Claims (1)
- 【特許請求の範囲】 1.ブラジキニンB1レセプター又はその機能的誘導体をコードする単離精製D NA分子。 2.ヌクレオチド配列: (配列番号1)又はその機能的誘導体を有する請求項1に記載の単離精製DNA 分子。 3.前記DNA分子がゲノムDNAである請求項1に記載の単離精製DNA分子 。 4.組換え宿主でブラジキニンB1レセプターを発現させるための発現ベクター であって、ブラジキニンB1レセプター又はその機能的誘導体をコードする組換 え遺伝子を含む前記発現ベクター。 5.発現ベクターがヌクレチオチド配列: (配列番号1)を有するブラジキニンB1レセプター又はその機能的誘導体をコ ードするクローン化遺伝子を含む請求項4に記載の発現ベクター。 6.発現ベクターがブラジキニンB1レセプターをコードするゲノムDNAを含 む請求項4に記載の発現ベクター。 7.ブラジキニンB1レセプター又はその機能的誘導体をコードする組換えクロ ーン化遺伝子を含む組換え宿主細胞。 8.ブラジキニンB1レセプターをコードする前記遺伝子がヌクレチオチド配列 : (配列番号1)又はその機能的誘導体を有する請求項7に記載の組換え宿主細胞 。 9.ブラジキニンB1レセプターをコードする前記クローン化遺伝子がゲノムD NAである請求項7に記載の組換え宿主細胞。 10.ブラジキニンB1レセプターとして機能する実質的に純粋な形態のタンパ ク質。 11.アミノ酸配列:(配列番号2)又はその機能的誘導体を有する請求項10に記載のタンパク質。 12.ブラジキニンB1レセプターに対して免疫反応性の単一特異抗体。 13.抗体がブラジキニンB1レセプターの活性を阻止する請求項12に記載の 抗体。 14.組換え宿主細胞におけるブラジキニンB1レセプタータンパク質の発現方 法であって、 (a)請求項4に記載の発現ベクターを適切な宿主細胞に移入する段階と、 (b)発現ベクターからブラジキニンB1レセプタータンパク質を発現させる条 件下で段階(a)の宿主細胞を培養する段階を含む前記方法。 15.ブラジキニンB1レセプター活性を調節する化合物の同定方法であって、 (a)ブラジキニンB1レセプター活性の推定モジュレーターをブラジキニンB1 レセプターと結合する段階と、 (b)レセプターに及ぼすモジュレーターの作用を測定する段階を含む前記方法 。 16.レセプターに及ぼすモジュレーターの作用がB1レセプターリガンドの結 合の阻害又は強化である請求項15に記載の方法。 17.レセプターに及ぼすモジュレーターの作用がB1レセプターにより媒介さ れるシグナル形質導入の刺激又は阻害である請求項15に記載の方法。 18.レセプターに及ぼすモジュレーターの作用がB1レセプターにより媒介さ れるシグナル形質導入であり、前記シグナル形質導入がホスファチジルイノシト ール加水分解、細胞内貯蔵Ca2+の放出及びアラキドン酸放出から構成される群 から選択される請求項17に記載の方法。 19.ブラジキニンB1レセプターのモジュレーターである請求項15に記載の 方法で活性な化合物。 20.ブラジキニンB1レセプターのアゴニスト又はアンタゴニストである請求 項15に記載の方法で活性な化合物。 21.ブラジキニンB1レセプターの発現モジュレーターである請求項15に記 載の方法で活性な化合物。 22.ブラジキニンB1レセプター活性のモジュレーターである請求項15に記 載の方法で活性な化合物を含む医薬組成物。 23.ブラジキニンB1レセプターにより媒介される症状の治療を必要とする患 者の治療方法であって、請求項15に記載の方法で活性なブラジキニンB1レセ プター調節化合物を投与することからなる前記方法。 24.ブラジキニンB1レセプターにより媒介され且つ痛覚過敏、急性炎症又は 慢性炎症により特徴付けられる症状の治療を必要とする患者の治療方法であって 、請求項15に記載の方法で活性なブラジキニンB1レセプター調節化合物を投 与することからなる前記方法。 25.ブラジキニンB1レセプター活性を調節する化合物の同定方法であって、 (a)ブラジキニンB1レセプター活性の推定モジュレーターを、組換えブラジ キニンB1レセプターを発現する細胞と結合する段階と、 (b)レセプターに及ぼすモジュレーターの作用を測定する段階を含む前記方法 。 26.段階(b)でレセプターに及ぼすモジュレーターの作用がB1レセプター リガンドの結合の阻害又は強化である請求項25に記載の方法。 27.段階(b)でレセプターに及ぼすモジュレーターの作用がB1レセプター により媒介されるシグナル形質導入の阻害又は強化である請求項25に記載の方 法。 28.シグナル形質導入がホスファチジルイノシトール加水分解、細胞内貯蔵C a2+の放出及びアラキドン酸放出から構成される群から選択される請求項27に 記載の方法。 29.ブラジキニンB1レセプターのモジュレーターである請求項25に記載の 方法で活性な化合物。 30.ブラジキニンB1レセプターのアゴニスト又はアンタゴニストである請求 項25に記載の方法で活性な化合物。 31.ブラジキニンB1レセプターの発現モジュレーターである請求項25に記 載の方法で活性な化合物。 32.ブラジキニンB1レセプター活性のモジュレーターである請求項25に記 載の方法で活性な化合物を含む医薬組成物。 33.ブラジキニンB1レセプターにより媒介される症状の治療を必要とする患 者の治療方法であって、請求項25に記載の方法で活性なブラジキニンB1レセ プター調節化合物を投与することからなる前記方法。 34.ブラジキニンB1レセプターにより媒介され且つ痛覚過 敏、急性炎症又は慢性炎症により特徴付けられる症状の治療を必要とする患者の 治療方法であって、請求項25に記載の方法で活性なブラジキニンB1レセプタ ー調節化合物を投与することからなる前記方法。
