Description
Technical Field
The application relates to the field of shampoo products, in particular to a composition for removing dandruff and relieving itching, a preparation method of the composition, shampoo, a preparation method of the shampoo and a medicament.
Background
Dandruff are fine white or grey scales that accumulate on the surface of the scalp. Dandruff is closely related to stratum corneum dysfunction. Most of the dandruff is believed to be caused by malassezia fungal infections. Malassezia is a lipophilic fungus, is a normal flora of the scalp, and is distributed on the surface of the scalp through a pilosebaceous canal under the influence of androgens after puberty, and after the malassezia ingests sebum, a large amount of malassezia is propagated, free fatty acids are generated, and the fatty acids penetrate into the stratum corneum to damage the skin barrier. Disruption of the barrier in turn results in excessive synthesis and secretion of sebum, which in turn provides a rich diet for the proliferation of malassezia, thus forming a vicious circle. Another cause of dandruff is dry scalp. Dry skin or scalp skin barrier is damaged due to frequent washing, scalding, dyeing, etc., so that protease activity responsible for exfoliation of epidermal cells is reduced, and epidermal cells cannot be normally exfoliated, thereby causing dandruff.
Currently, common antidandruff agents in shampoo products are ZPT (zinc pyrithione), OCT (piroctone olamine), ganbazole, and the like. Although ZPT has a good antibacterial effect and has been widely used as an anti-dandruff ingredient for more than 60 years, the european commission revised the european cosmetics regulation annex II ban ingredient list at 11, 2021, in which ZPT was formally effective at 3-1-2022 due to the GHS classification having reproductive toxicity 1B, which also means that the cosmetic anti-dandruff agent ZPT was formally banned in the european cosmetics. At present, ZPT is still listed in a limit raw material catalog and a standard preservative catalog of a cosmetic safety technical specification (2015) edition in China, but the requirements of use concentration/use specification are strict, and the use of ZPT in China is expected to be tightened in the future.
The piroctone olamine salt (P.O.) has the same anti-dandruff effect as ZPT, and has been widely used in daily chemical industry at home and abroad, but is mainly used in middle-high-end washing and caring products due to higher production cost. And its antipruritic effect is poor.
The gamboge has a certain inhibiting effect on the oomycetes or the oopityriasis causing dandruff and the candida albicans and the trichophyton, but the sole use of the gamboge has poor effect of removing dandruff. And it is harmful to aquatic organisms, and can pollute the aquatic environment.

A shampoo was provided, and the respective component contents of the shampoo provided in each comparative example are shown in table 2; the preparation method is the same as the preparation steps of the shampoo provided in the previous examples.
TABLE 2
The performance of the shampoos provided in the examples and comparative examples is tested in the following.
Test example 1
In vitro anti-dandruff performance test:
and (3) testing the inhibition effect of the shampoo on malassezia furfur by adopting a bacteriostasis ring method, and carrying out a bacteriostasis ring test for evaluating the anti-dandruff effect of each group of anti-dandruff shampoo under the same test condition.
Experimental materials
1. The main instrument is as follows: constant temperature incubator, superclean bench, puncher, pipette, forceps, sterilization flat board.
2. Culture medium: leemming & Notman medium was modified.
3. And (3) strain: malassezia furfur.
4. Test sample: shampoo of example 3, example 4, example 5, example 6 and comparative examples 7, 2, 3, 8.
Experimental details
1. Malassezia furfur is prepared into bacterial suspension with the concentration of about 105 cfu.
2. Uniformly coating on a Leemung & Notman culture medium flat plate, and then placing in a sterile operation table to blow dry the bacterial liquid on the flat plate.
3. And quantitatively adding each group of shampoo samples into the flat plate, culturing in a constant temperature incubator at 30 ℃ for 5-7 days, observing results, and recording and measuring the condition of the inhibition ring.
Experimental results
Recording the results as the diameter of the inhibition ring, unit: millimeter.
Among them, fig. 1 shows the effect of shampoo on inhibition of malassezia furfur, and as can be seen from fig. 1, both comparative example 2 (without dichlorobenzyl alcohol) and comparative example 7 (with 0.02% less dichlorobenzyl alcohol) have unsatisfactory antibacterial effects, and example 3 (0.04% dichlorobenzyl alcohol) has good effect of inhibiting malassezia furfur of dandruff microorganisms. As can be seen from comparison of example 3 and example 4, the effect of inhibiting malassezia furfur is better as the amount of dichlorobenzyl alcohol is increased, but if the amount exceeds 0.15%, the limit of the 2015 cosmetic safety specification is not met, and the cost is too high to have an economic effect.
Sodium salicylate inhibition malassezia synergy test: as shown in fig. 2, the shampoo has a certain antibacterial effect in comparative example 3 (without sodium salicylate) and comparative example 8 (with sodium salicylate being 0.05% lower), but the antibacterial effect is quite different, and the effect of inhibiting the pityromonas versicolor of the dandruff microorganism in example 5 (0.1% sodium salicylate) is obviously enhanced, which means that the sodium salicylate with 0.1% has a better synergistic effect on inhibiting the pityromonas versicolor, the better the synergistic effect on inhibiting the pityromonas versicolor with the increase of the dosage is, the dosage is up to 2% (example 6), the antibacterial effect reaches the highest peak, but the dosage is more than 2%, the sodium salicylate causes a certain stimulation to the scalp, and the cost is too high, so that the shampoo is more suitable for 0.1-2.
Test example 2
Moisture retention performance test:
1. polyglycerol-10 laurate moisture retention test: the tester washes hair with the shampoo of examples and comparative examples at 40 c and dries after washing.
The scalp cuticle moisture content was measured for 0 hours, again for 4 hours in a constant temperature and humidity room, and for 8 hours in a constant temperature and humidity room.
Skin moisture content test probe (instrument software CK-MPA-Multi-Pro be-Ada pterFB) was used: corneometer CM825 measures scalp stratum corneum moisture content. Fig. 3 is a graph of scalp cuticle moisture content (where n=12 indicates the number of test persons and the sample size, and the average result is taken from the data; and the ordinate unit: CU), and as is clear from fig. 3, comparative example 1, in which polyglycerin-10 laurate was not added, has the lowest scalp cuticle moisture content, and next, comparative example 4, in which polyglycerin-10 laurate content is 0.05%, and after the polyglycerin-10 laurate usage amount is more than 0.1% (example 1), the moisturizing effect of the shampoo starts to be greatly improved, and after the polyglycerin-10 laurate usage amount is 2% (example 2), the moisturizing effect is improved to a limited extent. In view of the moisturizing effect and the economical efficiency of the cost, the added amount of 0.1 to 2% of polyglycerol-10 laurate is preferable. In addition, the moisture content of the scalp cuticle after 0h,4h and 8h of comparative example 1, in which no polyglycerol-10 laurate was added, was not greatly different, but the moisture content of the scalp cuticle increased with time after the addition of polyglycerol-10 laurate, indicating that polyglycerol-10 laurate also has a long-lasting moisturizing effect.
2. C12-13 sodium lactate alcohol moisturizing and synergy test; the tester washes hair with 40 ℃ water using the shampoo of the examples and the comparative examples, dries and measures the moisture content of scalp cuticle after washing for 0 hours, measures the moisture content of scalp cuticle again in a constant temperature and humidity room for 4 hours, and measures the moisture content of scalp cuticle again in a constant temperature and humidity room for 8 hours.
Skin moisture content test probe (instrument software CK-MPA-Multi-Pro be-Ada pterFB) was used:
corneometer CM825 measures scalp stratum corneum moisture. Fig. 4 is a graph of scalp cuticle moisture content (where n=12 refers to the number of test persons and the sample size, and the average result of data) and as can be seen from fig. 4, the scalp cuticle of comparative example 5 without added sodium C12-13 lactate and comparative example 10 with added 0.03% has a certain moisture content, but the moisture retention effect of the shampoo after use of the shampoo after added sodium C12-13 lactate (example 9) with 0.05% starts to be improved, and the moisture retention effect is improved greatly after the use amount is 2% (example 10), which indicates that the sodium C12-13 lactate has the effect of enhancing the moisture retention and enhancing effects. However, since the foamability of the shampoo is greatly reduced when the amount of the sodium lactate C12-13 alkoxide added exceeds 2%, the amount of the sodium lactate C12-13 alkoxide added is preferably 0.05 to 2%.
3. Allantoin scalp repair test: the testers weekly, wednesday, friday were evaluated for room moisture content, and shampoo compositions of examples 7, 8 and 4 and 9 were rinsed with 40 ℃ water, dried after rinsing, and measured for moisture content on day 0, day 7, day 14, and scalp stratum corneum.
Skin moisture content test probe (instrument software CK-MPA-Multi-Pro be-Ada pterFB) was used: corneometer CM825 measures scalp stratum corneum moisture. From fig. 5, it is clear that allantoin can increase the moisture content of the scalp stratum corneum, and the moisturizing effect increases with the increase of the amount. The more pronounced the increase in stratum corneum moisture content with increasing trial time (e.g., day 28 data), which suggests that the moisturizing condition of the scalp stratum corneum is improved, especially the increase in allantoin levels is pronounced from 0.05% to 2% (examples 7-8), with more than 2% increasing more slowly.
4. Clinical test of integral anti-dandruff moisturizing performance:
the shampoo of the example 2 and the comparison example 1, the comparison example 2, the comparison example 3, the comparison example 4 and the comparison example 5 and the commercially available active type of the cleaning and man anti-dandruff shampoo are packaged in test packaging bottles of 60 parts to 200mL and sent to test crowd (60 people) for comparison. Each shampoo was used for 4 weeks continuously, and the tester scored the questionnaire test item according to the sense of use. The scoring criteria used 7 points, 1 point being the worst and 7 points being the best, the higher the score the better. The final score is shown in figure 6 (ordinate unit: score) as a statistical average score.
As can be seen from fig. 6, the compositions of the present application were evaluated more highly in each study dimension than the commercial cleaning men's anti-dandruff shampoo sports type.
Further, the composition has synergistic interaction, and the comparison of the comparative example 1 and the example 2 shows that the polyglycerol-10 laurate indirectly increases the efficacy of removing dandruff and inhibiting itch besides obviously increasing moisture; as can be seen by comparing comparative example 2 with example 2, the addition of dichlorobenzyl alcohol enhances the anti-dandruff and antipruritic effects; as can be seen from comparison of comparative example 3 with example 2, sodium salicylate has effects of removing cutin and sterilizing, and further improving capability of removing dandruff and relieving itching; comparison of comparative example 4 with example 2 shows that allantoin has a moisturizing effect, and significantly increases the moisture content of the scalp. Comparison of comparative example 5 with example 2 shows that sodium lactate C12-13 alkoxide has moisturizing and antipruritic effects; 5 substances are synergistic with each other, so that the effects of removing dandruff, relieving itching and preserving moisture are improved.
The foregoing description is only of the preferred embodiments of the present application and is not intended to limit the same, but rather, various modifications and variations may be made by those skilled in the art. Any modification, equivalent replacement, improvement, etc. made within the spirit and principles of the present application should be included in the protection scope of the present application.
