Abstract
本发明公开了一种生产5’‑胞苷酸的方法,属于基因工程和微生物工程领域。本发明通过基因敲除技术对大肠杆菌的胞苷单磷酸焦磷酸化酶基因ushA和核苷酸5'‑单磷酸核苷酶基因ppnN进行敲除,获得可高效积累5’‑胞苷酸(5’‑CMP)的大肠杆菌突变体。进一步通过过量表达尿苷激酶基因udk,获得高效生产5’‑CMP的重组大肠杆菌,5’‑CMP的发酵水平可达31.8 g/L以上。这种高效的大肠杆菌菌株可用于实现5’‑CMP的工业化生产,利于降低其生产成本及环境污染,实现绿色生物制造。

技术领域
本发明涉及一种生产5’-胞苷酸的方法,属于基因工程和微生物工程领域。
背景技术
核苷酸主要参与构成核酸,且具有重要的生物学功能。核苷酸及其衍生物可广泛应用于农业生产、食品、医药等领域。化学法合成核苷酸通常以三氯氧磷为磷酸供体,将核苷直接磷酸化生产相应核苷酸。但此种化学法用于合成5’-胞苷酸时,需要在核糖的2’,3’-羟基上加上保护基团,再进行磷酸化反应。化学法合成核苷酸操作步骤多、路线长、立体选择性差、试剂昂贵且试剂有毒、可操作性差、生产成本高,不适合大规模绿色生产。
核糖核酸(RNA)降解也可以用于生产5’-胞苷酸(5’-CMP),这种方法可以一次获得4种产物,除了5’-胞苷酸外,其他三种均为反应的副产物,需要经过进一步的分离纯化才能获得纯的5’-胞苷酸。同时,作为降解的原料-RNA,也存在诸多限制,如RNA的来源并不广泛,而且该降解RNA生产5’-胞苷酸工艺中的分离操作复杂,目标产物收率降低。
CN 111269870 A披露了一种高产胞苷酸的重组大肠杆菌及其应用,从胞苷酸生产菌株中克隆得到的胞苷激酶基因,重组菌经破碎后得到的粗酶液有着良好的催化活性和稳定性,加入胞苷、三磷酸腺苷(ATP)以及Mg2+,反应得到胞苷酸。但,该方法需要对重组菌进行破碎、再对酶进行分离,且产量不高。
发明内容
[技术问题]
本发明要解决的技术问题是现有技术中缺少能够一步高产5’-胞苷酸的重组菌株。
[技术方案]
本发明采用基因敲除技术对大肠杆菌基因组上胞苷单磷酸焦磷酸化酶基因ushA进行敲除,切断由5’-胞苷酸催化生产5’-胞苷的路径,使5’-胞苷酸获得积累;采用基因敲除技术对大肠杆菌基因组上核苷酸5'-单磷酸核苷酶基因ppnN进行敲除,切断由5’-胞苷酸催化生产胞嘧啶的路径,使5’-胞苷酸获得积累;并过量表达尿苷激酶基因udk,增强5’-胞苷至5’-胞苷酸的转化,可以高效生产5’-胞苷酸(5’-CMP),促进其在医药等领域中的应用。
本发明提供一种用于生产5’-胞苷酸的重组菌,是敲除大肠杆菌或枯草芽孢杆菌基因组上的胞苷单磷酸焦磷酸化酶基因ushA和核苷酸5'-单磷酸核苷酶基因ppnN,并过量表达尿苷激酶基因udk实现高效生产5’-胞苷酸(5’-CMP)。
进一步地,所述大肠杆菌选自:DH5α、BL21(DE3)、JM109、HB101;所述枯草芽孢杆菌选自:WB600、WB800。
进一步地,所述的重组菌,优选重组大肠杆菌BL21(DE3)。
进一步地,所述的重组菌,过量表达尿苷激酶基因udk时采用质粒pET22b、pET28a或pRSFDuet-1等进行游离表达,或将尿苷激酶基因udk整合至大肠杆菌基因组上进行整合表达。
本发明还提供应用所述重组菌生产5’-胞苷酸的方法,将重组菌株接种至含有卡那霉素的LB培养基中培育得到种子液;将种子液转接至发酵培养基中,在30-42℃和100-300 rpm条件下进行发酵培养,关联溶氧补料,当溶氧高于20-50%时进行补料,采用氨水控制pH中性;当发酵至菌浓OD600达15-28时,添加诱导剂IPTG进行诱导,同时降低培养温度至33℃。
进一步地,将重组菌株接种至含有卡那霉素的LB培养基中培育得到种子液,将种子液按照6%接种量,转接至含有卡那霉素的发酵培养基中,在37℃和200 rpm条件下进行发酵培养,30L发酵罐的初始装液量为13 L;发酵过程中,关联溶氧补料,当溶氧高于35%时进行补料,并采用氨水控制pH为7.0;发酵至菌浓OD600达15-28时,添加终浓度为0.6 mM的诱导剂IPTG进行诱导,同时培养温度降低为33℃。
进一步地,发酵培养基为:甘油30 g/L、氯化铁 8 mg/L、MgSO4 1 g/L、柠檬酸钠8mg/L、氯化钙150 mg/L、磷酸氢二钠3 g/L、磷酸二氢钠 3.0 g/L、氯化锌20 mg/L、酵母粉 6g/L、蛋白胨 5 g/L、微量元素溶液 10 mL;其中,微量元素溶液含有氯化铜21 g/L、硫酸锌18 g/L、钼酸钠25 g/L。
进一步地,补料培养基为:甘油600 g/L、蛋白胨6 g/L、酵母粉6 g/L。
[有益效果]
本发明采用基因敲除技术对大肠杆菌基因组上胞苷单磷酸焦磷酸化酶基因ushA进行敲除,切断由5’-胞苷酸催化生产5’-胞苷的路径,使5’-胞苷酸获得积累;采用基因敲除技术对大肠杆菌基因组上核苷酸5'-单磷酸核苷酶基因ppnN进行敲除,切断由5’-胞苷酸催化生产胞嘧啶的路径,使5’-胞苷酸获得积累;并过量表达尿苷激酶基因udk,增强5’-胞苷至5’-胞苷酸的转化,可以高效生产5’-胞苷酸(5’-CMP)。
采用本发明提供的敲除胞苷单磷酸焦磷酸化酶基因ushA和核苷酸5'-单磷酸核苷酶基因ppnN,并过量表达尿苷激酶的重组大肠杆菌,可以高效生产5’-胞苷酸,产量达到31.8 g/L,基本不积累中间代谢产物,有利于5’-胞苷酸的分离纯化。
本发明提供的重组大肠杆菌,能够以甘油为底物,一步发酵得到5’-胞苷酸,发酵上清液中5’-胞苷酸浓度高,便于分离纯化,工艺简单。

具体实施方式
胞苷酸的HPLC检测条件:液相色谱仪岛津 10A,色谱柱:INERTSIL ODS-SP 5μm4.6*250 mm,流动相:缓冲液配制:0.1 mol/L磷酸二氢钾水溶液:0.01 mol/L的四丁基氢氧化铵:甲醇=95:95:10,然后磷酸调节pH至4.5,波长:276 nm,流速:1.0 ml/min。
实施例1:用于生产5’-胞苷酸的重组大肠杆菌的构建方法
(1)构建用于过表达尿苷激酶基因的重组质粒pRSFDuet-udk
采用引物udk-FW: CATGCCATGGGCactgatcagtctcatcagtg(SEQ ID NO:7)和udk-RS: CGGGATCCTTATTCAAAGAACTGACTT (SEQ ID NO:8),以大肠杆菌基因组为模板,进行PCR扩增,获得udk目的片段(核苷酸序列如SEQ ID NO:5所示,编码的酶的氨基酸序列如SEQ IDNO:6所示)。PCR扩增的条件:预变性98℃,5 min;变性95℃,30 s;退火55℃,30 s;延伸72℃,1min;设置循环29次;再延伸72℃,10 min;最后16℃保温。PCR扩增的体系:5×PSbuffer 20μL,dNTP 10μL,上游/下游引物(10 μmol·L-1) 2μL,模板 1μL,酶1μL,水 66μL。采用限制性内切酶Nco I和BamH I酶切PCR获得的udk基因片段和pRSFDuet-1质粒。利用连接酶将酶切后的udk基因片段和pRSFDuet-1质粒进行连接,转化大肠杆菌JM109,涂布LB平板筛选获得阳性克隆,提取质粒进行基因测序验证正确,构建获得的重组质粒pRSFDuet-1-udk(图1C)。
(2)敲除胞苷单磷酸焦磷酸化酶基因ushA
采用基因敲除的手段,敲除大肠杆菌 BL21(DE3)基因组上的胞苷单磷酸焦磷酸化酶基因ushA(核苷酸序列如SEQ ID NO:1所示,编码的酶的氨基酸序列如SEQ ID NO:2所示),构建可高效积累5’-胞苷酸的重组菌株。
具体采用引物:ΔushA-1-FW:ATATGGTGAATATGGTCTGG (SEQ ID NO:9);ΔushA-1-RS:CTAGAAAGTATAGGAACTTCGGGTCGCCGCGATAATAATG(SEQ ID NO:10);ΔushA-2-FW: TTCTAGAGAATAGGAACTTCTGGATTGTGCAGGCGCATGA (SEQ ID NO:11);ΔushA-2-RS:ATTGATTACTTGACCGCCGT (SEQ ID NO:12),以大肠杆菌基因组为模板,通过融合PCR技术获得敲除胞苷单磷酸焦磷酸化酶基因ushA的敲除框ΔushA。采用引物Sg-ushA-FW: GTCCTAGGTATAATACTAGTCTGCATACCAATGATCATCAGTTTTAGAGCTAGAAATAGC (SEQ ID NO:13);pTarget-RS: CGGACTAGTATTATACCTAGGACTGAGC (SEQ ID NO:14),构建含有导向RNA的重组质粒pTarget。PCR扩增的条件:预变性98℃,5 min;变性95℃,30 s;退火55℃,30 s;延伸72℃(酶的延伸速度为1 kb/min,具体时间根据扩增片段的长度来设置);设置循环30次;再延伸72℃,10 min;最后16℃保温。PCR扩增的体系:5×PS buffer 20μL,dNTP 10μL,上游/下游引物(10 μmol·L-1)2μL,模板 1μL,酶1μL,水 66μL。将质粒pCas转化大肠杆菌 BL21(DE3),在抗性平板筛选获得重组大肠杆菌菌株。将pTarget和敲除框ΔushA电转化入含有质粒pCas的重组大肠杆菌菌株,在含有博来霉素和卡那霉素的抗性平板上筛选。将获得的单菌落,采用引物ΔushA-1-FW:ATATGGTGAATATGGTCTGG (SEQ ID NO:15)和ΔushA-2-RS:ATTGATTACTTGACCGCCGT (SEQ ID NO:16)进行菌落PCR验证,由电泳图可以看出(图2A),筛选获得的大肠杆菌A1-①(ΔushA)和A1-②(ΔushA)通过PCR获得的条带均比对照菌株小,说明胞苷单磷酸焦磷酸化酶基因ushA已经敲除,同时将该序列送至测序公司测序验证正确。
将构建获得的菌株A1-①采用LB培养基活化后,接种发酵培养基发酵培养,采用HPLC方法测定5’-胞苷酸的含量。发酵培养基为:甘油30g/L、氯化铁 8 mg/L、MgSO4 1 g/L、柠檬酸钠8 mg/L、氯化钙150 mg/L、磷酸氢二钠3 g/L、磷酸二氢钠 3.0 g/L、氯化锌20 mg/L、酵母粉 6 g/L、蛋白胨 5 g/L、微量元素 10 mL(含有氯化铜21 g/L、硫酸锌18 g/L、钼酸钠25 g/L。补料培养基:甘油600 g/L、蛋白胨6 g/L、酵母粉6 g/L。培养方法为:将菌株接种至含有卡那霉素的LB培养基中,37℃和200 rpm,培养8 h;将LB培养基中的种子液按照6%接种量,转接至含有卡那霉素的发酵培养基中(30L发酵罐,初始装液量为13 L),在37℃和200rpm条件下进行发酵培养,关联溶氧补料,当溶氧高于35%时进行补料。采用氨水控制pH为7.0。发酵至菌浓OD600达15-28时,添加诱导剂IPTG(终浓度为0.6 mM)进行诱导,同时培养温度降低为33℃。发酵结束后,离心除菌体,取上清液,采用HPLC测定其中的5’-胞苷酸的含量。通过测定发现,没有敲除ushA基因的对照菌株,几乎检测不到5’-胞苷酸的含量;敲除ushA基因的A1-①(ΔushA)菌株,可以检测到0.4 g/L的5’-胞苷酸的含量(图4)。
(3)在敲除胞苷单磷酸焦磷酸化酶基因ushA的基础上,进一步敲除ppnN
在步骤(2)构建的胞苷单磷酸焦磷酸化酶基因ushA敲除菌株的基础上,采用基因敲除的手段,继续敲除大肠杆菌基因组上的核苷酸5'-单磷酸核苷酶基因ppnN(核苷酸序列如SEQ ID NO:3所示,编码的酶的氨基酸序列如SEQ ID NO:4所示),构建可高效积累5’-胞苷酸的重组菌株。
具体采用引物:ΔppnN-1-FW:TGGATATGCTTAAACGCACCGC(SEQ ID NO:17);ΔppnN-1-RS:AGGATCAATGGTAAAACCTGAGCTCACGCAGGCCCAGCTG(SEQ ID NO:18);ΔppnN-2-FW: CAGCTGGGCCTGCGTGAGCTCAGGTTTTACCATTGATCCT(SEQ ID NO:19);ΔppnN-2-RS:CGTGCAGATTTCGTAGCAAGGGA(SEQ ID NO:20),以大肠杆菌基因组为模板,通过融合PCR技术获得敲除胞苷单磷酸焦磷酸化酶基因ppnN的敲除框ΔppnN。采用引物Sg-ppnN-FW: GTCCTAGGTATAATACTAGTCTGCATACCAATGATCATCAGTTTTAGAGCTAGAAATAGC (SEQ ID NO:21);pTarget-RS: CGGACTAGTATTATACCTAGGACTGAGC (SEQ ID NO:22),构建含有导向RNA的重组质粒pTarget。PCR扩增的条件:预变性98℃,5 min;变性95℃,30 s;退火55℃,30 s;延伸72℃(酶的延伸速度为1 kb/min,具体时间根据扩增片段的长度来设置);设置循环30次;再延伸72℃,10 min;最后16℃保温。PCR扩增的体系:5×PS buffer 20μL,dNTP 10μL,上游/下游引物(10 μmol·L-1) 2μL,模板 1μL,酶1μL,水 66μL。将质粒pCas转化大肠杆菌 A1-①(ΔushA),在抗性平板筛选获得重组大肠杆菌菌株。将pTarget和敲除框ΔppnN电转化入含有质粒pCas的重组大肠杆菌菌株,在含有博来霉素和卡那霉素的抗性平板上筛选。将获得的单菌落,采用引物ΔppnN-1-FW:GTGCTGGATTCACGCAGAAGGTT(SEQ ID NO:23)和ΔppnN-2-RS:CTGTGATGATATTTGTCGCGTGAG(SEQ ID NO:24)进行菌落PCR验证,由电泳图可以看出(图2B),筛选获得的大肠杆菌B1-①(ΔushA/ΔppnN)和B1-②(ΔushA/ΔppnN)通过PCR获得的条带均比对照菌株小,说明胞苷单磷酸焦磷酸化酶基因ppnN已经敲除,同时将该序列送至测序公司测序验证正确。
将构建获得的菌株B1-①采用LB培养基活化后,接种发酵培养基发酵培养,采用HPLC方法测定5’-胞苷酸的含量。采用步骤(2)中的发酵培养基和培养条件,进行发酵生产5’-胞苷酸。发酵结束后,离心菌体,取上清液,采用HPLC测定其中的5’-胞苷酸的含量。通过测定发现,敲除ppnN和ushA基因的B1-①(ΔushA/ΔppnN)菌株,可以检测到1.6 g/L的5’-胞苷酸的含量(图4)。
(4)构建重组大肠杆菌菌株ΔushA/ΔppnN+OE_udk
将步骤(1)构建获得的重组质粒pRSFDuet-udk转化入胞苷单磷酸焦磷酸化酶基因ushA敲除大肠杆菌菌株A1-①(ΔushA),涂布LB平板筛选获得阳性克隆重组大肠杆菌菌株ΔushA+OE_udk(图1A)。
将步骤(1)构建获得的重组质粒pRSFDuet-udk转化入胞苷单磷酸焦磷酸化酶基因ushA和核苷酸5'-单磷酸核苷酶基因ppnN双敲除的大肠杆菌菌株B1-①(ΔushA/ΔppnN),涂布LB平板筛选获得阳性克隆重组大肠杆菌菌株ΔushA/ΔppnN+OE_udk(图1B)。
将构建获得的重组大肠杆菌菌株ΔushA+OE_udk和ΔushA/ΔppnN+OE_udk分别进行摇瓶发酵培养,采用HPLC检测5’-胞苷酸的含量。采用步骤(2)中的发酵培养基和培养条件,进行发酵生产5’-胞苷酸。发酵结束后,离心菌体,取上清液,测定其中的5’-胞苷酸的含量。发酵结束后,离心菌体,取上清液,测定其中的5’-胞苷酸的含量。
通过测定发现,与步骤(2)中构建的敲除ushA基因但不过量表达尿苷激酶基因udk的菌株相比较,敲除ushA基因同时过量表达尿苷激酶基因udk的菌株ΔushA+OE_udk的5’-胞苷酸产量显著增加,5’-胞苷酸含量达6.1 g/L(图4)。同时,与步骤(3)中构建的敲除ppnN和ushA基因但不过量表达尿苷激酶基因udk的B1-①(ΔushA/ΔppnN)菌株相比较,敲除ppnN和ushA基因同时过量表达尿苷激酶基因udk的菌株ΔushA/ΔppnN+OE_udk的5’-胞苷酸产量显著增加,5’-胞苷酸含量达31.8 g/L,且杂质尿嘧啶、鸟苷的含量非常低(图4、图5)。
虽然本发明已以较佳实施例公开如上,但其并非用以限定本发明,任何熟悉此技术的人,在不脱离本发明的精神和范围内,都可做各种的改动与修饰,因此本发明的保护范围应该以权利要求书所界定的为准。

Claims (10)
1.一种用于生产胞苷酸的重组菌,其特征在于,是敲除大肠杆菌或枯草芽孢杆菌基因组上的胞苷单磷酸焦磷酸化酶基因ushA和/或核苷酸5'-单磷酸核苷酶基因ppnN,并过量表达尿苷激酶基因udk。
2.根据权利要求1所述的重组菌,其特征在于,所述大肠杆菌选自:DH5α、BL21(DE3)、JM109、HB101、MG1655。
3.根据权利要求1所述的重组菌,其特征在于,所述枯草芽孢杆菌选自:WB600、WB800、168。
4.根据权利要求1所述的重组菌,其特征在于,尿苷激酶选自大肠杆菌、枯草芽孢杆菌、无乳链球菌、莱氏无胆原体。
5.根据权利要求1或2所述的重组菌,其特征在于,在大肠杆菌中过量表达尿苷激酶基因udk时,采用质粒pET22b、pET28a或pRSFDuet-1进行游离表达。
6.根据权利要求1或2所述的重组菌,其特征在于,在大肠杆菌中过量表达尿苷激酶基因udk时,将udk整合至大肠杆菌基因组上进行整合表达。
7.根据权利要求1所述的重组菌,其特征在于,敲除大肠杆菌BL21(DE3)基因组上的胞苷单磷酸焦磷酸化酶基因ushA和核苷酸5'-单磷酸核苷酶基因ppnN,并利用pRSFDuet-1过量表达尿苷激酶基因udk。
8.应用权利要求1~7任一所述重组菌生产5’-胞苷酸的方法,其特征在于,将重组菌株接种至含有卡那霉素的LB培养基中培育得到种子液;将种子液转接至发酵培养基中,在30-42℃和100-300 rpm条件下进行发酵培养,关联溶氧补料,当溶氧高于20-50%时进行补料,采用氨水控制pH中性;当发酵至菌浓OD600达15-28时,添加诱导剂IPTG进行诱导,同时降低培养温度至33℃。
9.根据权利要求8所述的方法,其特征在于,发酵培养基为:甘油30 g/L、氯化铁 8 mg/L、MgSO4 1 g/L、柠檬酸钠8 mg/L、氯化钙150 mg/L、磷酸氢二钠3 g/L、磷酸二氢钠 3.0 g/L、氯化锌20 mg/L、酵母粉 6 g/L、蛋白胨 5 g/L、微量元素溶液 10 mL;其中,微量元素溶液含有氯化铜21 g/L、硫酸锌18 g/L、钼酸钠25 g/L。
10.根据权利要求9所述的方法,其特征在于,补料培养基为:甘油600 g/L、蛋白胨6 g/L、酵母粉6 g/L。
