Claims (11)
1. Postupak proizvodnje transgene biljke ili transgenog biljnog tkiva,
naznačen time, da postupak obuhvaća sljedeće:
pripravljanje molekule nukleinske kiseline koja obuhvaća najmanje dva područja sekvence nukleinske kiseline kojima nedostaje homologna sekvenca s genomskom DNK od biljne stanice domaćina, i najmanje jedno mjesto za prepoznavanje ciljne endonukleaze, pri čemu najmanje dva područja sekvence nukleinske kiseline kojima u biti nedostaje homologna sekvenca s genomskom DNK od biljne stanice domaćina, zakriljuju najmanje jedno mjesto prepoznavanja ciljne endonukleaze, gdje najmanje dva područja sekvence nukleinske kiseline kojima nedostaje homologna sekvenca s genomskom DNK od biljne stanice domaćina, ne obuhvaćaju otvoreni okvir za iščitavanje,
dok su pritom najmanje dva područja sekvence nukleinske kiseline kojima nedostaje homologna sekvenca s genomskom DNK od biljne stanice domaćina, odabrana od jedne ili više skupina koje se sastoje od sekvenci SEQ ID NO:1 do SEQ ID NO:14, te 17 ili 18, ili pri čemu najmanje dva područja sekvence nukleinske kiseline kojima nedostaje homologna sekvenca s genomskom DNK od biljne stanice domaćina, zajedno s mjestima ciljne endonukleaze, obuhvaćaju sekvence SEQ ID NO:15, 16 i 19;
transformiranje biljne stanice ili biljnog tkiva koja/e obuhvaća biljnu stanicu s molekulom nukleinske kiseline, gdje je molekula nukleinske kiseline stabilno integrirana u genom biljne stanice; i
generiranje biljnog tkiva ili regeneriranje biljke iz biljne stanice koja je transformirana s molekulom nukleinske kiseline.
2. Postupak prema zahtjevu 1, naznačen time, da molekula nukleinske kiseline nadalje obuhvaća neidentična mjesta restrikcije, a poželjno ta neidentična mjesta restrikcije obuhvaćaju kompatibilne krajeve jednostruke lančane strukture, koji dopuštaju konkatemerizaciju za višestrukih zemljanih podloga koje su podvrgnute genetskom inženjeringu (ELP – engineered landing pad).
3. Postupak prema zahtjevu 1 ili 2, naznačen time, da najmanje dva područja sekvence nukleinske kiseline kojima nedostaje homologna sekvenca s genomskom DNK od biljne stanice domaćina, obuhvaćaju od oko 50 bp do oko 3 kb.
4. Postupak prema bilo kojem od zahtjeva 1 do 3, naznačen time, da mjesto ciljne endonukleaze predstavlja mjesto nukleaze cinkovih prstiju.
5. Postupak prema bilo kojem od zahtjeva 1 do 4, naznačen time, da je molekula nukleinske kiseline stabilno integrirana nasumice u genomu biljne stanice, ili je stabilno integrirana u jedno ili više poznatih ciljnih mjesta u genomu biljne stanice, ili je stabilno integrirana u amplificirano područje kromosoma ili minikromosoma.
6. Postupak prema bilo kojem od zahtjeva 1 do 5, naznačen time, da je molekula nukleinske kiseline zakriljena na svakom kraju pomoću jednog ili više dodatnih mjesta prepoznavanja ciljne endonukleaze.
7. Postupak prema zahtjevu 6, naznačen time, da nadalje obuhvaća uvođenje u biljno tkivo ili u biljku, jedne ili više ciljnih endonukleaza koje prepoznaju jedno ili više dodatnih mjesta prepoznavanja ciljne endonukleaze, pri čemu se molekula nukleinske kiseline odstranjuje iz genoma biljnog tkiva ili biljke.
8. Postupak proizvodnje transgene biljke, naznačen time, da postupak obuhvaća sljedeće:
pripravljanje stanice ili tkiva izolirane(og) iz transgenog biljnog tkiva ili transgene biljke prema zahtjevu 3;
pripravljanje najmanje jedne ciljne endonukleaze ili prve molekule nukleinske kiseline koja obuhvaća sekvencu nukleinske kiseline koja kodira najmanje jednu ciljnu endonukleazu, pri čemu najmanje jedna ciljna endonukleaza prepoznaje najmanje jedno mjesto prepoznavanja ciljne endonukleaze koja je uvedena u genom transgene biljke;
pripravljanje druge molekule nukleinske kiseline koja obuhvaća sekvencu nukleinske kiseline od interesa, i dvije dodatne sekvence nukleinske kiseline koje zakriljuju sekvencu nukleinske kiseline od interesa, pri čemu je svaka od dviju dodatnih sekvenci nukleinske kiseline, homologna s jednim od dva područja sekvence nukleinske kiseline kojima nedostaje homologna sekvenca s genomskom DNK;
uvođenje (i) najmanje jedne ciljne endonukleaze ili prve molekule nukleinske kiseline; i (ii) druge molekule nukleinske kiseline u bilnju stanicu ili biljno tkivo, pri čemu je sekvenca nukleinske kiseline od interesa stabilno integrirana u genom biljne stanice; i
regeneriranje biljke iz biljne stanice ili biljnog tkiva transformirane(og) putem prve i druge molekule nukleinske kiseline.
9. Transgena biljka, naznačena time, da se proizvodi pomoću postupka prema bilo kojem od zahtjeva 1 do 8.
10. Sjeme, naznačeno time, da se proizvodi pomoću transgene biljke prema zahtjevu 9.
11. Postupak prema zahtjevu 7 ili 8, naznačen time, da ciljna endonukleaza predstavlja nukleazu cinkovih prstiju.
