Disclosure of Invention
In order to have a good anti-drop effect for androgenetic alopecia patients, the application provides a composite herbal plant composition for removing dandruff, preventing alopecia and relieving itching, a fermentation process and application thereof.
In a first aspect, the application provides a composite herbal plant composition for removing dandruff, preventing alopecia and relieving itching, which adopts the following technical scheme:

Step 3), cooling the pre-extract to room temperature, inoculating a compound bacterium compounded by schizosaccharomyces pombe, powdered pichia pastoris and succinic acid-producing filamentous bacillus, and fermenting for 96-120 hours to obtain a fermentation extract;
step 4), adding ethanol into the fermented extract, heating to 68-72 ℃, extracting for 90-120min, cooling, filtering to obtain filtrate, removing impurities, and obtaining the anti-dandruff, anti-drop and antipruritic composite herbal plant composition;
In the step 1), the mass ratio of the kuh-seng to the biota orientalis, the angelica, the gentiana macrophylla, the cnidium fruit and the viola philippica is 4.5-5.5:0.5-1.5:1.5-2.5:1.5-2.5:1.5-2.5:0.5-1.5;
In the step 3), the mass ratio of the schizosaccharomyces pombe, the pichia pastoris powder and the succinic acid-producing filamentous bacillus is 0.0002-0.0004:0.0005-0.0007:0.0004-0.0006.
By adopting the technical scheme, the active ingredients in the kuh-seng, the biota orientalis, the angelica sinensis, the gentiana macrophylla, the cnidium fruit and the viola yedoensis are compounded and extracted, so that the kuh-seng, the biota orientalis, the angelica sinensis, the gentiana macrophylla, the cnidium fruit and the viola yedoensis can be well matched with each other, the composite bacteria compounded by the schizosaccharomyces pombe, the pichia pastoris and the succinic acid-producing filamentous bacillus are adopted for fermentation, the cell walls of plants can be well destroyed, the active ingredients in the composite plants can be better extracted, the active ingredients in the composite plants have higher content, the active ingredients after the active ingredients are fermented by the composite bacteria form a small molecular structure which is easier to absorb, the active ingredients after fermentation can have a remarkable effect of inhibiting the generation of the Dihydrotestosterone (DHT), the composite is more gentle to androgen alopecia patients, the negative effects of the composite on the anti-dandruff anti-itching effect can be reduced, and the composite is better in anti-itching effect after the composite is added into the anti-itching articles.
Preferably, the mass ratio of the composite plant particles to the composite bacteria is 10-16:0.0011-0.0017.
By adopting the technical scheme, the fermentation effect of the composite bacteria is better, the waste of the composite bacteria is reduced, and the composite bacteria has higher economic value.
Preferably, the mass ratio of the composite plant particles to the water is 4-5:2-3.
By adopting the technical scheme, the dissolution performance of the solvent is better, the extraction effect of the active ingredients is better, and the anti-dandruff, anti-falling and antipruritic effects of the prepared composition are better by specifically selecting the ratio of water to ethanol.
Preferably, in the step 4), after adding ethanol, continuously introducing carbon dioxide until the heating process is finished, and introducing 100-200mL/h of carbon dioxide into each 1kg of water.
By adopting the technical scheme, the pH value in the water is more suitable for the growth of the compound bacteria by introducing the carbon dioxide, the activity of the compound bacteria is effectively improved, the fermentation efficiency is better, and the anti-dandruff, anti-drop and antipruritic effects of the composition are better.
Preferably, in the step 4), during the impurity removal, D113 resin and RS407 resin are added into the filtrate, stirred for 30-40min, and filtered, thus the impurity removal is completed.
By adopting the technical scheme, the D113 resin and the RS407 resin are added into the filtrate to selectively remove impurities, so that the purity of effective active ingredients in the composition is higher, and the composition has better effects of removing dandruff, preventing alopecia and relieving itching.
Preferably, the mass ratio of the filtrate to the D113 resin to the RS407 resin is 10:1.3-1.7:1.3-1.7.
By adopting the technical scheme, the impurity removal effect is better, the active ingredients in the composition are purer, and the effects of removing dandruff, preventing alopecia and relieving itching are better by specifically selecting the mass ratio of the filtrate, the D113 resin and the RS407 resin.
In a second aspect, the application provides a preparation method of a composite herbal plant composition for removing dandruff, preventing alopecia and relieving itching, which adopts the following technical scheme:
A composite herbal composition for removing dandruff, preventing alopecia and relieving itching is prepared by fermenting the composite herbal composition.
By adopting the technical scheme, the composite herbal plant composition for removing dandruff, preventing alopecia and relieving itching has the effect of remarkably inhibiting the generation of DHT, has a good anti-alopecia effect on androgenetic alopecia patients, and has good anti-dandruff, anti-alopecia and antipruritic effects.
In a third aspect, the application provides an application of a composite herbal plant composition for removing dandruff, preventing alopecia and relieving itching, which adopts the following technical scheme:
The application of the anti-dandruff, anti-falling and antipruritic composite herbal plant composition is that the anti-dandruff, anti-falling and antipruritic composite herbal plant composition is added into a washing and caring product to prepare the washing and caring product with the anti-dandruff, anti-falling and antipruritic effects, and the mass percentage of the anti-dandruff, anti-falling and antipruritic composite herbal plant composition in the washing and caring product is 10-15%.

In summary, the application has the following beneficial effects:

According to the application, the active ingredients in the kuh-seng, the biota orientalis leaves, the angelica sinensis, the gentiana macrophylla, the cnidium fruit and the viola yedoensis are compounded and extracted, so that the kuh-seng, the biota orientalis leaves, the angelica sinensis, the gentiana macrophylla, the cnidium fruit and the viola yedoensis can be well matched with each other, the cell wall of a plant can be well destroyed by adopting the compound bacteria compounded by schizosaccharomyces pombe, the active ingredients in the compound plant can be better extracted, the active ingredients in the composition have higher content, the active ingredients form a micromolecular structure which is easier to absorb after the fermentation of the compound bacteria, the active ingredients after fermentation can have a remarkable effect of inhibiting the generation of Dihydrotestosterone (DHT), the anti-drop effect on androgenic alopecia patients is better, the irritation components can be effectively reduced, the composition is milder, the negative influence of the irritation substances on the anti-drop effect of the composition on the anti-drop effect is reduced, and the anti-drop effect of the composition can be well achieved after the composition is added into the anti-drop for the hair care product.
2. In the application, the proportion of water and ethanol is preferably selected in particular, so that the solvent has better dissolution performance, the active ingredient has better extraction effect, and the prepared composition has better effects of removing dandruff, preventing alopecia and relieving itching.
3. In the present application, it is preferable that the D113 resin and the RS407 resin are added to the filtrate to selectively remove impurities, so that the purity of the active ingredient in the composition is higher, and the composition has a better anti-dandruff, anti-slip and antipruritic effect.
Detailed Description
The present application will be described in further detail with reference to examples.
Example 1
A fermentation process of a composite herbal plant composition for removing dandruff, preventing alopecia and relieving itching comprises the following steps:
Step 1), 4.5kg of kuh-seng, 0.5kg of biota orientalis, 1.5kg of angelica, 1.5kg of gentiana macrophylla, 1.5kg of fructus cnidii and 0.5kg of viola philippica are broken into particles, and the particles are sieved by a 10-mesh sieve to obtain the composite plant particles.
And 2) putting 10kg of the composite plant particles and 40kg of water into a reaction kettle, heating to boiling, condensing and refluxing, and extracting for 2 hours to obtain the pre-extract.
And 3) cooling the pre-extract to room temperature, inoculating a compound bacterium compounded by 0.0002kg of schizosaccharomyces pombe, 0.0005kg of powdered Pichia pastoris and 0.0004kg of filamentous bacillus succinogenes, and fermenting at a constant temperature of 32 ℃ for 96 hours to obtain a fermentation extract.
And 4) adding 20kg of ethanol into the fermented extract, introducing carbon dioxide, heating to 68 ℃ with the carbon dioxide introducing amount of 4000mL/h, extracting for 90min, cooling to room temperature, filtering by a filter cloth, discarding filter residues to obtain filtrate, carrying out rotary evaporation concentration on the filtrate at the rotary evaporation temperature of 60 ℃, carrying out rotary evaporation pressure of 0.01Mpa, concentrating the filtrate with the mass of 10kg, then adding 1.3kg of D113 resin and 1.3kg of RS407 resin into the filtrate, stirring for 30-40min, filtering by the filter cloth, discarding the filter residues, and taking the filtrate to obtain the anti-dandruff, anti-falling and antipruritic composite herbal plant composition.
The kuh-seng, the biota orientalis, the angelica, the gentiana macrophylla, the cnidium fruit and the viola philippica are all commercially available and are sun-cured traditional Chinese medicinal materials.
The water is deionized water, and the water is self-made.
Ethanol is purchased from Jinan Yuyi chemical Co., ltd., absolute ethanol, CAS number 64-17-5.
Schizosaccharomyces pombe was purchased from Beijing Bai-Ou Bo Wei Biotechnology Co., ltd., platform No. Bio-58027.

Example 2
A fermentation process of a composite herbal plant composition for removing dandruff, preventing alopecia and relieving itching comprises the following steps:
Step 1), 5kg of radix sophorae flavescentis, 1kg of biota orientalis, 2kg of Chinese angelica, 2kg of gentiana macrophylla, 2kg of fructus cnidii and 1kg of herba violae are broken into particles, and the particles are sieved by a 14-mesh sieve, so that the composite plant particles are obtained.
And 2) putting 13kg of composite plant particles and 45kg of water into a reaction kettle, heating to boiling, condensing and refluxing, and extracting for 2.5h to obtain a pre-extract.
And 3) cooling the pre-extract to room temperature, inoculating a compound bacterium compounded by 0.0003kg of schizosaccharomyces pombe, 0.0006kg of powdery pichia pastoris and 0.0005kg of filamentous bacillus succinogenes, and fermenting at the constant temperature of 32.5 ℃ for 108 hours to obtain a fermentation extract.
And 4) adding 25kg of ethanol into the fermented extract, introducing carbon dioxide, heating to 70 ℃ with the carbon dioxide introducing amount of 7000mL/h, extracting for 105min, cooling to room temperature, filtering by a filter cloth, discarding filter residues to obtain filtrate, carrying out rotary evaporation concentration on the filtrate at the rotary evaporation temperature of 60 ℃, carrying out rotary evaporation pressure of 0.01Mpa, concentrating the filtrate with the mass of 13kg, then adding 1.5kg of D113 resin and 1.5kg of RS407 resin into the filtrate, stirring for 30-40min, filtering by the filter cloth, discarding the filter residues, and taking the filtrate to obtain the anti-dandruff, anti-falling and antipruritic composite herbal plant composition.
The kuh-seng, the biota orientalis, the angelica, the gentiana macrophylla, the cnidium fruit and the viola philippica are all commercially available and are sun-cured traditional Chinese medicinal materials.

Example 3
A fermentation process of a composite herbal plant composition for removing dandruff, preventing alopecia and relieving itching comprises the following steps:
Step 1), 5.5kg of kuh-seng, 1.5kg of biota orientalis, 2.5kg of angelica, 2.5kg of gentiana macrophylla, 2.5kg of fructus cnidii and 1.5kg of viola philippica are broken into particles, and the particles are sieved by a 20-mesh sieve to obtain the composite plant particles.
And 2) putting 16kg of composite plant particles and 50kg of water into a reaction kettle, heating to boiling, condensing and refluxing, and extracting for 3 hours to obtain the pre-extract.
And 3) cooling the pre-extract to room temperature, inoculating a compound bacterium compounded by 0.0004kg of schizosaccharomyces pombe, 0.0007kg of powdery pichia pastoris and 0.0006kg of filamentous bacillus succinogenes, and fermenting for 120 hours at a constant temperature of 33 ℃ to obtain a fermentation extract.
And 4) adding 30kg of ethanol into the fermented extract, introducing carbon dioxide, heating to 72 ℃ with the carbon dioxide introducing amount of 10000mL/h, extracting for 120min, cooling to room temperature, filtering by a filter cloth, discarding filter residues to obtain filtrate, carrying out rotary evaporation concentration on the filtrate at the rotary evaporation temperature of 60 ℃, carrying out rotary evaporation pressure of 0.01Mpa, concentrating the filtrate with the mass of 16kg, then adding 1.7kg of D113 resin and 1.7kg of RS407 resin into the filtrate, stirring for 30-40min, filtering by the filter cloth, discarding the filter residues, and taking the filtrate to obtain the anti-dandruff, anti-falling and antipruritic composite herbal plant composition.

Experiment 1
Anti-dandruff and antipruritic experiment:
The method for preparing the shampoo to be tested comprises the following steps:
1.5kg of cocamide MEA, 5kg of cocamidopropyl betaine, 6kg of glycerol, 12kg of laureth sulfate sodium, 0.5kg of guar gum hydroxypropyl trimethyl ammonium chloride, 1kg of sunflower glucoside, 0.5kg of polydimethylsiloxane, 13kg of anti-dandruff, anti-drop and antipruritic composite herbal plant composition, 0.2kg of sclerotium rolfsteak, 0.1kg of EDTA disodium and 60.2kg of deionized water are weighed into a stirring kettle, and the mixture is stirred for 15min and uniformly mixed to obtain shampoo to be tested.
The anti-dandruff, anti-drop and antipruritic composite herbal composition is the anti-dandruff, anti-drop and antipruritic composite herbal composition of each example and comparative example.
The volunteers were summoned in groups using the shampoo to be tested to which the anti-dandruff, anti-hair loss and antipruritic composite herbal composition of each example or comparative example was added, and each 10 human volunteers were shampooed 1 time per 4 days with the shampoo to be tested, and the number of dandruff and whether scalp itching was recorded before each shampoo was tested 5 times in total.
The method for testing the dandruff comprises separating hair along the vertex center line to form a hair boundary, attaching the dandruff test card to the hair boundary for 5s, removing the dandruff test card, recording the amount of dandruff adhered on the test card, and taking the average value of all experimental data.
The experimental data of experiment 1 are shown in tables 1 and 2.

According to the comparison of the data in tables 1-2, the effect of well inhibiting dandruff is achieved after three times of shampoo, the shampoo is not washed for four days, the quantity of dandruff is small, the anti-dandruff effect of each example is obviously higher than that of each comparative example, and it is seen that the anti-dandruff antipruritic effect of the obtained anti-dandruff antipruritic composite herbal composition is better after the composite plant particles compounded by the schizosaccharomyces pombe, the pichia pastoris powder and the succinic acid producing filamentous bacillus are fermented by the composite bacteria compounded by the kuh-seng, the biota orientalis, the angelica, the gentiana macrophylla, the fructus cnidii and the viola yedoensis.
Comparative examples 1-4 the properties of the fermentation products were changed by destroying the complex systems of schizosaccharomyces pombe, pichia pastoris powder, and filamentous succinic acid-producing bacillus, resulting in a significant decrease in the anti-dandruff and antipruritic efficacy of the resulting compositions.
Comparative examples 5 to 7 showed a decrease in the effect of removing impurities specifically due to the destruction of the complex system of the D113 resin and the RS407 resin, and the obtained complex herbal composition showed a decrease in the effect of removing dandruff and relieving itching.
Experiment 2
5 Alpha reductase inhibition assay:
1. Principle of experiment
Androgenic alopecia (AGA) is caused by the fact that slaughter ketone (T) is converted into more active Dihydrotestosterone (DHT) by 5 alpha-reductase, which is sensitive to hair follicles and affects hair follicle development, resulting in alopecia. Whether the test substance can inhibit 5α -reductase, thereby inhibiting DHT production, can be used as one of the basis for determining whether the hair-growing efficacy is achieved.
2. Experimental materials
Type 2.1 type II 5 alpha-reductase extracted from rat prostate tissue, stored at-80 ℃.
2.2 Positive Control (PC) 1. Mu.M finasteride solution.
2.3 Sample preparation (TA) the composite herbal composition of each example and comparative example was diluted to 5% (v/v) with PBS buffer solution.
2.4 Blank control Phosphate Buffer (PBS)
3. Experimental procedure
3.1 Preparation of type II 5 alpha-reductase: male rats, 10, were taken out, cervical vertebrae were removed and sacrificed after no water retention after overnight fast, and the prostate was taken out and minced on an ice table. Homogenizing at 0deg.C with precooled buffer solution in a glass homogenizer at a ratio of 1:5, centrifuging at 3000g for 10min, collecting supernatant 10000g, centrifuging for 30min, collecting supernatant, packaging into EP tubes with 1mL of enzyme extract, and storing in a refrigerator at-80deg.C.
3.2 Type II 5 alpha-reductase Activity assay: the 2mL reaction system contains 0.25mL of phosphate buffer, 0.70mL of enzyme extract, 0.1mL of testosterone solution (0.5 mg/mL), 0.35mL of NADPH solution (1 mg/mL), 0.6mL of finasteride in the positive control group and 0.6mL of sample solution in the complete reaction group. The control group replaces enzyme extract with phosphate buffer solution, and adds 2.0mL of dichloromethane to stop the reaction, the complete reaction group, the positive group and the sample group react for 30min at 37 ℃, after the reaction is finished, 2.0mL of dichloromethane is added to stop the reaction, after shaking for 1min, 5000r/min is centrifugated for 10min. The upper aqueous phase was removed, about 1.0mL of the organic phase was removed and evaporated to dryness, and the residue was dissolved in 1.0mL of methanol and the testosterone peak area (T) was determined by high performance liquid chromatography. Each reaction was set up with 3 parallel tubes.
In the blank control group, testosterone cannot be converted into dihydrotestosterone without the action of type II 5 alpha-reductase, and the content of testosterone per se is finally measured.
Complete response group testosterone (T) is converted to dihydrotestosterone by 5 a-reductase and the final measurement is of the residual testosterone content after the response.
Positive substance/sample group by inhibiting activity of 5 alpha-reductase type II, inhibiting conversion of testosterone (T) to dihydrotestosterone, measuring content of testosterone remaining after reaction, and determining whether activity of 5 alpha-reductase type II is effectively inhibited by calculating inhibition rate and statistical analysis.
3.3 Analysis of results:

According to the comparison of the data in Table 3, the inhibition rate of each example is close to that of the positive group and is obviously higher than that of each comparative example, and it is seen that each example ferments the composite plant compounded by the schizosaccharomyces pombe, the pichia pastoris powder and the succinic acid-producing filamentous bacillus through the composite bacteria compounded by the schizosaccharomyces pombe, the biota orientalis, the angelica sinensis, the gentiana macrophylla, the cnidium fruit and the viola yedoensis, and the prepared composition has good effect of inhibiting DHT generation and has good anti-drop effect for androgenic alopecia patients.
In each comparative example, although the compound plants formed by compounding kuh-seng, biota orientalis, angelica sinensis, gentiana macrophylla, fructus cnidii and herba violae are adopted, the fermentation effect is obviously changed due to the damage of the compound systems of schizosaccharomyces pombe, pichia pastoris powder and filamentous bacillus succinogenes, and the fermentation product cannot achieve the effect of obviously inhibiting DHT generation, so the anti-alopecia effect for androgenetic alopecia patients is obviously reduced.
