賦活剤は、目的の効果を得ようとして製剤に多量に配合すると、変色したり変質するなど製剤に悪影響を与えたり、あるいは所期の薬効が得られない場合があり、その改善が望まれていた。

【発明が解決しようとする課題】
本発明は、上記の事情に鑑みなされたもので、製剤中に多量に配合しても経時的に安定であり、製剤の外観等にも悪影響を与えずに使用することができ、細胞賦活効果又は老化防止効果が高く発揮できる細胞賦活剤又は老化防止剤を提供することを目的とする。また、この細胞賦活剤、老化防止剤を含有する皮膚外用剤を提供することを目的とする。

【課題を解決するための手段】
本発明者等は、従来の製剤中の水と同様に、多量に配合しても経時的に安定で、製剤の外観等に悪影響を与えずに使用することができ、かつ効果を高く発揮することが可能な薬効剤について鋭意検討した結果、ウリ科植物果実搾汁液を濃縮する工程又は蒸留する工程で搾汁液より分離して得られる液体が細胞賦活効果を有し、老化防止剤として優れたものであることを見出した。更に、このような効果を有するウリ科植物果実水は、皮膚外用剤中に用いられる従来の精製水と同様に使用することが可能な上、他の薬効剤と組み合せることにより、皮膚外用剤としてより優れた細胞賦活効果、老化防止効果が得られることを見出し、本発明を完成した。

すなわち本発明は、ウリ科植物果実搾汁液を濃縮する工程で搾汁液より分離して得られる液体(以下、ウリ科植物果実水と言うことがある)を有効成分とする老化防止剤である。また、ウリ科植物果実搾汁液を蒸留する工程で搾汁液より分離して得られる液体(以下、ウリ科植物果実水と言うことがある)を有効成分とする老化防止剤である。更に、本発明は、ウリ科植物果実搾汁液を濃縮する工程で搾汁液より分離して得られる液体を有効成分とする細胞賦活剤である。また、ウリ科植物果実搾汁液を蒸留する工程で搾汁液より分離して得られる液体を有効成分とする細胞賦活剤である。上記のウリ科植物果実搾汁液としてメロン果実搾汁液が好ましく用いられる。すなわち、本発明はメロン果実搾汁液を濃縮する工程又は蒸留する工程で搾汁液より分離して得られる液体(以下、メロン水と言うことがある)を有効成分とする老化防止剤又は細胞賦活剤である。更に本発明は、この老化防止剤又は細胞賦活剤を含有することを特徴とする皮膚外用剤である。

(A)上記の老化防止剤又は細胞賦活剤、
(B)他の細胞賦活剤、抗酸化剤、保湿剤及び紫外線防止剤から選ばれる薬効剤の一種または二種以上、

【発明の実施の形態】
本発明に用いられるウリ科植物果実は、トウガン属のトウガン(Benincasa hispida Cogn.)、スイカ属のスイカ(Citrullus vulgaris Schrad.)、コロシントウリ(Citrullus colocynthis Schrad.)、キュウリ属のメロン(Cucumis melo L.)、マクワウリ(Cucumis melo L.var.makuwa Makino)、キュウリ(Cucumis sativus L.)、カボチャ属のカボチャ(Cucurbita moschata Duch.)、テッポウウリ属のテッポウウリ(Ecballium elaterium A.Richard)、アマチャツル属のアマチャツル(Gynostemma pentaphyllum Makino)、ヘムスレヤ属のヘムスレヤ・アマビリス(Hemsleya amabilis Diels)、ユウガオ属のユウガオ(Lagenaria siceraria Standl.)、ヘチマ属のヘチマ(Luffa cylindrica Roem.)、ツルレイシ属のツルレイシ(Momordica charantia L.)、モクベツシ(Momordica cochinchinensis Spr.)、カラスウリ属のカラスウリ(Trichosanthes cucumeroides Maxim.)などであり、それらの産地は特に限定されない。

本発明のウリ科植物果実としてメロン果実が好ましく用いられるが、メロン(Cucumis melo L.)は、果実を食用に利用されるウリ科のつる性の植物で、植物学的に多くの変種からなることが知られている。大きく分けて雨量の少ない地方で発達した欧米系のメロン類と湿潤な地方で発達をした東洋系のマクワウリに分けられる。メロンは品種の分化や品種群間の交配を通じて多くの品種が作られることが知られているが、通常、分類的に、Cucumis melo var. reticulatus Naud、Cucumis melo var. cantalupensis Naud 、Cucumis melo var. indorus Naudの3変種に分けられる欧米系のメロン類相互の交配、あるいはマクワウリとの交雑品種をメロンと総称するが、本発明に用いられるメロン果実の種類や産地は特に限定されない。

ウリ科植物果実搾汁液は、一般に、ウリ科植物果実を絞って得られるものであるが、搾汁までの過程で必要に応じて水を加えてもよく、更に必要に応じてパルプ分離処理、脱臭脱色処理等を施して得ることもできる。本発明で用いるウリ科植物果実水は、ウリ科植物果実搾汁液を濃縮又は蒸留する工程で搾汁液より分離して得られる液体であり、従来は廃棄されていたものである。ウリ科植物果実水を作成するに当たってのウリ科植物果実搾汁液濃縮方法は特に限定されないが、例えば減圧蒸留により濃縮する工程又は蒸留する工程で生じる蒸気留分を一か所に集め冷却したものを挙げることができる。

本発明のウリ科植物果実水を有効成分とする老化防止剤又は細胞賦活剤は、常法に従い通常の皮膚外用剤に使用される種々の形態の基剤に配合し、製剤化することにより皮膚外用剤を得ることができる。本発明の老化防止剤又は細胞賦活剤の有効成分であるウリ科植物果実水の皮膚外用剤中における含有量は0.1質量%(以下単に「%」で示す)以上が好ましい。含有量が0.1%以上であれば、本発明で目的とする細胞賦活効果、老化防止効果を発揮することができる。本発明の老化防止剤又は細胞賦活剤の有効成分であるウリ科植物果実水は、各種形態の皮膚外用剤において従来の精製水の代わりに用いることもできる。その場合には、ウリ科植物果実水は水としての役割を発揮しつつ、細胞賦活効果、老化防止効果を発揮する。また、本発明の老化防止剤又は細胞賦活剤は、更に他の薬効剤と組み合せることにより、より細胞賦活効果、老化防止効果の優れた皮膚外用剤が得られる。

本発明において、ウリ科植物果実水を有効成分とする老化防止剤又は細胞賦活剤(成分(A))と組み合わせて使用する他の薬効剤(成分(B))は、ウリ科植物果実水以外の他の細胞賦活剤、抗酸化剤、保湿剤及び紫外線防止剤から選ばれるものである。これらは一種又は二種以上を組み合わせて用いることができる。これらの薬効剤としては、例えば、それぞれ以下に示すものが挙げられる。ここで「誘導体」には形成可能なエステルや塩が含まれる。なお、植物名の後のかっこ内にその植物の別名、生薬名などを記載した。また、薬効を二種以上有するものは重複して記載した。

本発明の皮膚外用剤における上記成分(B)、すなわち成分(A)以外の細胞賦活剤、抗酸化剤、保湿剤、紫外線防止剤の皮膚外用剤の含有量は、薬効剤の種類により相違するが、以下に示す範囲とすることが好ましい。この範囲であれば、成分(A)の老化防止剤、細胞賦活剤と組み合わせた場合、皮膚外用剤及び皮膚外用剤中の成分(A)の老化防止剤、細胞賦活剤の経時安定性に影響を及ぼすことがなく、相乗的により高い老化防止効果を発揮させることができる。

アルコールは、溶解、清涼感、防腐、保湿等の目的で、エチルアルコール等の一価アルコールや、多価アルコールを用いることができる。油剤は、使用性、使用感を良くするものとして、その由来、性状は問わず使用することができる。例えば流動パラフィン、スクワラン、トリグリセライド油、エステル油、ロウ類、脂肪酸類、高級アルコール、シリコン油、フッ素系油、各種ワックス等である。界面活性剤は、油剤等の乳化や可溶化等のために用いられ、陰イオン性、陽イオン性、非イオン性及び両性の活性剤を用いることができる。増粘剤としては、カルボキシビニルポリマー、カラギーナン、寒天、キサンタンガム、デキストリン脂肪酸エステル、有機変性粘土鉱物等、化学合成品又は天然物由来に関わらず用いることが可能である。また、これらの成分を系の粘度調整だけでなく、ゲル化、保湿、皮膜形成等のために用いることもができる。

粉体としては、形状や粒子の大きさ、多孔性の有無、結晶構造等を問わず、使用性や使用感を良くするために、複合化や表面処理を行なったものでもよい。タルク、マイカ、セリサイト、無水ケイ酸等の無機粉体、ナイロンパウダー等の有機粉体、魚鱗箔、オキシ塩化ビスマス等のパール顔料、酸化鉄、カーボンブラック、群青等の無機顔料、タール色素及びそのレーキ、天然色素等が用途に応じて用いられる。系中の成分の品質劣化を防ぐために、EDTA等のキレート剤、乳酸−乳酸ナトリウム等のバッファーによるpH調整剤を用いることもできる。

常法で破砕したメロン果実破砕物に、ペクチナーゼ及び水を原料果実に対してそれぞれ0.01%及び40%になるように添加し、一晩放置した。翌日、この果実破砕物を油圧プレス機を用いて圧搾搾汁し、次いで一旦加熱した後遠心分離を行ない、メロン果実搾汁液を上澄み液として得た。このメロン果実搾汁液を真空濃縮機を用いて減圧濃縮した。このとき蒸発する留分を復水器で冷却し液化して集め、メロン水を得た。

試験例1 細胞培養による細胞賦活試験
ヒト新生児由来の線維芽細胞NB1RGBを使用した。24穴プレートに培地を適量とり、線維芽細胞NB1RGBを播種し、37℃、二酸化炭素濃度5vol%の雰囲気中にて静置した。翌日、参考例1で得たメロン水及び参考例2で得たスイカ水をそれぞれ最終濃度が0μL/mL(対照)、1μL/mL、10μL/mL、100μL/mLとなるように検体調製液を添加し混和した。培養4日目に培地を交換し、再度検体調製液を添加した。翌日、培地を除き、細胞をリン酸緩衝液にて洗浄した後回収し、各検体調製液で生育させた線維芽細胞NB1RGBの細胞数を対照と比較し細胞増殖率として評価した。
また、比較例としてすでに細胞賦活作用のあることが知られている大豆抽出物(参考例3)についても同様の試験を行った。
