Abstract
The invention relates to a high purity streptokinase effective for the treatment of thrombosis which is an amorphous solid, soluble in water and having a molecular weight of about 47,000, a sedimentation rate constant of about 3.0 x 10-13, a diffusion constant of about 6.20 and 10-7 and an electrophoretic mobility (m ) of 4.19 x 10-5cm2sec-1volt-1, the preecding physical properties being measured in phosphate buffer at pH 7.5 and an ionic strength of 0.15. Crude streptokinase is adsorbed on an ionizable cellulose-type chromotographic column, desorbing purified streptokinase therefrom by means of gradient elution by means of a phosphate buffer having a molarity of 0.1 to 0.005 M, having a pH of 5.8 to 8.5 and a temperature of 0 DEG C. to 10 DEG C., recovering the streptokinase from the eluate by treatment with conventional precipitating agents for streptokinase, adsorbing said purified streptokinase on a second ionizable cellulose type chromatographic column, desorbing further purified streptokinase from said second column by means of gradient elution using the aforesaid phosphate buffer and recovering said further purified streptokinase from the eluate by conventional precipitating agents for streptokinase. Specifications 714,803, 715,103, 715,110, 715,572 and 715,930 are referred to.ALSO:An intravenous solution effective for the treatment of thrombosis comprises an aqueous solution of a purified streptokinase prepared by adsorption on a cellulose anion exchanger and elution with a phosphate buffer (see Group IV (b)). Preservatives, colouring-agents and other non-therapeutic injectable substances may be present. 50,000 to 100,000 units of streptokinase per hour may be administered.
