Example 1
1. Effect of water-soluble tomato concentrate on proliferation of dermal papilla cells
1.1 preparation of Water-soluble tomato concentrate
The method as described in patent "CN200980148421.6" of p Luo Weisi natural products limited: tomato extracts were prepared using as starting material a commercially available cold crushed tomato pulp having a Brix of 28-30 ° (i.e. 28-30% solids, w/w) of <0.350AU (Brix) with a browning index (absorbance at 420m of a solution with a concentration of 12.5g soluble solids/L). The slurry was diluted with ultrapure water (1:5) and large particulate matter was removed by centrifugation and then clarified at room temperature using a Westfalia MSB-14 separator (centrifugal disk clarifier). Smaller particulate material is then removed by microfiltration at a temperature of no more than 45 ℃ to give a clear pale yellow solution free of insoluble, sedimentable (spin-down) solids and capable of passing through a 0.2 μm filter without loss of soluble solids. The solution was concentrated by evaporation to a 65 ° brix slurry under carefully controlled conditions and at temperatures not exceeding 50 ℃ to limit non-enzymatic browning reactions. A rapid pasteurization step (t=105 ℃,3 seconds) was introduced at the beginning of the evaporation step. The final product was characterized by a browning index <0.600AU and a total plate count of microorganisms <1000.
In order to obtain a raw material containing a more concentrated bioactive ingredient, the sugar in the product is removed as described below. A column of L30L resin containing FPX66 resin (Rohm and Haas) was prepared and equilibrated in ultrapure water at 4 ℃. The above material was diluted to about 8 Brix with ultrapure water and passed through the resin column at a flow rate of about 260L/min, with the temperature maintained at 4 ℃. The column filtrate was discarded. Once all the required material has passed through the column, about 130L of flush water is passed through the column and discarded. Thereafter, the compounds that have remained on the resin are eluted by the following steps: 130L of hot water (75 ℃) was passed through the column, then 130L of 80% ethanol was passed through the column, and then another 130L of hot water was passed through the column. All eluting materials were retained and combined to give about 400L of an approximately 25% ethanol solution containing the compound of interest. The dilute solution containing the compound of interest was concentrated by reverse osmosis using a Trisep AC5 membrane at a temperature of about 30 ℃. The ethanol/water solvent passes through the membrane while all compounds dissolved therein remain within the membrane. Once the dilute solution was concentrated 10-fold, i.e. the volume was reduced to 40-50L, diafiltration was started, during which ultrapure water was added to the retentate at the same rate as the permeate removal rate. Thus, the ethanol concentration of the solution will gradually decrease from 25% to <5%.
The ethanol solution containing about 15-20% solids was then spray dried using an Anhydro spray dryer to give a golden fine powder with a water content of <6%, which is the final enriched water-soluble tomato extract.
The water-soluble tomato extract mainly comprises: (a) glycosylated phenolic acid or phenolic ester or derivative thereof; (b) glycosylated flavonoid; and (c) a nucleoside. The water-soluble tomato extract is separated from the pericarp, seed fragments, high molecular weight proteins, pectin and carotenoids (e.g. lycopene) by means of separation, and is a gaseous lipid stabilized in the form of droplets in an aqueous solution in the presence of pectin and proteins. I.e. the water-soluble tomato extract used in the present application is substantially free of lycopene.
1.2 principle of experiment
The CCK-8 cell viability detection kit (CK 04, dojindo, japan) contains WST-8 (2- (2-methoxy-4-nitrophenyl) -3- (4-nitrophenyl) -5- (2, 4-disulfophenyl) -2H-tetrazolium monosodium salt), and when the WST-8 is oxidized and reduced by intracellular dehydrogenase in the presence of an electronic carrier, water-soluble orange yellow formazan dye generated can be dissolved in a tissue culture medium, the color depth of the orange yellow formazan dye is in direct proportion to cell proliferation and in inverse proportion to cytotoxicity, the color depth of the orange yellow formazan dye is in linear relation to the cell number of the same cell, and the OD value is measured at the wavelength of 450nm by using an enzyme-labeled instrument, so that the number of living cells can be indirectly reflected.
1.3 Experimental methods
Taking cultured primary rat papilla cells, and adjusting cell suspension concentration to 5.0X10 4 And inoculating the cells to a 96-well cell culture plate for 24 hours per ml, and carrying out CCK8 experiments after the cells are attached. Adding culture medium containing 8 kinds of water-soluble tomato concentrates with different concentrations into experimental group, and parallel control group containing 5 kinds of common tomato powder with different concentrations (product of fructus Lycopersici Esculenti after heat crushing, pulping, vacuum concentrating, spray drying); the blank group is added with a culture medium without medicines, after incubation for 48 hours, CCK8 detection reagent is added, and after incubation is continued for 4 hours, OD value of each hole at 450nm wavelength is detected.
Cell proliferation rate calculation method: cell proliferation% = (experimental OD-blank OD)/(control OD-blank OD) ×100%.
1.4 experimental results
The experimental results of the effect of the water-soluble tomato concentrate on dermal papilla cell proliferation are shown in table 1.
TABLE 1
Note that: "/" indicates that proliferation was not detected.
As shown in Table 1, the water-soluble tomato concentrate can stimulate proliferation of hair papilla cells at 62.5-1000. Mu.g/mL, and the proliferation-promoting effect of 500. Mu.g/mL is optimal, compared with the control group, the proliferation-promoting rate of the water-soluble tomato concentrate is 188%, and the common tomato powder basically does not stimulate proliferation of hair papilla cells, and the high concentration can inhibit cell proliferation.
2. Effect of water-soluble tomato concentrate on reducing dermal papilla apoptosis
2.1 principle of experiment
Deoxyribonucleotide terminal transferase mediated gap end labeling (terminal-deoxynucleotidyl transferase mediated nick end labeling, TUNEL): when cells undergo apoptosis, some DNA endonucleases are activated, which cleave the genomic DNA between nucleosomes. Upon cleavage of genomic DNA, the exposed 3'-OH can be catalyzed by terminal deoxynucleotidyl transferase with the addition of green fluorescent probe Fluorescein (FITC) -labeled dUTP (Fluorescein-dUTP), so that apoptotic cells can be detected specifically and accurately by fluorescence microscopy or flow cytometry, while normal or proliferating cells have little cleavage of DNA and thus no 3' -OH formation and can be stained very rarely.
2.2 Experimental methods
Three optimal drug concentrations of 250 mug/mL, 500 mug/mL and 750 mug/mL obtained by CCK8 experiments are selected, the culture medium containing water-soluble tomato concentrate is added into an experimental group, the culture medium without drug is added into a blank group, and after incubation and culture for 48 hours, 125 mug/mL of common tomato powder is used in a parallel control group. Staining was performed with TUNEL staining kit (C1086, beyotime, china), and observation and photographing were performed with a fluorescence microscope.
2.3 experimental results
The apoptosis rate of the control group is 7.90+/-2.13%, the apoptosis rate of the tomato concentrate treated with 500 mug/mL is 3.42+/-1.48% (p < 0.05), the apoptosis rates of the tomato concentrate treated with 250 mug/mL and 750 mug/mL are 8.49+/-2.45% (p > 0.05) and 24.40+/-7.13%; the apoptosis rate of the common tomato powder treated with 125 mug/mL is 8.21+/-1.54% (p > 0.05).
FIG. 1 is a graph of the results of TUNEL staining of cells after treatment with water-soluble tomato concentrate at various concentrations, as described in connection with FIG. 1 and the above results: tomato concentrate at low concentration (250 μg/mL) is not sufficient for hair papilla anti-apoptotic capacity, but combined with its effect of promoting cell proliferation, the overall benefit to cells is greater than bad. While high concentrations (750 μg/mL) of tomato concentrate, while helpful in proliferation of hair papilla cells, also induced apoptosis, affecting the overall life span of the cells. The tomato concentrate with medium concentration (500 mug/mL) has the effects of promoting proliferation and division of cells on hair papilla cells, resisting apoptosis on hair papilla cells, promoting cell growth and prolonging cell life. And the common tomato powder cannot play a remarkable role.
3. Effect of Water-soluble tomato concentrate on dermal papilla cell growth factor expression
3.1 Experimental methods
Three optimal drug concentrations of 250 μg/mL, 500 μg/mL and 750 μg/mL obtained by CCK8 experiments were selected, the experimental group was added with the medium containing the water-soluble tomato concentrate, the blank group was added with the medium containing no drug, after incubation for 48 hours, the cell culture supernatant was taken and ELISA was performed using VEGF (EMC 103.96, neobioscience, china), IGF-1 (EMC 125.96, neobioscience, china), FGF-7 (XY 9M1023, XYBIotechnology, china) and TGF- β1 kit (EMC 107b.96, neobioscience, china).
3.2 experimental results
The standard curve is drawn, taking VEGF as an example, and the results are shown in Table 2 and FIG. 2, wherein the X-axis of the abscissa in FIG. 2 represents the concentration of water-soluble tomato concentrate and the Y-axis of the ordinate represents the OD value.
TABLE 2
Similarly, standard curves for each cytokine can be obtained according to the instructions of the kit, and the factor concentration calculated according to the standard curves, and the results are shown in Table 3.
TABLE 3 Table 3
From the above results, it can be seen that the water-soluble tomato concentrate can increase VEGF (P>0.05)、IGF-1(Represents p<0.05 And significantly reduces TGF- β1 production (+)>Represents p<0.05,/> Representing P<0.01)。
The catagen (catagen) of the hair cycle is an apoptosis-driven process, accompanied by terminal differentiation, proteolysis and matrix remodeling. There are studies showing that VEGF accelerates hair regeneration and increases the size of hair follicles and hair shafts by inducing perifollicular angiogenesis, improving head blood circulation and nutrient supply; IGF-1 is an important factor for regulating hair follicle mitosis and growth differentiation, and the signal transduction pathway has the effects of affecting hair follicle proliferation, tissue remodeling, hair growth cycle conversion and hair follicle differentiation, and up-regulating IGF-1 contributes to the restoration of dermal hair follicle papilla cells and maintains hair induction capacity. Meanwhile, as inhibitors of keratinocyte proliferation and inducers of keratinocyte apoptosis, transforming growth factor beta 1 (TGF-beta 1) has been proposed to play an important role in the regulation of the catagen phase, and the water-soluble tomato concentrate of the present application is capable of significantly reducing TGF-beta 1 production (p < 0.05) in primary papilla, helping to induce or maintain the growth phase of hair follicles, helping to achieve self-remodeling of hair follicles.
The results show that the water-soluble tomato concentrate can obviously promote proliferation of hair papilla cells, inhibit apoptosis of hair papilla cells, especially at 500 mug/mL, promote expression of VEGF and IGF-1 in hair papilla, and reduce expression of TGF-beta 1, and the water-soluble tomato concentrate has the effects of promoting hair follicle to maintain a growing period and inhibiting entering a retrograde period, so that the water-soluble tomato concentrate has good effects of preventing alopecia and promoting hair growth.
4. Effect of Water-soluble tomato concentrate on hair growth in mouse alopecia models
4.1 Experimental methods
Experimental animals: c57BL 6 mice, females, 8 weeks, 15-20g, syngeneic. The skin color pink was selected and the hair was a resting stage mouse. The phase change of the hair period can be judged according to the skin color of the mice, the skin in the hair growth period is black, the retrograde period is gray, and the resting period is pink. Establishing a murine alopecia model: mixing rosin and paraffin at a ratio of 1:1, heating to melt, uniformly coating on the back of a mouse, wherein the coating area is about 3cm multiplied by 3cm, cooling and solidifying, and removing the back hair. The anagen phase is confirmed to be in the telogen phase based on the skin color.
Grouping 8 mice confirmed to be in resting stage; wherein: experimental group: gastric administration of 500mg/mL water-soluble tomato concentrate; positive control group: 2% minoxidil solution; negative control group: purifying the water.
The administration mode is as follows: the medicine is filled into stomach and is administrated 1 time a day, the daily administration amount is 0.15mL, and the medicine is continuously administrated for 21 days.
Results observations and recordings: the skin and hair color changes of the mice are observed visually every day, and the time from the administration to the beginning of blackening of the skin color of the hair removal area at the back of each animal is recorded; the negative control group records the time from day 1 after depilation to the onset of skin darkening in the dehairing area; the dehairing area begins to darken to the time of full hair growth.
4.2 experimental results
As a result, as shown in fig. 3, the mice of the experimental group developed a black local skin on day 5 after depilation, the hair was grown in the anagen phase, the hair was grown after about 10 days, the skin color was changed to pink (telogen phase) again via gray (anagen phase) on day 21, and the skin black and gray remained for about 16 days in total; the positive control mice gradually appear black on local skin on day 5 after dehairing, hair enters anagen phase, new hair grows after 9 days, skin color changes to pink (rest phase) through gray (retrogressive phase) on day 21, and skin black and gray last for 16 days; the local skin of the mice in the negative control group changed from pink to black on the 6 th day after depilation, new hair grew out after 11 days, and the skin color changed from gray to pink again after 20 days, and the skin black and gray lasted for 14 days.
The experiment shows that compared with the mice in the negative control group, the skin color of the mice in the experiment group is changed from pink to black 24 hours in advance, the skin color of the mice in the experiment group is changed from black to gray 24 hours later, the time of the growing period of the hair follicle is about 48 hours longer than that of the mice in the negative control group, the result is similar to that of the positive control group, and the solvent group and the negative control group have no obvious difference, so that the oral water-soluble tomato concentrate can promote the hair follicle to enter the growing period, prolong the growing period and delay the entering the retrograde period.
In conclusion, under the experimental conditions, the water-soluble tomato concentrate has the effects of promoting hair follicles to enter a growing period, prolonging the growing period and delaying entering a withdrawal period for a mouse alopecia model, so that the water-soluble tomato concentrate has good effects of preventing alopecia and promoting hair growth.

(2) Preparing 130L of resin column containing FPX66 resin, balancing in ultrapure water at 4 ℃, diluting the intermediate product to have a Brix of 8 DEG, loading the resin column at 260L/min, maintaining the temperature at 4 ℃, once all substances are adsorbed on the resin column, eluting with 130L of washing water, discarding the eluent, then eluting with 130L of hot water at 75 ℃, then eluting with 130L of 80% ethanol, finally eluting with 130L of hot water at 75 ℃, collecting and combining the eluents to obtain 400L of ethanol dilute solution with a concentration of 25%, concentrating the 400L of ethanol dilute solution with a concentration of 25% by reverse osmosis by using Trisep AC5 membrane at 30 ℃, allowing ethanol/water solvent to pass through the membrane, keeping all compounds dissolved in the membrane, once the dilute solution is concentrated to 1/10 of the original volume, namely the volume of the solution is reduced to 40L, beginning to permeate, adding ultrapure water into the retentate at the same rate as the removal rate of the filtered solution, finally gradually reducing the concentration of ethanol dilute solution from 25% to a concentration of <5% by reverse osmosis, and finally obtaining the water-soluble fine powder with a concentration of <6% by spray of water-mist of water-soluble tomato.
2. The use according to claim 1, wherein the product further comprises other substances having an auxiliary effect on hair growth and hair growth, the substances comprising one or more of cowberry fruit extract, grape seed extract, saw palmetto extract, wheat germ extract, semen Cuscutae extract, ginseng extract, chamomile extract, turmeric extract, silybum marianum extract, millet extract, linseed oil, DHA, EPA and vitamin E.
3. The use according to claim 2, wherein the dosage form of the oral product comprises solutions, powders, tablets, granules, capsules, emulsions and suspensions.
4. The use according to claim 3, wherein the product further comprises a health food or a pharmaceutically acceptable adjuvant.
