The testing method comprises the following steps: putting the test object into a patch tester with the dosage of 5g, covering the patch tester with the test object on the back or the curved side of the forearm of a subject by using a non-stimulated cloth-based adhesive tape, lightly pressing the patch tester with the palm to uniformly adhere the patch tester on the skin surface for 24 hours, removing the test object for 30 minutes, and observing skin reaction after the indentation disappears. If the result is negative, the observation is carried out again 24 hours and 48 hours after the patch test, respectively.
Evaluation criteria:
Level 0: a negative reaction;
stage 1: suspicious reactions, only weak erythema;
2 stages: weak positive response, erythema, infiltration, edema, and possibly papules;
3 stages: strong positive reaction, erythema, infiltration, edema, papule, and reaction beyond the test area;
4 stages: very strong positive response, marked erythema, severe infiltration, edema, blepharospermia, and response beyond the test area.
Test results: all subjects were negative for skin response.
The test results show that the acne-removing facial cleanser composition and the skin care product bath cream provided by the invention are mild and free of irritation to skin, and are safe to use.
Cytotoxicity test
Human corneal epithelial cells HCEC with stable growth state were taken, and the cell concentration was adjusted to 1X 10 5/mL, and inoculated into 96-well cell culture plates (except outermost peripheral wells and blank wells) at 100. Mu.L per well. 200. Mu.L of PBS was added to the outermost wells of the cell culture plates to reduce evaporation of the medium. Placing the cells in a carbon dioxide incubator, and culturing at 37+/-1 ℃ under the condition that the concentration of CO 2 is 5.0+/-1% until the cell fusion reaches more than 90%. Samples were added at a concentration of 10mg/L, and a blank, sample set, 100. Mu.L per well, was set. And (3) starting to add samples (sample groups) and PBS (blank control), keeping the time interval of adding samples in every two rows consistent, ensuring that the intervention time of each test object to cells is 5 min, sequentially sucking the test objects after 5 min, adding PBS into each hole, and cleaning the test objects for 2 times. The liquid in the hole is sucked by a vacuum suction pump or a discharge gun, and the operation is carefully performed so as not to suck out the living cells to influence the experimental result. 100. Mu.L of complete medium and 10. Mu.L of CCK-8 solution were added to each well, and the wells were placed in an incubator for cultivation for about 2 hours. At 450 nm wavelengths, absorbance (OD) values were measured and cell viability calculated; the results are shown in Table 6.
As can be seen from the results of the experiments in Table 4, the cell viability was maintained at 95% or more. The experiment collocation composition has small influence on HCEC cell viability.
Antibacterial agent for acne bacillus
Referring to WS/T650-2019, 24h fresh bevel cultures of acne bacillus were washed with PBS and diluted to 5.0X10 5CFU/mL-5.0×106 CFU/mL for use using a bacteriostatic ring test.
Preparation of antibacterial tablets: the cut diameter was 5 mm and the thickness was no more than 4 mm pieces of filter paper, each 4 pieces being a set immersed in 10mL of sample solution.
Preparation of negative control plaques: taking samples of the same material, preparing into discs (blocks) with the same size as the test group, and soaking in sterile water.
The acnes suspension with the concentration of 5.0X10 5CFU/mL-5.0×106 CFU/mL is dipped by a sterile cotton swab and evenly smeared on the surface of a proper culture medium flat plate for 3 times. And (3) each time of smearing, the flat plate rotates for 60 degrees, and finally, the cotton swab is smeared around the edge of the flat plate for one circle. The plate was covered and left to dry at room temperature for 5min. 1 bacteria-staining plate is attached to each test, and 4 test pieces, 1 negative control piece and 5 pieces are attached to each plate. The sample piece is stuck on the surface of the flat plate by using sterile forceps, the negative control sample piece is stuck on the center of the flat plate, the test sample piece is stuck on the periphery, and after the sample piece is stuck, the sample piece is lightly pressed by using the sterile forceps, so that the sample piece is clung to the surface of the flat plate. The distance between the centers of the sample pieces is more than 25 mm and the distance between the centers of the sample pieces and the periphery of the flat plate is more than 15 mm. The plate is covered, incubated at 36+ -1deg.C for 16-18 h, and the test is repeated 3 times. The diameter of the inhibition ring (including the patch) is measured by a vernier caliper and recorded, when the inhibition ring is measured, the inhibition ring which grows uniformly and completely in a sterile mode is selected for carrying out the measurement, and the diameter of the inhibition ring is measured by taking the outer edge of the inhibition ring as a boundary.
The negative control sample should not have a bacteriostasis ring, and the test sample has a bacteriostasis effect when the diameter of the bacteriostasis ring is more than 7 mm; and if the diameter of the inhibition ring is less than or equal to 7 mm, judging that the inhibition ring has no inhibition effect. And (3) judging that the test is qualified if the test has an antibacterial effect in 3 repeated tests. See table 7 for specific results.
The results show that: the comparison of the examples and the comparative examples shows that the examples are superior to the comparative examples in bacteriostasis performance, so that the preparation is carried out according to the proportion defined by the invention, and the effect achieved by the invention cannot be achieved by any default. And the default of the comparative example also proves that certain synergistic effect exists between the proportions defined by the invention.
Fourth, percutaneous Water loss (TEWL) test
The transcutaneous water loss (TEWL) is an important parameter in assessing the barrier function of the skin and is indicative of the loss of moisture in the deep dermis through evaporation of the epidermis. The higher the rate of percutaneous loss of water indicates that the more moisture is lost through the skin, the weaker the skin's ability to retain moisture.
The rate of percutaneous water loss is closely related to the hydration of the stratum corneum and is a common indicator reflecting the barrier function of the stratum corneum. In the development process of the cosmetics, the efficacy of the moisturizing cosmetics can be evaluated by testing the TEWL value of skin moisture loss. In addition, the percutaneous water loss rate can also be applied to allergic patch tests, contact dermatitis, physiotherapy, burn and monitoring of new tissues, and whether the protective function of the skin is destroyed can be found in time.
Therefore, the percutaneous water loss rate is an important parameter for evaluating skin health, and has important significance for developing and evaluating cosmetics.
190 Healthy subjects between 18 and 60 years old, with intact persons on the inner side of the arm, were selected for grouping, 10 persons in each group, and 5 men and 5 women in each group, respectively. Sample and blank groups (i.e., negative control group: no test sample) were set, and the test areas were randomly distributed in the area 1cm 2 inside the arm of the volunteer. One area was used with 1g of test product and the other area was blank, as a negative control area, and before the use of the product (T0), 2 hours after the use of the product (T1), the forearm skin percutaneous water loss rate (TEWL) was collected, and the moisturizing effect of the product was evaluated by using the variation difference of the skin index of the product treatment area and the negative control area before and after the use of the product, and the average value was taken, to obtain the following results in table 8.
The experiments show that the shower gel prepared by the application examples 15-21 has good moisturizing effect on skin compared with the shower gel prepared by the comparative examples 23-34, can obviously increase the moisture content of skin horny layer and reduce the moisture loss of skin outside, and fully proves that the shower gel disclosed by the invention scientifically customizes a compound formula aiming at the moisturizing requirement of each layer of skin epidermis, so that the full-effect moisturizing care of reducing the percutaneous water loss rate, promoting the generation of hyaluronic acid, enhancing the internal cell water circulation, strengthening the surface water locking barrier and the like is achieved.
Fifthly, human body sensory test
1. Foam Properties
Reference GB/T13173-91, measured using the Ross-Miles method. 10g of the samples of examples 15-21 and comparative examples 23-34 were weighed, dissolved in 150mg/kg of hard water and fixed to a 1000mL volumetric flask, and the prepared solution was placed in a 40℃constant temperature water bath for 0.5h. After 200mL of the test solution was flowed from a height of 90cm to the surface of 50mL of the same test solution at the bottom of the graduated cylinder at 40℃and the liquid collided to generate foam, a stopwatch was immediately started and the corresponding foam height of the system at 30s and 5min was recorded. The foaming capacity is corresponding to the foam height (H α) corresponding to 30s, and the stability of the foam is corresponding to the ratio (H β/Hα) of the corresponding foam height (H β) to H α after 5 min. See table 9 for specific test data.
