Abstract
1397406 Determination of hydroxysteroids NYEGAARD & CO AS 19 June 1972 [22 June 1971] 29286/71 Heading G1B A reagent for determination of hydroxysteroids comprises a lyophilized mixture of a hydroxysteroid dehydrogenase, a coenzyme which is nicotinamide adenine dinucleotide or thionicotinamide adenine dinucleotide, and optionally a neutral buffer, a polyhydroxy compound, a chelating agent, and/or a thiol. The polyhydroxy compound may be a dextranor sugar (sucrose exemplified); the chelating agent may be EDTA; the thiol may be dithiothreitol (exemplified), mercaptoethanol, cysterine or glutathione. For determinations on urine, the sample is treated with conc-hydrochloric acid and formalin and the hydroxysteroids are extracted using first a non-polar then a polar solvent. The mixed extracts are evaporated to dryness and the residue dissolved in methanol for testing by mixing with the enzyme and co-enzyme. The buffer may provide pH 6-10, preferably 9-10, and may be a tris (2-amino-2-hydroxymethyl-1,3-propane diol)/hydrochloric acid buffer.
