Description
- Die vorliegende Erfindung bezieht sich auf eine Zusammensetzung für den Einsatz als anti-seborrhoische Formulierung für die Behandlung der klinischen Krankheit seborrhoische Dermatitis bei Tieren wie etwa Hunden.
- Seborrhoe ist eine chronische Hautkrankheit, die als Defekt der Keratinisierung angesehen wird mit einer verstärkten Schuppenbildung. Dandruff ist eine milde Form. Seborrhoe wird in drei klinische Formen aufgeteilt. Seborrhoe Sicca ist charakterisiert durch trockene Schuppenbildung der Haut. Seborrhoe Oleosa ist charakterisiert durch lokale diffuse Schuppenbildung assoziiert mit exzessivem Talg. Seborrhoische Dermatitis ist charakterisiert durch Schuppenbildung und Fettigkeit mit starkem Auftreten lokaler oder diffuser Entzündung. Es gibt Unterschiede hinsichtlich der Rassen für Cocker-Spaniel, Springer-Spaniel, Basset-Hunde und insbesondere tritt die schwierigste Form bei Westhigland-White- Terriern auf. Sie wird angesehen als ein primärer Keratiniserungsdefekt mit genetischem Ursprung. Obwohl einige Gründe bekannt sind, zeigen sich diese als designierte Sekundärseborrhoen. Primäre idiopathische seborrhoische Dermatitis wird gegenwärtig angesehen als chronische Krankheit, die gemildert, jedoch nicht geheilt werden kann.
- Standardheilungsbehandlungen sind normalerweise Shampoos mit einem Gehalt Salicylsäure, Schwefel, Selensulfid, Teer und Antiseptika. Diese ge ben jedoch nur eine zeitlich sehr begrenzte Besserung der Symptome, normalerweise höchstens einige Tage.
- Menschliche seborrhoische Dermatitis wurde kürzlich assoziiert mit der Hefe Malassezia (Pityrosporum), ovale. Malassezia pachydermatis wurde im Zusammenhang mit einer Dermatitis bei Hunden beschrieben. Hunde mit der klassischen seborrhoischen Dermatitis bei Westhigland-Whites sind jedoch mit Ketaconazoletabletten oral behandelt worden, ohne daß sich eine verläßliche Heilung ergab. Die Symptome traten wieder auf oder verschlechterten sich während einer Behandlung.
- Es ist ein Gegenstand der vorliegenden Erfindung, eine antiseborrhoische Formulierung zur Verfügung zu stellen, die wirksam ist gegen seborrhoische Dermatitis und einen therapeutischen Erfolg erzielt, der von Standardbehandlungen bislang nicht bekannt war.
- Die Erfindung löst diese Aufgabe durch die Bereitstellung einer Zusammensetzung für die Behandlung der seborrhoischen Hautkrankheit mit einem Breitbandspektrum-Arzneistoff gegen Pilze und/oder einem topischen Antiseptkum.
- Vorzugsweise wird die Zusammensetzung formuliert für topische Anwendung und kann eine keratolytische/keratoplastische Verbindung enthalten.
- Vorzugsweise ist der Breitbandspektrumarzneistoff gegen Pilze aus der Imidazol/Triazol-Gruppe z. B. Miconazol.
- Vorzugsweise ist das topische Antiseptikum von Phenolbiguanid- Typ, wie etwa Chlorhexidin oder Tricolsan.
- Vorzugsweise ist die keratolytische/keratoplastische Verbindung Selensulfid.
- Vorzugsweise wird die Zusammensetzung zur Verfügung gestellt auf einer Shampoo-Basis, bei welcher es sich um jede Standardshampoo-Basis handeln kann, insbesondere kommt eine typische Antischuppenshampoo-Basis zum Einsatz.
- Die Erfindung soll nun beschrieben werden unter Bezugnahme auf die Darstellung in Fig. 1, welche die Ergebnisse eines Versuchs einer besonderen Zusammensetzung gemäß der Erfindung erläutert.
- Beim Testen einer Zusammensetzung gemäß der Erfindung wurden 41 Hunde mit seborrhoischer Dermatititis diagnostiziert. Diese 41 Hunde umfaßten eine unproportional hohe Zahl von Westhighfand-White-Hunden, verglichen mit der normalen Hundepopulation. Dies ist zu erwarten auf der gegebenen Rassenanfälligkeit gegenüber dieser Krankheit. Ein doppelter klinischer Zufallsversuch wurde mit zwei Shampoobehandlungen ausgelegt. Ein Austausch von einer Behandlungsgruppe zur anderen wurde zugelassen, wenn die erste Behandlung versagte.
- Die Hunde wurden behandelt entweder mit einem industriellen Standard-Selensulfid-Shampoo (Selsun Blue oder Seleen) hergestellt von Abbots Laboratories oder der folgenden erfindungsgemäßen Zusammensetzung: ein Shampoo enthaltend 0,25% Selensulfid, 2% Chlorhexidin und 2% Mineonazol. Alle Bestandteile wurden gewogen. Die Wirkung wurde dann gemessen anhand eines objektiven Maßstabes durch Inaugenscheinnahme einer Verbesserung der klinischen Symptome nach 2 und 4 Wochen.
- Die Fig. 1 zeigt grafisch die Ergebnisse des Versuches, welche anzeigen, daß die Shampoozusammensetzung gemäß der Erfindung, d. h. die mit I bezeichnete Darstellung zu Ergebnissen führte, die weit besser waren als die Standardbehandlung mit einem 1%igen Selensulfidshampoo angezeigt durch die Grafik II.
- Imidazol- und Triazolarzneien sind bekannte wirksame Fungizidchemikalien, die in der Medizin und der Landwirtschaft zum Einsatz kommen. Beispiele sind Ketoconazol, Miconazol, Econazol und Elinconazol.
- Eines der wirksamsten systemischen Imidazole ist Ketoconazol. Allein als Tablette hat es versagt, verläßliche Heilung bei Versuchen an seborrhoischen Westhighland-White-Hunden zu zeigen. Teilwirkungen bei einigen Hunden folgte ein Wiederaufflammen der Seborrhoe noch während der Behandlung.
- Chlorhexidin ist ein phenolverwandtes Biguanid-Antiseptikum, wobei es sich um ein antimikrobielles Breitbandarzneimittel handelt. Es wurde eingesetzt als topisches Wasch-/Spülmittel in einem Shampoo für Veterinär- und Humanmedizin seit mehr als 30 Jahren. Obwohl es in großem Rahmen zum Einsatz gekommen ist, hat es sich nicht als wertvoll gezeigt bei seborrhoischer Dermatititis.
- Selensulfid in einer Shampoo-Basis kommt in großem Rahmen zum Einsatz als antiseborrhoisches und Antischuppenmittel für Menschen und Hunde. Es gibt jedoch lediglich zeitlich begrenzte Erleichterung. Es besitzt keratoplastische und kerotolytische Eigenschaften. Es wirkt somit durch Unterdrücken und Aufbrechen von Schuppen.
- Verschiedene Kombinationen der vorgenannten aktiven Bestandteile wurden untersucht in einer Shampoo-Basis an Hunden mit seborrhoischer Dermatitis. Es hat sich gezeigt, daß die Kombinationen von 1,0% Selensulfid und 1,0% Chlorhexidin, 1,0% Selensulfid, 1% Chlorhexidin und 1,0% Miconazol sowie 0,25 % Selensulfid mit 2% Chlorhexidin und 2,0% Miconazol alle viel wirksamer waren als das Standard 1,0% Selensulfid allein. Ein 2% Shampoo mit Imidazolen, wie Ketaconazol, Miconazol, Econazol und Elinconazol erzeugte ebenfalls brauchbare Ergebnisse. Die besten Ergebnisse wurden jedoch erzielt mit der Zusammensetzung, wie sie oben unter Bezugnahme auf Fig. 1 beschrieben wurde.
Claims (14)
1. Antiseborrhoische Zusammensetzung für die Behandlung von Hunden, die:
(a) einen Breitspektrum-Arzneistoff gegen Pilze aus der Gruppe der
Imidazole und Triazole sowie
(b) ein topisches Antiseptikum aus der Gruppe Chlorhexidin und Triclosan
enthält.
2. Antiseborrhoische Zusammensetzung nach Anspruch 1, wobei es sich bei
dem Breitspektrum-Arzneistoff gegen Pilze um Ketaconazol, Miconazol,
Econazol oder Elinconazol handelt.
3. Antiseborrhoische Zusammensetzung nach Anspruch 2, wobei es sich bei
dem topischen Antiseptikum um Chlorhexidin und bei dem Breitspektrum-
Arzneistoff gegen Pilze um Miconazol handelt.
4. Antiseborrhoische Zusammensetzung nach Anspruch 3 mit 2,0 Gew.-%
Miconazol und 2,0 Gew.-% Chlorhexidin.
5. Antiseborrhoische Zusammensetzung nach einem der Ansprüche 1 bis 4,
die als Shampoo formuliert ist.
6. Antiseborrhoische Zusammenstzung nach Anspruch 5, die eine
keratolytische oder keratoplastische Verbindung enthält.
7. Antiseborrhoische Zusammenstzung nach Anspruch 6, dadurch
gekennzeichnet, daß es sich bei der keratolytischen oder keratoplastischen
Verbindung um Selensulfid handelt.
8. Antiseborrhoische Zusammensetzung nach Anspruch 7 mit 0,25 Gew.-%
Selensulfid, 2,0 Gew.-% Chlorhexidin und 2,0 Gew.-% Miconazol.
9. Antiseborrhoische Zusammensetzung nach Anspruch 7 mit 1 Gew.-%
Selensulfid, 1 Gew.-% Chlorhexidin und 1 Gew.-% Miconazol.
10. Verwendung einer Verbindung mit einem Arzneistoff gegen Pilze und einem
topischen Antiseptikum zur Herstellung eines Arzneimittels für die
Behandlung von seborrhoischen Hautkrankheiten beim Hund.
11. Verwendung nach Anspruch 10, wobei es sich bei dem topischen
Antiseptikum um einen Phenol-Biguanid-Typ und bei dem Arzneistoff gegen Pilze

Antiseptikum um Chlorhexidin und bei dem Arzneistoff gegen Pilze um Miconazol
handelt.
13. Verwendung nach Anspruch 10, wobei es sich bei der Zusammensetzung
um eine Zusammensetzung nach einem der Ansprüche 1 bis 9 handelt.
14. Verwendung einer Zusammensetzung mit 1% Selensulfid und 1%
Chlorhexidin in einer Shampoo-Grundlage bei der Herstellung eines Arzneimittels
für die Behandlung von seborrhoischen Hautkrankheiten beim Hund.
