Abstract
A method for preparing a purified cold-water insoluble, neutral galactose polymeric constituent of agar from an agar which contains such a polymeric constituent and, in addition, contains calcium and a cold-water insoluble, sulphated polysaccharide, comprises rendering the sulphated polysaccharide cold water soluble by adding to the agar, under aqueous conditions, a solubilizing agent which is pectinase enzyme or a calcium sequestrant, and thereafter separating the cold-water insoluble constituent from the cold-water solubilized sulphated polysaccharide. Specified calcium sequestrants are dibasic sodium triphosphate, tribasic sodium phosphate, sodium pyrophosphate, sodium triphosphate, sodium hexametaphosphate, sodium tripolyphosphate, EDTA and alkali metal salts thereof. The purified product may be bleached, e.g. with sodium hypochlorite solution, and may be incorporated in culture medium compositions.
