Abstract

1340921 Sustained release compositions PHARMACIA AS 8 June 1971 [8 June 1970] 19387/71 Heading A5B Sustained release compositions in oral dosage form comprise a sintered porous matrix of a pharmaceutically acceptable thermoplastic material which is insoluble in gastrointestinal fluids, and particles of a physiologically active substance dispersed in the pores of the matrix, the particles carrying a coating of a pharmaceutically acceptable hydrophobizing material. The thermoplastic material may be paraffin wax, spermaceti, cetanol, a macrogol, carnauba wax or hydrogenated castor oil; the hydrophobizing material may be a silicone resin, glyceryl stearate or palmitate; and the physiologically active substance may be an inorganic salt such as potassium chloride, ferrous sulphate, magnesium sulphate or ammonium chloride; a vitamin, antipyretic, analgesic, antibiotic, diuretic (such as cyclothiazide) or psychopharmaca. The composition may be in the form of a tablet or pill comprising (a) a main body comprising a sintered porous matrix and the hydrophobized particles of a physiologically active substance and (b) a secondary tablet body, having a composition different from that of the main body, compressed into a surface part of the main body to form a unitary coherent combined tablet which may comprise in addition an exterior coating covering the unitary coherent combination tablet. An oral dosage form is prepared by coating particles of a physiologically active substance wholly or partially with a pharmaceutically acceptable hydrophobizing material, blending the coated particles with particles of a pharmaceutically acceptable thermoplastic material, compressing the blended mixture into the form of tablets or pills and thereafter subjecting the tablets or pills to a sintering treatment by heating to a temperature below the melting point of the thermoplastic material e.g. at 50-80‹C for ¢ to 3 hours. The tablet or pill may also be. applied with an exterior coating which may contain a physiologically active substance, in particular the core may contain potassium chloride and the coating a diuretic such as cyclothiazide.
