Abstract

Novel reagent for counting basophils and opt. other leukocytes in a biological medium (esp. a blood sample) consists of a hypotonic aq. soln. contg. a metachromatic reagent (pref. toluidine blue), a fixative for leukocytes and basophils, and an acid capable of contributing to the disruption of erythrocytes in blood samples. The acid is present in such an amt. that the final pH of the reagent is 3-5. The process comprises directly mixing the sample, pref. in a single step, with the above reagent. Visualisation and counting of basophils is used in a variety of diagnostic procedures, pref. for the diagnosis of anaphylactic sensitivities, esp. allergies, involving incubating several fractions of a blood sample with 1 agent (esp. an allergen) having a degranulating effect on basophils.
