Claims (1)
- 【特許請求の範囲】 1.基本認識ユニットから伸びた露出したループ構造を特徴とする、標的分子 の活性部位またはクレフトと相互作用する能力を有する、可能な認識分子。 2.ループ構造が、標的分子の活性部位またはクレフトに対する結合特異性を 有するラクダ科の重鎖抗体のCDR3またはこのようなCDR3の誘導バージョ ンである、請求項1記載の認識分子。 3.CDR3の誘導バージョンが、少なくとも一つの天然アミノ酸が、1個ま たはそれ以上の他のアミノ酸によって置換されている突然変異CDR3である、 請求項1または2記載の認識分子。 4.CDR3の誘導バージョンが、1個またはそれ以上の追加のアミノ酸が付 加されるかおよび/またはその天然アミノ酸配列内に組み込まれている突然変異 CDR3である、請求項1または2記載の認識分子。 5.基本認識ユニットが、標的分子に対する結合親和性をもつ抗体型構造によ って形成されている、請求項1から4に記載の認識分子。 6.抗体型構造が、ラクダ科重鎖抗体またはその修飾バージョンの少なくとも 一部によって形成されている、請求項5に記載の認識分子。 7.ラクダ科抗体の修飾バージョンが、少なくとも一つの天然アミノ酸が、1 個またはそれ以上の他のアミノ酸によって置換されているバージョンである、請 求項6に記載の認識分子。 8.ラクダ科抗体の修飾バージョンが、1個またはそれ以上の追加のアミノ酸 が付加されるかおよび/またはその天然アミノ酸配列内に組み込まれているバー ジョンを含む、請求項6または7に記載の認識分子。 9.ラクダ科抗体の修飾バージョンが、第二のアミノ酸配列に融合しているバ ージョンである、請求項6、7または8に記載の認識分子。 10.ラクダ科抗体の修飾バージョンが、生物学的に活性な分子に連結してい るバージョンである、請求項6、7または8に記載の認識分子。 11.標的分子の生物学的機能の中和において使用する、請求項1から10に 記載の認識分子。 12.治療、診断、ワクチンおよび標的分子の単離または精製において使用す る、請求項1から10に記載の認識分子。 13.標的分子が、細菌毒素、ヘビ毒液由来の毒素、ミツバチ毒液由来の毒素 、クモ毒素、酵素、特にウイルスおよび細菌の酵素、ならびに受容体から選ばれ る、請求項11または12に記載の認識分子。 14.1種またはそれ以上の請求項1から13に記載の認識分子および適当な 賦形剤を含む、治療用組成物。 15.1種またはそれ以上の請求項1から13に記載の認識分子含む、診断用 試験キット。 16.1種またはそれ以上の請求項1から13に記載の認識分子を含むワクチ ン。 17.担体に結合した1種またはそれ以上の請求項1から13に記載の認識分 子を有する該担体からなる精製材料。 18.担体がカラム材料であり、好ましくは親和性カラムである、請求項17 記載の精製材料。 19.抗原に特異的である、請求項1から13に記載の認識分子の製造方法で あって: a)ラクダ科重鎖抗体を準備し; b)コーディング配列を単離し、ファージディスプレイベクターにおいてクロ ーニングし; c)該ベクターを宿しているファージにおいて該コーディング配列を発現し; 次いで d)ファージを固定抗原で選り分けることによって抗原に特異的な認識分子を 選択すること、を特徴とする方法。 20.抗原に特異的である、請求項1から13に記載の認識分子の製造方法で あって: a)ラクダ科重鎖抗体をランダムに選び; b)コーディング配列を単離し、ファージディスプレイベクターにおいてクロ ーニングし; c)該コーディング配列を少なくともひとつのコドンにおいてランダムに置換 することにより修飾し; d)ファージディスプレイベクターにおいてランダムに突然変異したコーディ ング配列のライブラリを作成し; e)該ベクターを宿しているファージにおいて該コーディング配列を発現し; 次いで f)ファージを固定抗原で選り分けることによって抗原に特異的な認識分子を 選択する、ステップを特徴とする方法。 21.抗原に特異的である、請求項1から13に記載の認識分子の製造方法で あって: a)認識分子またはその先駆体をコードするDNA配列を単離し; b)必要に応じて、1個またはそれ以上の塩基置換、欠失または挿入を導入す ることによって該分子またはその先駆体を修飾し; c)このようにして得られた、必要に応じて修飾された、DNA配列を適当な 宿主に移入し;次いで d)宿主内でDNA配列を発現する、ステップを特徴とする方法。
