Abstract

1507215 Process for producing α-globulin formulation GREEN CROSS CORP 19 Aug 1975 34465/75 Heading A5B [Also in Division C3] A process for producing an IRA (immunoregulatory α-globulin) preparation comprises treating an aqueous solution of human blood or an α 2 -globulin-containing fraction thereof with an acid at pH of 5.0 or less at ambient temperature for 30 minutes to 24 hours, filtering the treated solution with a filter capable of leaving in the filtrate a fraction having a M.W. of 13000 or less, removing impurities in the filtrate by adsorption on a weakly acid cation exchanger, adsorbing the IRA on an inorganic (including active carbon) adsorbent and eluting the IRA with an aqueous acid or organic solvent. The blood fraction may be plasma or placental blood, particularly Cohn's Fraction IV. The eluate may be further purified by extraction with a halogenated hydrocarbon or an ether. Any HB-Ag activity in the preparation may be inactivated by heating in the presence of a stabilizer which. may be a neutral amino acid, a sugar alcohol, a monosaccharide or disaccharide, which is removed by dialysis.
