Abstract
A process for the manufacture of material containing antigens, obtained by the use of carcinogenic inactivating agents defined as to mean b -propiolactone or compounds of the formula <FORM:0806950/VI/1> where R is methyl, ethyl and normal or isopropyl, which includes the step of freeze drying in order to reverse excess of the carcinogenic agent. The material containing antigens is prepared by subjecting infectious micro-organisms to the action of the above inactivating agents, e.g. by subjecting a 20 per cent w/v. suspension of the micro-organism and animal tissue in sterile saline, to b -propiolactone or N-acetylethyleneimine in the form of an aqueous solution in which the concentration of said agent is between 0.005 per cent v/v. and 1 per cent v/v. based on the combined volumes of the, e.g. suspension of micro-organism and the solution of said agent. Sugars, e.g. sucrose may preferably be added to the material prior to freeze drying, which step is carried out at a low pressure, for example between 0.01 mm. and 0.0001 mm., during a period of several hours, e.g. 24 hours. The freeze-drying is conveniently carried out in ampoules. Specification 806,949 is referred to.
