Description
Composition hémostatique et cicatrisanteHaemostatic and healing composition
The present invention relates to a haemostatic and healing composition. This composition is advantageously used in the treatment of wounds and ensures good haemostasis and good healing, as well as a rapid relief of pain in case of burning of the skin or insect sting.
The skin provides many functions, especially protection against external elements. A burn or wound breaks these functions, which are restored during healing.
It thus appears desirable to have compositions having a haemostatic and healing effect satisfactory and to soothe pain in case of burning or sting.
The present invention aims to achieve these objectives. The subject of the invention is thus a hemostatic and healing composition.
The composition according to the invention is in an aqueous and thermogelifiable form, in particular when in contact with the skin, and comprises a mixture of Aloe Vera, a water-soluble alginate salt, and a thermo-gelling polymer soluble in water.
Thus, advantageously, the thermo-gelling polymer allows the gelation of the product in contact with the skin, because of the temperature difference between the ambient temperature and the skin temperature. The gel obtained makes it possible to locally treat the zone of application of the composition, since it does not flow outside this zone. The specific combination of YAloe vera and water-soluble alginate salt helps to soothe pain caused by burns or punctures. It also stops bleeding and greatly promotes healing. The use of the composition is recommended on superficial burns, for example domestic, or in case of sunburn of the first and second degree, on superficial injuries of everyday life, by direct application of the composition on the injured area .
The composition is considered a class Mb device according to Rule 4 of Annex IX of Directive 93-42-EEC.
The composition according to the invention makes it possible to stop the bleeding, to quickly soothe the burns as well as the pains related to insect bites. It has the effect of a barrier between the damaged skin and the external environment. It also participates in the healing process. Aloe vera can be in the form of powder or gel, especially concentrated gel.
The powder can be obtained by lyophilization of Aloe Vera gel.
For example, Aloe vera leaves can be peeled to obtain the netting (the gelatinous inner part), which is rinsed to remove the latex containing the aloin, an irritant and very laxative compound. The tenderloin can then be crushed to obtain the Aloe vera juice that is concentrated. This concentrated gel can be dried by lyophilization to obtain the powder.
The water-soluble alginate salt is preferably a monovalent salt, typically sodium alginate, potassium alginate, lithium alginate or ammonium alginate. Alginic acid and its derivatives (base conjugates, salts and esters) alginates are polysaccharides mainly obtained from a family of brown algae: laminaria or fucus. Alginate is the common name for a family of polymers consisting of two monomers bonded together: mannuronate or mannuronic acid and guluronate or guluronic acid.
La formule générale de l’alginate est la suivante :The general formula of alginate is as follows:
The proportion and distribution of these two monomers are decisive for a broad expansion of the physical and chemical properties of alginate, so-called gluronic mannuronic ratio or M / G ratio. Its chemical composition varies among different species of algae, different parts of the same plant and is subject to seasonal changes. Nevertheless, by selecting raw materials with different properties, it is possible to manufacture a variety of alginate with constant characteristics.
Alginate products have intrinsic, natural, very effective properties against wound treatment. These products are in particular intended for hemostasis and cicatrization.
The term "thermo-gelling polymer" means a polymer which, at low concentration in water, is in liquid form (low viscosity) at room temperature and which gels at body temperature, the phenomenon of thermo-gelling being perfectly reversible. By "ambient temperature" is meant a temperature of between 18 and 25 ° C.
The thermo-gelling polymer may be chosen from the family of polyethers, copolymers of vinylcaprolactone or polyacrylamides.
The thermo-gelling polymers that may be used for the purposes of the invention may comprise, on the one hand, water-soluble units and, on the other hand, units having in water a lower critical demixing temperature (abbreviated as "LCST"). > for "lower critical solution temperature"). Below the LCST, the polymer is perfectly soluble in water, while above this temperature the LCST portions combine and lose their solubility in water, thus forming cross-linking nodes between the chains. polymers. The polymer then resembles a three-dimensional network, which leads to obtaining a gel. This association of LCST chains within the hydrophobic micro-domains beyond the demixing temperature being of a physical nature, the gelling phenomenon is then perfectly reversible. This gelation property with temperature is only possible if the polymer concentration is greater than the critical micelle concentration to allow interactions between the LCST units carried by different polymer chains.
The polymers used in the invention may be either block polymers or graft polymers which comprise, on the one hand, water-soluble units and, on the other hand, units with an LCST. These water-soluble units may be polymers of natural origin or of synthetic origin obtained by chain polymerization or by polycondensation.
The units with an LCST will preferably be chosen from: polyethers such as poly (propylene oxide) (POP) and its random and block copolymers with poly (ethylene oxide) (POE), N-substituted derivatives acrylamide such as poly (N-isopropylacrylamide) or poly (N-ethylacrylamide), polyvinylcaprolactam and copolymers of vinylcaprolactam.
Les polymères thermogélifiants susceptibles de convenir à l'invention peuvent être, par exemple, choisis parmi ceux décrits dans les demandes de brevets et brevets suivants : FR2694939, FR2788008, FR2820976, FR2856923, GB2408510, EP0629649, EP1307501, EPI407791, W097/00275, WO98/06438, W098/29487, W098/48768, W098/50005, WOOO/00222, WOOO/07603, WOOO/35961, WOOO/38851, WO01/41735, W002/032560, WO02/076392, W003/008462, WO03/106536, W004/006872, US2003/0099709, US6645476, US6689855, US6689856, US6870012 et US6878754.The thermogelling polymers that may be suitable for the invention may be, for example, chosen from those described in the following patent applications and patents: FR2694939, FR2788008, FR2820976, FR2856923, GB2408510, EP0629649, EP1307501, EPI407791, WO97 / 00275, WO98 / 06438, WO98 / 29487, WO98 / 48768, WO98 / 50005, WO00 / 00222, WO00 / 07603, WO00 / 35961, WO00 / 38851, WO01 / 41735, WO02 / 032560, WO02 / 076392, WO00 / 008462, WO03 / 106536 , W004 / 006872, US2003 / 0099709, US6645476, US6689855, US6689856, US6870012 and US6878754.
US 7339013.Particularly suitable thermogelling polymers are polyurethanes comprising poly (ethylene oxide-b-propylene oxide-b-ethylene oxide) (POE-b-POP-b-POE) groups such as those described in the patents and patent applications FR2840907, EP692506, EP1407791, WO 03 106536, US 7339013.
These polyurethanes are obtained by polycondensation of diisocyanates and POE-b-POP-b-POE triblock diols which are thermosensitive in a non-anhydrous medium and may comprise urea and / or allophanate groups. These polymers, known under the name of ExpertGel® and marketed by the company PolymerExpert, have many advantages, in particular: a low concentration of polymers is necessary for a significant increase in the viscosity as a function of the temperature, they have properties important lubricants, they are muco-adhesive, - they exhibit the same thermogelling behavior in a pH range from 1 to 12, they are compatible with salts, surfactants and most pharmaceutical and cosmetic excipients. By way of example, mention may be made of the thermo-gelling polymer EG230, which is a branched polyurethane containing POE-b-POP-b-POE units, whose INCI designation (International Nomenclature of Cosmetic Ingredients) in the English language is Bis -Methoxy PEG-13 PEG-502 / PPG-57 SMDI Copolymer.
Mention may also be made of the thermo-gelling polymer EG56, which is a polyurethane-urea based on POE-POP-POE containing about 75% POE, whose INCI designation (International Nomenclature of Cosmetic Ingredients) in the English language is Bis-Methoxy PEG-13 PEG-438 / PPG-110 SMDI Copolymer.
The Aloe vera content may be between 0.01 and 10% of the total weight of the composition, and preferably between 0.01 and 2% of the total weight of the composition.

The content of thermo-gelling polymer soluble in water may be between 1 and 10% of the total weight of the composition, and preferably between 3 and 8% of the total weight of the composition.
The composition may also comprise sodium chloride (as binder, thickener and masking agent), potassium sorbate (as a preservative), benzaikonium chloride (as preservative, antibacterial agent, antiseptic), menthol (feeling of freshness), eosin (as a dye, antiseptic) or chlorophyllin (as a dye, antioxidant), or a combination of these compounds.

The composition may be in various forms or packages, and especially in the form of an aerosol, an ampoule, especially a flexible bottom, an envelope provided with a breakable zone, a tube, or alternatively 'a flask. The invention also relates to a composition described above for hemostatic, healing, and / or soothing use. The invention is illustrated in a nonlimiting manner by the following example. The percentages are expressed by weight.
Exemple : préparation d’une composition hémostatique, cicatrisante et apaisante pouvant être déposée sous forme d’aérosol sur la peauExample: preparation of a haemostatic, healing and soothing composition that can be deposited in the form of an aerosol on the skin
On prépare la composition aqueuse thermo-gélifiante suivante :The following thermo-gelling aqueous composition is prepared:
The composition is prepared in the following manner. The polymer marketed under the name ExpertGel® 230 (EG230) is dissolved by PolymerExpert in the volume of demineralized water adapted, with mechanical stirring at 400 rpm, for 40 minutes. The EG230 polymer has its solubility increased in cold water. Once the EG230 polymer solubilized, the sodium alginate is added with stirring at 500 rpm for 20 minutes. Glycerol and NaCl are added successively, with the same stirring, for 5 and 10 minutes, respectively. Finally, YAloe vera solution is dissolved for 10 minutes at 500 rpm. The solution is ready and is sterilized quickly or stored in the refrigerator to avoid contamination (presence of plant material, Aloe vera and alginate, easily contaminating in water).
The ExpertGel® 230 or EG230 polymer is a semi-crystalline thermo-gelling polymer. It is film-forming, non-irritating, with a pH between 6 and 8 and is therefore compatible with the pH of the skin. Sodium alginate is a thickening and gelling agent which makes it possible to increase the viscosity of the composition.
La glycérine permet aussi de diminuer la température de gélation de l’EG230.Glycerine has the property of being moisturizing, emollient and also moisturizing. It therefore improves the lubricity and lubrication of the composition. Glycerin also reduces the freezing temperature of EG230.
Sodium chloride (NaCl) acts as a binder, thickener and masking agent. Like glycerine, sodium chloride lowers the freezing temperature of EG230. Aloe vera has many properties. It is both film-forming, antibacterial, antifungal, emollient, humectant, softening and anti-irritant and moisturizing.
The amount of each constituent is advantageously chosen so that the composition is soluble in its container, can thus be expelled in aerosol form and quickly gel on the skin. The active components are present in sufficient quantity with regard to the intended application.
An in vitro study is conducted with the creation of a lesion of a fibroblast cell mat (dermal cells). The composition prepared above is applied to the lesion and monitoring over time is achieved. The same composition is also applied to the lesion, but without alginate, and monitoring over time is performed. 48 hours after the lesion, it is found that the cicatrization is total with the composition according to the invention, while the healing is only partial with a composition without alginate.
