- 【特許請求の範囲】 1.特定の酵素活性を有するクローンを同定するための方法であって、 (i)特定の酵素活性を有するタンパク質をコードする核酸配列の少なくとも 一部を含む少なくとも一種の核酸または核酸様ハイブリダイゼーションプローブ を使用することにより、少なくとも二種の微生物から得た核酸集団から標的核酸 を選択及び回収し、 (ii)回収した標的核酸を用いて宿主を形質転換して、特定の活性についてス クリーニングされる当初の核酸集団のサブセットを含むクローンのライブラリー を作製すること により調製したクローンのライブラリー中で該特定の酵素活性についてスクリー ニングすることを含む前記方法。 2.前記の微生物の核酸から得られる核酸が、 二本鎖核酸を一本鎖核酸に変換し、 変換された一本鎖核酸から、プローブ核酸にハイブリダイズする標的一本鎖 核酸を回収し、 宿主を形質転換するために回収した標的一本鎖核酸を二本鎖核酸に変換する ことにより選択される請求項1に記載の方法。 3.前記プローブを固相に直接または間接的に結合し、これによりそのようにハ イブリダイズしない一本鎖核酸からそれを分離する請求項2に記載の方法。 4.一本鎖核酸を前記プローブから遊離させ、その遊離した一本鎖核酸を二本鎖 核酸に変換する前に増幅する工程をさらに含む請求項3に記載の方法。 5.前記標的核酸が遺伝子クラスターまたはその部分をコードする請求項1に記 載の方法。 6.少なくとも一種の微生物から得られた核酸ライブラリーを選択手順にかけて そこから特定のタンパク質活性を有するタンパク質をコードする核酸配列の全部 あるいは一部である一種以上のプローブ核酸配列にハイブリダイズする二本鎖核 酸を選択する方法であって、 (a)ハイブリダイゼーションを可能とする条件下で、二本鎖核酸集団を、リ ガ ンドに結合した核酸プローブと接触させて、プローブとそれにハイブリダイズす る核酸集団のメンバーの複合体を生成させ、 (b)(a)の複合体を前記リガンドの固相特異的結合相手と接触させて、固相複 合体を生成させ、 (c)(b)の未結合核酸集団から固相複合体を分離し、 (d)プローブから固相結合プローブに結合した集団のメンバーを遊離させ、 (e)(d)の二本鎖核酸を適当な宿主に導入して、選択された核酸を含む複数の クローンを含むライブラリーを形成し、 (f)該ライブラリーを特定のタンパク質活性についてスクリーニングするこ とを含む前記方法。 7.前記標的核酸が遺伝子クラスターまたはその部分をコードする請求項6に記 載の方法。 8.上記(a)の前に、さらに (i)所与のクラスのタンパク質にユニークな(i)のコンセンサス配列に相補的 なリガンド結合オリゴヌクレオチドプローブに、ハイブリダイゼーションを可能 とする条件下で(a)の二本鎖核酸集団を接触させて二本鎖複合体を形成させ、 (ii)(i)の複合体を該リガンドの固相特異的結合相手と接触させて固相複合 体を形成し、 (iii)該固相複合体を未結合核酸集団から分離し、 (iv)前記固相結合プローブに結合した集団のメンバーを遊離させ、 (v)該固相結合プローブをそれに結合した集団のメンバーから分離する 工程を含む請求項6に記載の方法。 9.異種核酸集団に由来する特定のタンパク質活性を有するタンパク質を得るた めの方法であって、該特定の活性が生じるように改変あるいは突然変異誘発され た異種核酸集団からの核酸を含むクローンのライブラリーを、前記特定の酵素活 性についてスクリーニングすることを含む方法。 10.前記突然変異誘発の前に、前記特定の活性と同じであってもよく異なっても よい共通の特徴をコードする核酸配列を含む核酸を異種核酸集団から選択的に回 収することをさらに含む請求項9に記載の方法。 11.核酸調製物の回収が、核酸集団を前記配列の少なくとも部分の特異的結合相 手と接触させることを含む請求項10に記載の方法。 12.特異的結合相手が固相結合ハイブリダイゼーションプローブである請求項11 に記載の方法。 13.前記共通の特徴が酵素活性の一種である請求項10に記載の方法。 14.前記酵素活性の一種を示す異種核酸集団からの核酸を含むクローンから核酸 を回収することを含む請求項13に記載の方法。 15.前記突然変異誘発が部位指向またはランダム指向突然変異誘発である請求項 9に記載の方法。 16.突然変異誘発にかける前に、前記特定の活性と同じであってもよく異なるも のであってもよい活性について前記クローンのライブラリーを予備スクリーニン グすることを含む請求項9に記載の方法。 17.対象となるタンパク質の発現について前記クローンを予備スクリーニングす ることを含む請求項16に記載の方法。 18.特定のタンパク質活性を有するタンパク質を得るための方法であって、その 特定のタンパク質活性と同じであってもよく異なるものであってもよい一以上の 所望の特徴を有するタンパク質をコードする核酸をクローンのライブラリーで生 成する試みにおいて突然変異誘発にかけられた核酸集団のプールからの核酸を含 むクローンのライブラリーを、その特異的な活性についてスクリーニングするこ とを含む前記方法。 19.前記突然変異誘発の前に、特定のタンパク質活性と同じであってもよく異な るものであってもよい少なくとも1つの共通のタンパク質の特徴をコードする核 酸配列を含む核酸を前記異種核酸集団から選択的に回収することをさらに含む請 求項18に記載の方法。 20.前記少なくとも1つの共通の特徴が少なくとも一の通常の酵素活性である請 求項19に記載の方法。 21.核酸調製物の回収が、核酸集団を、前記配列の少なくとも部分の特異的結合 相手と接触させることを含む請求項20に記載の方法。 22.前記特異的結合相手が固相結合ハイブリダイゼーションプローブである請求 項 21に記載の方法。 23.酵素活性の一種を示す異種核酸集団からの核酸を含むクローンから核酸を回 収することを含む請求項22に記載の方法。 24.前記突然変異誘発が部位指向またはランダム突然変異誘発である請求項18に 記載の方法。 25.低温において改良された酵素活性を有し、酵素からなる群から選択されるメ ンバーである熱安定性酵素、または該酵素をコードするポリヌクレオチドを提供 する方法であって、 (a)少なくとも60℃の温度で安定な少なくとも一種の酵素を突然変異誘発に かけ、 (b)少なくとも60℃の温度で安定で、その至適温度範囲の少なくとも10℃低 い温度で酵素活性を有し、工程(a)の酵素よりも高い活性を有する突然変異酵素 、または突然変異酵素をコードするポリヌクレオチドについて(a)で得られた突 然変異体をスクリーニングすること を含む前記方法。 26.少なくとも一種の微生物から得られた核酸ライブラリーを選択手順にかけて そこから特定のタンパク質活性を有するタンパク質をコードする核酸配列の全部 あるいは一部である一種以上のプローブ核酸配列にハイブリダイズする二本鎖核 酸を選択する方法であって (a)ハイブリダイゼーションを可能とする条件下で、二本鎖核酸集団を、リ ガンドに結合した核酸プローブと接触させて、プローブとそれにハイブリダイズ する該核酸集団のメンバーの複合体を生成させ、 (b)未結合核酸集団から該複合体を分離し、 (c)該プローブから該プローブに結合した集団のメンバーを遊離させ、 (d)(c)の二本鎖核酸を適当な宿主に導入して、選択された核酸を含む複数の クローンを含むライブラリーを形成し、 (e)該ライブラリーを該特定のタンパク質活性についてスクリーニングする ことを含む前記方法。
