Abstract

The invention comprises aza-bicycloalkyl-(N)-alkylguanidines of the general formula: <FORM:0910427/IV (b)/1> and their acyl derivatives, quaternary ammonium compounds and acid addition salts (including ascorbates), in which A stands for a 1-5 membered hydrocarbon chain which may also contain a double bond or be interrupted by a nitrogen atom which may be unsubstituted or substituted by an alkyl or aralkyl group, m represents any one of the numbers from 0 to 6, m1 any one of the numbers from 1 to 7, n and n1 any one of the numbers from 0 to 3. R and R1 stand for hydrogen atoms or C1-7-alkyl groups and G for a guanido group, with the proviso that CmH2m separates the bridgehead atoms by at most 2 carbon atoms, and in which <FORM:0910427/IV (b)/2> when A forms a 3-5 membered isocyclic ring with the bridge members and <FORM:0910427/IV (b)/3> when A forms a 4-membered azacyclic ring with the bridge members, and the preparation thereof by reacting an aza - bicycloalkyl - (N) - alkylamine corresponding to the above general formula but wherein G represents a primary or secondary amino group with S-methylthiourea or a mineral acid addition salt thereof. Acyl derivatives may be prepared from mono or dicarboxylic acids or sulphonic acids which may contain further substituents. Quaternary ammonium salts are preferably prepared from esters of lower alkanols or aralkanols. The guanidines of the above general formula are useful as antihypertensive agents, and pharmaceutical compositions comprise the said compounds in admixture or in conjunction with an inert carrier suitable for enteral or parenteral administration. The compositions may contain other therapeutically useful substances, and may be in the form of tablets, dragees, capsules, solutions, suspensions or emulsions. Specification 910,428 is referred to.
