1292075 Anti-inflammatory substances ONO PHARMACEUTICAL CO Ltd 6 May 1970 [9 May 1969 26 Sept 1969] 21948/70 Heading A5B An anti-inflammatory substance obtained from a fermentation broth resulting from the cultivation of a microorganism of Aerobacter cloacae, Aerobacter aerogenes, Bacillus subtilis, Micrococus, lysodeikticus or Pseudomonas aeruginosa, and being odourless, tasteless, colourless, neutral with respect to pH, having a molecular weight higher than 200,000, soluble in water, insoluble in methanol, ethanol, acetone, benzene and ether, containing 5% ammonia nitrogen and 0.22% by weight phosphorus, giving positive m÷lisch, ninhydrin, biuret and chloroform-methanol extracted rhodamine stain reactions and negative xanthoproteic and direct rhodamine stain reactions, having an UV absorption spectrum showing absorption only at short waves and no maximum absorption at 280-260 mÁ and a specific rotation of [α]D<SP>26.5</SP> + 45‹ (c=1, water), may be prepared by cultivating a microorganism, as specified above, in a nutrient medium consisting of carbon and nitrogen sources and inorganic salts and recovering the anti-inflammatory substance from the resulting broth either (1) by precipitation from a filtrate of the fermentation broth by the addition of ethanol in the presence of potassium chloride or (2) by concentration of a filtrate of the fermentation broth, then dialysis in order to remove any low molecular weight substances and then subjection of the filtrate to gel-filtration in order to obtain a fraction containing the anti-inflammatory substance. In procedure (1), the potassium chloride may be added to the filtrate so that the concentration thereof is from 1 to 5% weight volume and then ethanol is added thereto in such an amount that the final alcohol concentration is from 40 to 80% weight/volume. The anti-inflammatory substance may be administered parenterally in the form of a saline solution.
