说明书
本发明涉及烷基-N-(3-二甲氨基)烷基氨基甲酸酯的制备方法。
烷基-N-(3-二甲氨基)烷基氨基甲酸酯及其盐可以用作杀真菌剂(参见GBl212708，ZA685172，DE1567169，DE1643040)。
正丙基-(3-二甲氨基)正丙基氨基甲酸酯是一种众所周知的杀真菌活性成份，通常以盐酸盐的销售来销售。该化合物通常是通过使3-(二甲氨基)正丙基氨与氯甲酸正丙酯在包括甲苯和醚在内的惰性溶剂中进行反应来制备的。
我们现在发现了一种新的方法，其特点是在变化的温度条件下使反应在醇中或醇的混合物与水、惰性有机溶剂或其混合物的混合物中进行。
因此，本发明提供了一种方法，用于制备烷基-N-(3-二甲氨基)烷基氨基甲酸酯，该方法包括使氯甲酸烷基酯在C1-C6脂族醇中进行反应。
较佳的，本发明的方法被应用于通过使氯甲酸正丙酯在C1-C6脂族醇中进行反应来制备正丙基-(3-二甲氨基)正丙基氨基甲酸酯。
利用本发明的方法，反应可以在大的温度范围内进行、达到很高的产率。
该反应能够以如此高的产率进行这一事实是出乎意料的，因为根据预期，醇会与氯甲酸正丙酯进行反应，特别是在如3-(二甲氨基)正丙基胺或者正丙基-(3-二甲氨基)正丙基氨基甲酸酯自身之类的酸受体存在的情况下是如此(例如。参见Houben-Weyl，第四版，1983年， E4，第68页)。
然而，以氯甲酸正丙酯计，采用本方法，可以得到95％的得率，而基于胺的得率甚至更高(高达97％)。
正如人们所推测，本发明提供了与在惰性溶剂或水溶剂中(参见DE1643040，DE1567169，GB1212708，ZA685172)或其它普通氨基甲酸酯工艺(例如，Houben-Weyl，第四版，1983年， 8，第138ff页，及 E4，第149ff页)相比高得率的正丙基-(3-二甲氨基)正丙基氨基甲酸酯。
本发明方法的另一优点在于不再需要额外的酸受体。
再一优点是反应可以在宽范围的温度内进行，从-20℃直到醇的沸点温度甚至更高，通常高达200℃。
优选温度范围为-20℃到110℃左右，特别优选的温度是-20℃到97℃左右。这些温度，进而反应的速度，总的来说明显地比采用惰性溶剂、水或其混合物的其它方法要高(例如，Houben-Weyl，第四版， 8，第138ff页，及E4，第149ff页)。
通常，当使用较高温度时选择性会降低，在甲苯例中，如果温度高于65℃，氯甲酸正丙酯的降解显著增加，产生大量的二氧化碳和正丙基氯。在其它文献例中(参见Houben-Weyl，第四版， 11/1，第985f页，及 E4，第153f页)，甚至在较低温度下这一降解也是主要的反应。
不同寻常的是，我们的方法中以两种混合物计的得率即使在高于85℃的温度下也能保持稳定，氯甲酸正丙酯的降解一般也非常有限。
根据本发明所采用的醇是C1-C6脂族醇，优选C1-C4的脂族醇。进一步优选的醇是丙醇，特别是正丙醇，而且采取循环的形式。而且优选采用混合溶剂，该混合溶剂由一种或多种这样的醇与水或惰性有机溶剂或水与惰性有机溶剂的混合物组成，最高达30％重量。惰性有机溶剂的例子包括甲苯、醚等，例如THF或甲基叔丁基醚。
对于本发明的方法，试剂既可以用化学计量的，也可以是最高50％摩尔数过量的，优选过量10％，最好是氯甲酸正丙酯最多过量10％摩尔数。
3-(二甲氨基)正丙基胺与氯甲酸正丙酯的摩尔比例一般在1∶0.75-1.5，优选1∶0.95-1.1。
因为其它的几个特征，本发明的方法在经济方面和环保方面都是有利的。
像正丙醇之类的醇与醚、二氯甲烷、或其它惰性溶剂相比，是在环保和技术方面都不会有问题的溶剂。再蒸馏的醇可以使用。
反应可以在高浓度进行。
在高反应温度的放出的热量很容易控制。
反应过程中几乎没有气体发生，不产生废水。
本发明由以下实施例来说明。
实施例
3-(二甲氨基)正丙基胺(184.4g)于20-40℃滴加到循环正丙醇(443.5g)中。在30-40分钟内滴加氯甲酸正丙酯(227.3g)。温度迅速升高，烧瓶从容地进行冷却使温度维持在80-85℃。在蒸馏除去正丙醇(以后循环使用)后，留下的即正丙基3-(二甲氨基)正丙基氨基甲酸酯盐酸盐，以3-(二甲氨基)正丙基胺计的得率为97％。这比采用甲苯的反应的得率高了4％。
以氯甲酸正丙酯计的得率为95％，比用甲苯的反应的得率高了至少16％。
甲苯工艺例：
氯甲酸正丙酯(180g)于20-40℃滴加到甲苯(650g)中。在30-40分钟内滴加3-(二甲氨基)正丙基胺(125g)。温度迅速升高，烧瓶进行冷却使温度维持在55-60℃。反应混合物冷却到40-45℃，加水。分相后，粗正丙基3-(二甲氨基)正丙基氨基甲酸酯溶液进行蒸馏得到正丙基3-(二甲氨基)正丙基氨基甲酸酯盐酸盐，以3-(二甲氨基)正丙基胺计的得率为93％。以氯甲酸正丙酯计的得率为79％。

Claims (8)

1.一种3-(二甲氨基)丙基氨基甲酸正丙酯盐酸盐的制备方法，其特征在于，包括使氯甲酸正丙酯与3-(二甲氨基)正丙基胺在C1-C6脂族醇中或者一种或多种这类脂族醇与最多30％重量的水或惰性有机溶剂或其混合物的混合物中进行反应。
2.如权利要求1所述的方法，其特征在于，所述的C1-C6脂族醇是C1-C4脂族醇
3.如权利要求1所述的方法，其特征在于，所述的C1-C6脂族醇是丙醇。
4.如权利要求1所述的方法，其特征在于，所述的C1-C6脂族醇是正丙醇或循环正丙醇。
5.如权利要求1所述的方法，其特征在于，反应进行的温度范围在-20℃到110℃。
6.如权利要求1到5中任一项所述的方法，其特征在于，两种试剂的用量是化学计量的。
7.如权利要求1到5中任一项所述的方法，其特征在于，两种试剂中的一种的用量最多过量50％摩尔数。
8.如权利要求1到5中任一项所述的方法，其特征在于，氯甲酸正丙酯最多过量10％摩尔数。
