Abstract
The invention discloses an anti-aging composition and application thereof, and relates to the technical field of cosmetics. The anti-aging composition comprises the following components: peony extract, beta-alanyl hydroxyproline diaminobutyric acid benzylamine, sea buckthorn fruit oil and jojoba oil. After the peony extract rich in polyphenols, the beta-alanyl hydroxypropionyl diaminobutyric acid benzylamine with excellent anti-wrinkle performance, the sea buckthorn fruit oil with good oxidation resistance and the jojoba oil with excellent permeability and affinity are matched in a balanced way, the synergistic effect is effectively exerted, the skin elasticity and the skin tightening sense can be effectively improved, the fine lines are reduced, and the skin dryness is improved.

Technical Field
The invention relates to the technical field of cosmetics, in particular to an anti-aging composition and application thereof.
Background
The free radicals may cause protein damage, lipid peroxidation, nucleic acid damage, etc., thereby inducing various diseases. The antioxidation mechanism plays a role in antioxidation by providing hydrogen for the antioxidase, buffering physiological pH, chelating metal ions, capturing free radicals and the like. CN110051593a discloses a peony essence and a preparation method thereof, and the usage amount of peony extract, squalane and other active substances in the invention is high.
Therefore, it is desirable to provide an anti-aging composition which requires less kinds and amounts of active ingredients and has a good anti-aging effect.
Disclosure of Invention
The present invention aims to solve at least one of the technical problems existing in the prior art. Therefore, the invention provides an anti-aging composition which can effectively improve skin elasticity and skin tightening sense, reduce fine lines, lighten fine lines and improve skin dryness.

An anti-aging composition according to an embodiment of the first aspect of the present invention comprises: peony extract, beta-alanyl hydroxyproline diaminobutyric acid benzylamine, sea buckthorn fruit oil and jojoba oil.
The anti-aging composition according to the embodiment of the invention has at least the following beneficial effects:
the anti-aging composition of the embodiment contains peony extract rich in polyphenols, beta-alanyl hydroxypropionyl diaminobutyric acid benzylamine with excellent anti-wrinkle performance, sea buckthorn fruit oil with good anti-oxidation and anti-wrinkle effects, and jojoba oil with excellent permeability and affinity. The composition is designed through water-oil balance, absorption promotion and multidimensional anti-aging mechanism, and the peony extract, beta-alanyl hydroxy prolyl diaminobutyric acid benzylamine, sea buckthorn fruit oil and jojoba oil achieve unexpected effects through synergistic effect, so that the composition can effectively improve skin elasticity and skin tightening sense, reduce fine lines, lighten fine lines and improve dry skin.

The preparation methods of the essence, the eye cream, the mask liquid and the sleeping mask are as follows:
1) Heating butanediol, p-hydroxyacetophenone, 1, 2-hexanediol and bis-PEG-18 methyl ether dimethyl silane to 60 ℃, and completely dissolving to obtain a pretreatment phase for later use;
2) Adding glycerol, carbomer, xanthan gum and acrylic acid (esters) or C10-30 alkanol acrylate crosslinked polymer into a stirring pot, heating to 80 ℃ and stirring until the crosslinked polymer is completely dissolved;
3) Adding glycerol stearate, PEG-100 stearate, caprylic/capric triglyceride, polydimethylsiloxane, cetostearyl alcohol, cetyl alcohol, jojoba oil and fructus Hippophae oil into oil pan, heating to 80deg.C, and dissolving completely to obtain material;
4) Pumping the material body obtained in the step 3) into the stirring pot obtained in the step 2), starting homogenizing and emulsifying, stirring at a speed of 2500-3000 r/min, homogenizing and emulsifying for 5-10 min, vacuum defoaming, cooling to 60 ℃, and adding the pretreated phase obtained in the step 1) to obtain a mixture for later use;
5) After the mixture is cooled to 45 ℃, adding aminomethyl propanol into a stirring pot, starting stirring at the rotating speed of 500-1000 r/min, and uniformly stirring; and (3) cooling to about 37 ℃, adding the beta-alanyl hydroxypropionyl diaminobutyric acid benzylamine, the peony extract and the phenoxyethanol into a pot, stirring uniformly, cooling to room temperature, sampling, detecting, and discharging qualified materials.
1. Skin tightening efficacy test effect
40 healthy volunteers were selected, one group of two random persons. The 20 groups of volunteers used sleep masks containing the anti-aging compositions of examples 1-14, comparative examples 1-6, respectively. Subjects were administered 3 times a week for 4 weeks.
Evaluation of skin condition of the test volunteers after sample use was collected by means of a questionnaire to analyze the effect of product use. The main dimensions for collecting feedback include: skin elasticity, skin tightening sense, fine lines reduction and wrinkle lightening effect. The specific evaluation is shown by a scoring mode, 1-10 points are shown, and the higher the score is, the better the sample is favored by volunteers, and the better the effect is.
The average score for each group of individuals is shown in table 5.

As can be seen from Table 5, the anti-aging compositions of examples 1 to 14 were found to have an obvious anti-aging effect, and the anti-aging compositions of comparative examples 1 to 6 were found to have no obvious effect. This demonstrates that jojoba oil, seabuckthorn fruit oil, peony extract, beta-alanyl hydroxyproline diamido butyric acid benzylamine have significant synergy.
2. Skin elasticity test
20 healthy volunteers were selected and each of the essences containing the anti-aging compositions of examples 1 to 14 and comparative examples 1 to 6 was used. Taking skin at the inner side of the forearm as a test part, sampling 1 time a day, smearing corresponding products on the test part, and wiping off the liquid on the surface with a clean paper towel after 10 min. The skin elasticity characterization values R before and after the trial for 28d were determined according to the body Standard T/ZHCA 005-2019 cosmetic impact skin elasticity test method, respectively. The larger the characterization value R, the better the skin elasticity.
The calculation formula of the characterization value R is as follows: r=u1/U2;
wherein U1 is rebound displacement (mm) of skin after negative pressure is eliminated, and U2 is maximum stretching displacement (mm) of skin when negative pressure is loaded.

As is clear from Table 6, the skin elasticity characterization values were improved after the test of the essence containing the anti-aging compositions of examples 1 to 14, compared with comparative examples 1 to 6, and the difference in the skin elasticity characterization values R before and after the test was not less than 0.0132.
As can be seen from the test results of example 1 and comparative example 1, although the peony extract of comparative example 1 has a certain effect even though the total polyphenol concentration is lower than 10mg/mL, the effect is significantly inferior to that of example 1.
From the test results of example 2 and examples 11-14, it can be seen that there is a difference in the skin elasticity characterization effect of the anti-aging compositions containing different peony extracts or combinations thereof. This is because peony extract is not only rich in polyphenols, but also contains amino acids, fatty acids, vitamins, trace elements essential to human body, etc., and different volunteers have differences in product absorption, etc.
From the test results of example 6 and comparative examples 2 to 5, it can be seen that the anti-aging effect is greatly impaired by the absence of any component in the anti-aging composition of the example. The skin elasticity characterization value differences of comparative examples 2-5 are respectively improved by 0.0062, 0.0033, 0.0052 and 0.0062 relative to the skin elasticity characterization value difference of comparative example 6, and the calculated theoretical skin elasticity characterization value difference of example 6 is 0.0075 which is far lower than the actual skin elasticity characterization value difference 0.0274 obtained in example 6, which indicates that the jojoba oil, the sea buckthorn fruit oil, the peony extract and the beta-alanyl hydroxypropionyl diaminobutyric acid benzyl amine have obvious synergistic effect. 3. Anti-wrinkle effect test
(1) 30 healthy volunteers were selected, and the mask patches prepared from the mask solution containing the anti-aging composition of example 6 were used 3 times per week, respectively, for 4 weeks according to the product usage method; the skin state of the sample after use is evaluated by a questionnaire mode, and the use effect of the product is analyzed.
The analysis results are shown in Table 7.

Wherein, the effective rate is more than or equal to 50 percent, and the product is considered to be effective, and the effective rate is = (the number of people with obvious effect+the number of people with obvious effect)/the total number of people.
As can be seen from table 7, after the subjects used for 4 weeks, 90% or more of the subjects approved that the skin dryness was improved, the skin became more tender and finer, and the skin elasticity and firmness were improved; 86.67% of subjects found a reduction in fine lines with a better lightening effect. (2) One volunteer was selected to use an eye cream containing the anti-aging composition of example 1, and the subject was used 1 time per day and 4 weeks after use, and the subject was subjected to comparative analysis of the skin texture at the corners of the eyes using a VISIA-CR facial image analyzer.
Figures 1 and 2 are images of the texture of the canthus skin of a subject before and after use, respectively. Table 8 is an instrument analysis of the texture data of fig. 1 and 2. From the data of fig. 1,2 and table 8, the texture of the canthus was significantly improved after 4 weeks of use by the subject. This demonstrates that the anti-aging composition of example 1 has a pronounced fringing effect.

4. Safety test
60 volunteers were selected, and the ages and sexes of the volunteers were randomly determined to meet the standards of "cosmetic health Specification" (2007 edition). Volunteers were randomly divided into 60 groups, and eye creams, mask solutions and essence prepared in examples 1 to 14 and comparative examples 1 to 6 were tried out, respectively, 3 to 5g were applied to facial skin, and after 10 to 20 minutes, the skin was washed, and whether there was a reaction or a delayed reaction in use and within 30 minutes after use was observed. Whether the sample caused the stimulating reaction and the reaction degree was judged according to the identification standard of Table 9, statistical analysis was performed after recording, and the detection results are shown in Table 10.

As is clear from Table 10, the anti-aging compositions of examples 1 to 14 and comparative examples 1 to 6 were excellent in safety and did not cause adverse effects on the human body.
The embodiments of the present invention have been described in detail with reference to the accompanying drawings, but the present invention is not limited to the above embodiments, and various changes can be made within the knowledge of one of ordinary skill in the art without departing from the spirit of the present invention.
Claims (8)
1. An anti-aging composition comprising: peony extract, beta-alanyl hydroxyproline diaminobutyric acid benzylamine, sea buckthorn fruit oil and jojoba oil;
the preparation method comprises the following steps of: 0.5 to 5 parts of peony extract, 0.001 to 0.02 part of beta-alanyl hydroxypropionyl diaminobutyric acid benzylamine, 0.05 to 5 parts of sea buckthorn fruit oil and 0.05 to 0.5 part of jojoba oil;
the total polyphenol content of the peony extract is more than or equal to 10mg/mL.
2. The anti-aging composition according to claim 1, comprising, in parts by mass: 1 to 5 parts of peony extract, 0.001 to 0.02 part of beta-alanyl hydroxypropionyl diaminobutyric acid benzylamine, 1 to 5 parts of sea buckthorn fruit oil and 0.1 to 0.5 part of jojoba oil.
3. The anti-aging composition according to claim 1 or 2, wherein the peony extract comprises at least one of peony extract, peony water, peony/flower/leaf extract, peony root bark extract.
4. Use of an anti-aging composition according to any one of claims 1 to 3 in the preparation of a cosmetic.
5. The use according to claim 4, wherein the cosmetic is a lotion, an emulsion, a cream or a mask.
6. A cosmetic product, characterized in that it comprises an anti-ageing composition according to any one of claims 1 to 3 and a cosmetically acceptable auxiliary.
7. The cosmetic according to claim 6, wherein the anti-aging composition is 0.5 to 16% by mass based on the total mass of the cosmetic.
8. The cosmetic product according to claim 7, wherein the auxiliary material comprises at least one of an emollient, an emulsifier, a thickener, a humectant, a pH adjuster, a skin protectant, and a preservative.
