Abstract

The invention comprises a process for the preparation of steroid compounds by contacting a 3-keto nuclear saturated steroid having from 18-21 atoms in the carbon skeleton with oxidizing enzymes of an organism of the genus Nocardia, and thereby producing a 3-keto- D 1,4steroid, a 3-keto- D 4-steroid or mixtures thereof. As species of the genus Nocardia there may be used N. convoluta, N. corallina, N. erythropolis, N. gardneri, N. minima, N. polychromegenes, N. opaca and N. globerula. As starting materials there are mentioned androstene-3-one, androstane-3,17-dione, bisnorcholane - 3 - one, pregnane - 3 - one, pregnane - 3,20-dione, 4-dihydrocorticosterone, 17a -hydroxyandrostane-3-one, estrane-3-one, 4-dihydrodesoxycorticosterone, allopregnane-3-one, allopregnane-3,20-dione, 17a -hydroxypregnane-3-one and 3,20-dion, 11b -hydroxypregnane-3-one and 3,20-dione, 20-hydroxypregnane-3-one, 14a -hydroxypregnane-3-one and 3,20-dione, pregnane and allopregnane - 3,11,20 - trione, 11b ,17a ,21 - trihydroxypregnane-3,20-dione, 17,21 - dihydroxypregnane - 3,11,20 - trione, 11b ,21 - dihydroxypregnane - 3,20 - dione, 11b ,14a ,17a ,21 - tetrahydroxypregnane - 3,20 - dione and 21-hydroxypregnane-3,20-dione. The oxidation of the steroid compound may be effected in the presence of the whole fermentation product, by cells removed from the growing culture and resuspended, or by oxidizing enzymes prepared from cells of the organism. Examples describe the preparation of prednisolone, prednisone, D 1,4-pregnadiene-21-ol-3,20-dione, desoxycorticosterone, and D 1,4-desoxycorticosterone.
