Abstract
The invention relates to the technical field of cosmetics, and particularly provides a composition, application thereof and a cosmetic. The composition of the invention comprises a compact Luo Mu extract, a gynostemma pentaphylla extract, a madecassoside and a lotus root extract. Compared with the prior art, the composition can prevent histamine release, inhibit the activity of hyaluronidase and repair skin barrier through the synergistic effect of the components, improve skin through the various effects, and improve the effects of anti-inflammatory, allergy-relieving and repairing; the skin care composition is safe and has no irritation, and has wide application range.
Description
Technical Field
The invention relates to the technical field of cosmetics, in particular to a composition, application thereof and cosmetics.
Background
The skin is the largest organ in the human body, is directly contacted with the external environment, and has the functions of protecting, excreting, regulating the body temperature, sensing external stimulus and the like.
In recent years, due to climate change, air pollution, and complicated types and components of cosmetics, the pressure of work and life is high, diet is irregular, and food is stimulated, so that the possibility of appearance and stimulation of sensitive skin is improved. Consumers affected and bothered by sensitive skin are increasing, and the perception rate and the attention of the sensitive skin are gradually increased in addition to the improvement of the health consciousness of the skin of people.
The dermatitis comprises allergic dermatitis, seborrheic dermatitis, hormone dermatitis, etc. There are two main categories of common allergic dermatitis: contact dermatitis and cosmetic dermatitis. Allergic reactions caused by different pathogenic factors are different. Among them, irritant contact dermatitis is the most common skin diseases (facial cleansers, pigments and astringents) using cosmetics, such as irritation, itching, redness, swelling and the like, and swelling and desquamation of skin may occur in severe cases. Allergic contact dermatitis occurs less frequently than irritant contact dermatitis. Cosmetic ingredients that can cause skin allergy include fragrances, preservatives and sunscreens, hair dyes, etc., can cause skin irritation, itching, ulceration, redness, swelling symptoms, and even skin foaming.
The cosmetic has the main ways of preventing and treating skin sensitivity and irritation: 1. inhibiting inflammatory mediator release such as histamine; 2. enhancing body barrier function; 3. scavenging excessive free radical in vivo.
Although some people add components with the conditioning effect on facial skin into the skin care product, the components are single, the overall consideration is not comprehensively given to the components, the effect of relieving and repairing is not obvious, and the effects of relieving and repairing are not well considered; how to achieve zero stimulation of the product, quickly permeate the skin and absorb the skin, and the effect components exert extremely good effects on the skin of the face, thus being a problem to be solved urgently in the research of the current skin care products.
Disclosure of Invention
Accordingly, the present invention has been made keeping in mind the above problems occurring in the prior art, and an object of the present invention is to provide a composition which can prevent histamine release, inhibit hyaluronidase activity, repair skin barrier, improve skin by the above-mentioned various actions, and enhance anti-inflammatory and allergy-relieving effects, and a cosmetic, and an application thereof; the skin care composition is safe and has no irritation, and has wide application range.
In order to achieve the above purpose, the present invention adopts the following technical scheme:
A composition comprises extract of Mi Luo Mu, extract of Gynostemma Pentaphyllum, madecassoside, and extract of Nelumbo nucifera Gaertn.
In the present invention, the composition comprises, in parts by weight:
0.1-2.5 parts of extract of Mi Luo Mu;
0.2-2.5 parts of gynostemma pentaphylla extract;
0.1-2 parts of hydroxy asiaticoside;
0.1-3 parts of lotus root extract.
The invention has no limitation on the sources of the extract of the compact Luo Mu, the extract of the gynostemma pentaphylla, the madecassoside and the extract of the lotus root, and the invention can be realized by adopting the conventional preparation method in the field or the commercially available products.
In the present invention, the composition comprises, in parts by weight:
0.1-2.5 parts of extract of Mi Luo Mu;
0.5-2.5 parts of gynostemma pentaphylla extract;
0.1-2 parts of hydroxy asiaticoside;
0.1-3 parts of lotus root extract.
Madecassoside is the main active ingredient of centella asiatica extract. Madecassoside reduces the production of pro-inflammatory mediators (IL-1, MMP-1), and increases and restores the self-barrier function of the skin, thereby preventing and correcting skin immune dysfunction. The madecassoside can promote the synthesis of collagen I and III, promote the secretion of mucopolysaccharide (such as hyaluronic acid synthesis), increase the water retention of skin, activate and update skin cells, make skin relax, promote and feel shiny, activate the fibroblast genes, strengthen the activity of basal layer cells of skin, keep skin elasticity and compactness, and smooth fine wrinkles on face.
The lotus root extract contains pectin, galactan, arabinose and mucin, has the effects of reducing inflammatory mediators, relieving inflammation, calming skin, and relieving skin allergy and redness.
In the present invention, the extract of Mime Luo Mu is Mime Luo Mushe/stem extract. The preparation method of the Mi Luo Mu extract comprises the following steps: crushing the honey Luo Mufen, and mixing with 8-10 times of water to obtain a honey Luo Mu extract; the preparation method of the Mi Luo Mu extract preferably comprises the following steps: crushing the honey Luo Mu, soaking the crushed honey in 8-10 times of water, and extracting for 1-3 hours to obtain the honey Luo Mu extract.
The extract of Mi Luo Mushe/stem has specific gallic acid, trehalose, and glyceroglycosides, which can effectively activate and regenerate skin, increase skin elasticity and moisture retention, relieve skin redness, resist erythema, enhance skin cell activity and metabolism, restore aging cells to young cell state, and promote skin activation and regeneration.
In the invention, the gynostemma pentaphylla extract is a gynostemma pentaphylla leaf extract. The preparation method of the gynostemma pentaphylla extract comprises the following steps: pulverizing herba Gynostemmatis, mixing with organic solvent, collecting filtrate, and concentrating to obtain herba Gynostemmatis extract;
The preparation method of the gynostemma pentaphylla extract preferably comprises the following steps: pulverizing herba Gynostemmatis, and sieving with 40 mesh sieve. Weighing 50g of powder, adding 500ml of 95% ethanol solution, reflux-extracting at 80deg.C for 2 times each for 2 hr, filtering, mixing the filtrates, concentrating under reduced pressure to recover solvent to obtain concentrated solution, and oven drying at 40deg.C under ventilation to obtain herba Gynostemmatis extract.
The extract of Gynostemma Pentaphyllum leaf contains phenols, sterols, steroid saponins and triterpenes, and has effects in increasing water channel number, promoting glycerol and water penetration, improving skin texture and enhancing skin barrier.
In the invention, the preparation method of the lotus root extract comprises the following steps: mixing lotus root with organic solvent in the material-liquid ratio of 1 (10-20) to obtain lotus root extract;
The preparation method of the lotus root extract preferably comprises the following steps: soaking lotus root in ethanol in the ratio of 1 (10-20), and extracting at 60-70 deg.c for 3-5 hr to obtain lotus root extract.

The experimental application example proves that the composition can effectively inhibit histamine release and hyaluronidase activity, prevent water loss on the surface of skin, and has good effects of resisting inflammation, relieving allergy and repairing skin.
The invention also provides cosmetics, comprising the composition and cosmetically acceptable auxiliary materials.
The cosmetic is preferably astringent, lotion, foundation, essence, facial mask, sunscreen, facial cream, eye cream, hand cream or body cream; the mask is preferably a patch type mask or a smearing type mask; the essence is preferably essence water and essence oil.
In the invention, the auxiliary materials are one or more of additives, moisturizers, thickeners, chelating agents, pH regulators and antioxidants;
the additive is panthenol and/or sodium hyaluronate.
In the present invention, the cosmetic preferably comprises, in parts by weight:
0.5-10 parts of the composition, 0.1-3 parts of panthenol, 0.01-0.3 part of sodium hyaluronate, 1-8 parts of humectant, 0.5-2 parts of thickener, 0.05-0.8 part of chelating agent, 0.1-0.5 part of pH regulator, 0.5-1.2 parts of antioxidant and water; water and 100 parts of the composition, panthenol, sodium hyaluronate, humectant, thickener, chelating agent, pH regulator and antioxidant.
In the invention, the humectant is one or more of glycerol, propylene glycol, 1, 2-hexanediol, octanediol, urea, trehalose, PCA-sodium and xylitol-based glucoside; in one embodiment of the invention, the humectant is urea, glycerol and xylitol-based glucoside in a mass ratio of 1:2.5:0.5; in one embodiment of the invention, the humectant is glycerin, octylglycol and urea in a mass ratio of 2:1:1;

(2) Cooling the mixed solution A, adding the skin care composition and the chelating agent, and uniformly stirring to obtain a mixed solution B;
(3) And adding a pH regulator and an antioxidant into the mixed solution B, uniformly stirring, and detecting to be qualified to obtain the cosmetic with the effects of comfort and repair.
Further, in the step (1), the heating temperature is 70-80 ℃, the stirring speed is 500-800r/min, and the stirring time is 20-30min;

The composition of the invention not only can effectively strengthen the skin against external stimulus, increase the natural restoration protection capability of the skin, moisten and soften the skin, reduce the release of inflammatory factors and remove redundant free radicals, but also can achieve the effects of skin relief and repair from different angles, strengthen the skin vitality, restore the cell vitality to be younger, isolate/reduce the contact and use of chemical substances from different sources, ensure that the product is safe and has no stimulus, and has wide application range.
The test examples prove that the composition can effectively prevent histamine release, inhibit the activity of hyaluronidase and repair skin barrier through the synergistic effect of the components, improve skin through the various effects, and improve the anti-inflammatory, allergy-relieving and repairing effects.

The preparation method of the lotus root extract comprises the following steps:
soaking lotus root in ethanol at a feed-liquid ratio of 1:10, and extracting at 70deg.C for 3 hr to obtain lotus root extract.

Comparative examples 1 to 10 were used
Except for the difference from application example 1 in that the skin care composition of application example 1 was replaced with the skin care compositions of application comparative examples 1 to 10, the remaining materials were unchanged, and the preparation method was the same as that of example 1.
Test example 1 product safety test
1. Test article cosmetic product original, application examples 1 to 6 and application comparative examples 1 to 10.
2. Negative control, blank control.
3. The subjects are 48 persons, 8 men and 40 women, and the ages of 22-58 years meet the volunteer selection standard of the subjects.
4. The test method comprises selecting qualified patch test equipment, placing test object about 0.020g-0.025g into patch test chamber, and placing control hole as blank (without any substances). The patch test with the test object is applied to the back or forearm of the subject by hypoallergenic tape, and is applied to the skin with palm light pressure for 24 hr. Skin reactions were observed according to the criteria of Table 2 for 30min (after disappearance of the indentations), 24h and 48h, respectively, after removal of the subject plaque tester, and observations were recorded.

Results: according to the skin response grading standard of the skin closed patch test of cosmetic safety technical Specification (2015 edition), the human skin patch test result of the test object shows that 48 subjects are negative in skin and should be scored (scoring grade: 0).
Test example 2 test for inhibition of histamine release
Peripheral blood basophils (KU 812) are often used as effector cells for studying allergic reactions, and the present invention uses a KU812 cell histamine release model to examine the effect of samples on KU812 cell pro-inflammatory cytokine expression.
Cell model establishment: cells were diluted with IMDM medium containing 10% fetal bovine serum, the cell density was adjusted to 2 x 10 6 cells/mL, and the resulting cell suspension was inoculated into well plates at an inoculum size of 150 μl per well.
Preparing and treating a tested sample: sample solutions of application examples 1 to 6 and comparative examples 1 to 10 were prepared in DMSO at a mass concentration of 10% and then diluted to a concentration of 0.3% by 10% fetal bovine serum culture medium to obtain test solutions.
Culturing and standing the cell suspension in a 36 ℃ and 5% CO 2 incubator for 10min, adding 20 mu L of test solution respectively, setting a control group without adding the test solution, pre-incubating for 2h in the incubator, and adding a stimulator (40 nM crotyl alcohol-12-tetradecanoate-13-acetate (PMA) and 1 mu m calcium ionophore A23187) respectively, wherein the PMA and the A23187 are prepared by using DMSO; after 2h, the cells were centrifuged, the cell supernatant was collected, and the histamine content in the supernatant was measured. The experimental results are shown in table 3.
Test example 3 test for percutaneous moisture loss
The principle of the transdermal moisture loss test is to measure the change in vapor pressure of moisture adjacent to the skin surface based on the diffusion principle. A cylindrical cavity measuring probe with two open ends is specially designed to form a relatively stable test small environment on the skin surface, the water vapor pressure gradient of the near epidermis (within about 1 cm) formed by the water loss of the stratum corneum at different two points is measured through two groups of temperature and humidity sensors, and the water amount evaporated through the epidermis is directly measured, so that the water loss condition of the skin surface is measured, and the moisturizing and skin barrier repairing effects of cosmetics on the skin surface can be evaluated. The calculation formula of the relative value of TEWL is:
TEWL relative value= (skin moisture loss of test sample-skin moisture loss of blank)/(skin moisture loss of positive control-skin moisture loss of blank).
The experimental method comprises the following steps: 48 healthy subjects 20-40 years old were selected and divided into 16 groups, and none of the subjects had a history of skin disease, the tested parts were normal, and no drugs or cosmetics irrelevant to the experiment were applied during the test. The temperature of the test environment is controlled at 25+/-1 ℃, the relative humidity is 40+/-5%, purified water at 36 ℃ is used for wiping the two arms of the subject before the test, and the subject is allowed to sit still for 30min in the test environment for the test.
Sample solution: the skin care compositions of application examples 1 to 6 and application comparative examples 1 to 10 were diluted in a mixed solvent of glycerin and water in a volume ratio of 1:5 to a skin care composition mass concentration of 1%, to prepare sample solutions; the positive control was sodium hyaluronate solution at the same concentration, and a blank control was set. Multiple regions were marked on each subject arm, typically 2 x 2cm 2 in size. And coating the tested sample, blank control and positive control sample on different areas of the arm of the same subject. The skin moisture loss in the test area was measured 3 hours after application by a skin moisture loss tester, the relative value of TEWL was calculated, and the experimental results are shown in table 3.
Test example 4 hyaluronidase in vitro inhibition assay:
1) The test components are divided into A, B, C, D groups, 0.5mL of hyaluronidase (500U/mL) is added to the group A and the group C respectively, 0.5mL of acetic acid buffer (pH value is 5.6) is added to the group B and the group D respectively, 0.1mL of CaCl 2 solution (2.5 mol/L) is added respectively, and the mixture is placed in a constant-temperature water bath kettle at 37 ℃ for 20 minutes;
2) Taking out the group A and the group B, respectively adding the sample solution into the group C and the group D, respectively adding distilled water into the group C and the group D, and then carrying out heat preservation for 20min in a constant-temperature water bath kettle at 37 ℃;
3) Taking out the group A and the group C, respectively adding 0.5mL of sodium hyaluronate solution (0.5 mg/mL), respectively adding 0.5mL of acetic acid solution (pH value is 5.6) into the group B and the group D, and then carrying out heat preservation for 30min in a constant-temperature water bath kettle at 37 ℃;
4) Taking out the groups A to D, standing at room temperature for 5min, adding 0.5mL of NaOH solution (0.5 mol/L) and 0.5mL of acetylacetone solution (3.5 mL of acetylacetone+50 mL of sodium carbonate solution with concentration of 1.0 mol/L) into each group, carrying out boiling water bath for 15min, ice water bath for 5min and standing at room temperature for 10min, adding 1mL of an Escherichia reagent (0.8 g of p-dimethylaminobenzaldehyde+15 mL of concentrated hydrochloric acid+15 mL of absolute ethyl alcohol) into each group, fully oscillating, adding 3mL of absolute ethyl alcohol into each group for dilution, standing at room temperature for 20min for color development, measuring absorbance value, and substituting the absorbance value into the following formula to calculate inhibition rate:
Hyaluronidase inhibition ratio (%) = [ (C-D) - (a-B) ]/(C-D) ×100%,
Wherein: a is the absorbance value of the sample solution, B is the absorbance value of the reagent blank solution, C is the absorbance value of the control solution, and D is the absorbance value of the blank control solution.
The in vitro inhibition rates of the anti-inflammatory allergy-free repair compositions of application examples 1 to 6 and application comparative examples 1 to 10 on hyaluronidase were measured by the above test methods, and the test results are shown in Table 3.

As shown in Table 3, the skin care composition with anti-inflammatory and allergy-relieving repairing effects of the present invention can prevent histamine release, inhibit hyaluronidase activity, repair skin barrier, improve skin through various effects, and improve anti-inflammatory allergy-relieving repairing effects through synergistic effects of the components. The skin-care gel is prepared by mixing various raw materials with good permeability and moisturizing effect, can continuously supplement nutrients for the skin, is more suitable for long-term use, has no stimulation, repairs the skin, improves the skin quality and increases the immunity of the skin.
From the results of the hyaluronidase inhibition rate experiments in table 3, it can be seen that: the application examples 1 to 6 all have the effect of inhibiting the activity of the hyaluronidase compared with the application comparative examples 1 to 10, but the anti-inflammatory and allergy-relieving repairing composition of the application examples 1 to 6 has the effect of inhibiting the hyaluronidase by more than 90 percent under the condition that the active ingredient is 6mg/mL, and has better effect than the anti-inflammatory and allergy-relieving repairing composition selected by the application comparative examples 1 to 10, which shows that the composition adopted by the invention has obvious synergistic effect.
