Abstract
The invention comprises tetrahydro-benzo[a]-quinolizines of the formula <FORM:1016518/C2/1> in which R1 is an alkyl, aryl or haloaryl group, R2 is an alkyl, alkenyl or aralkyl group, each of the symbols R3, R4 and R5 represents an alkoxy group or one is such a group and the other two collectively are an alkylenedioxy group and the broken lines indicate bonds to optional R-groups and wherein the ring C is associated with one cyclic ethylenic bond, and acid-addition salts thereof. The compounds are prepared by reacting a 2-hydroxy-benzo[a]-quinolizine of the formula <FORM:1016518/C2/2> wherein R1 to R5 and the broken lines are as defined above, or an acid addition salt thereof, with a dehydrating agent, e.g. a mineral acid. 2 - Hydroxy - benzo[a]quinolizines of the Formula II are prepared by (a) reacting an appropriate 2 - oxo - hexahydro - benzo[a]-quinolizine with an appropriate metal-organic acetylene compound, hydrolysing the condensation product and completely hydrogenating the acetylene residue in the 2-position, to form compounds of the a -series, or (b) by reacting an appropriate 2-oxo-hexahydrobenzo[a]-quinolizine with a Grignard compound of the formula R1-Mg-halogen, and hydrolysing the condensation product. Pharmaceutical compositions having sedative and anti-emetic properties for enteral or parenteral administration comprise a compound of the invention together with a pharmaceutical carrier. The compositions may be formulated as tablets, dragees, capsules, suppositories, solutions, suspensions or emulsions.
