Description
Eine alternative Lösung des Kreuzkontaminationsproblems besteht in der Verwendung einer porösen Membran, die die Aminosäurereaktionsmischung am Ort der Applikation
verwendet.
Das vollautomatische Waschen, Absättigen und Entschützen ist die bevorzugte Ausfuhrungsform bei der Herstellung der Substanzbibliothek (Figur 2).
Zur Montage wird die Kapillarplatte [4] auf das Unterteil eines Synthesetisches [6] gelegt, der fest mit einem Pipettierroboter verbunden ist. Dabei kommen die Randbereiche [5] der Kapillarplatte auf einer weichen, lösungsmittelresistenten Dichtung [7] zu liegen. Anschließend werden Abstandhalter [8] rings um die

Zur Inkubation wird bei verschlossenem Ablauf [26] und Einfüllstutzen [24] der Kolben [23] langsam nach oben bewegt, so daß die Luft aus den Kapillaren [1] in die Kavität [25] entweichen kann. Noch in Kapillaren [1] eingeschlossene Luft kann durch Beschallung mit dem Ultraschallgeber [22] entfernt werden. Wenn der Kolben [23] bis an die Kapillarplatte [4] herangeschoben wird, füllt sich die Kavität [25] vollständig mit Wirkstofflösung oder -suspension und sowohl die Luft als auch überzählige Wirkstofflösung bzw. -suspension weichen durch seitliche Kanäle [27] in die Ausgleichsgefäße [28] aus. Durch wiederholtes Heben und Senken des Kolbens [23] mit kurzem Hub wird dann die Lösung oder Suspension in den Kapillaren [1] mit der in der Kavität [25] befindlichen Lösung oder Suspension gemischt und noch in der Kavität [25] befindliche Luftblasen schrittweise in die Ausgleichsgefäße [28] vertrieben.
Das Waschen der Vorrichtung erfolgt in analoger Weise unter Verwendung geeigneter Waschlösungen.
