in a first aspect, the invention provides a composition with anti-allergy, relieving and anti-inflammation effects, which is prepared from the following raw materials in parts by weight: 50-70 parts of glycerol, 20-40 parts of water, 1-10 parts of trehalose, 0.1-1 part of hyaluronic acid, 0.1-1 part of mannitol, 0.01-0.05 part of sodium chloride, 0.1-1 part of balsam pear fruit extract, 10.5-2 parts of oligopeptide, 21-5 parts of oligopeptide-51-5 parts of oligopeptide and 0.5-3 parts of 1, 2-hexanediol.
More preferably, the composition is prepared from the following raw materials in parts by weight: 55-65 parts of glycerol, 25-30 parts of water, 1-8 parts of trehalose, 0.1-0.5 part of hyaluronic acid, 0.1-0.5 part of mannitol, 0.01-0.03 part of sodium chloride, 0.1-0.8 part of bitter gourd fruit extract, 10.5-1.5 parts of oligopeptide, 21-3 parts of oligopeptide-51-3 parts of oligopeptide and 0.5-2 parts of 1, 2-hexanediol.
Most preferably, the composition is prepared from the following raw materials in parts by weight: 60 parts of glycerol, 27.3 parts of water, 5 parts of trehalose, 0.4 part of hyaluronic acid, 0.3 part of mannitol, 0.01 part of sodium chloride, 0.5 part of bitter gourd fruit extract, 11 parts of oligopeptide, 22 parts of oligopeptide, 52 parts of oligopeptide and 1.5 parts of 1, 2-hexanediol.
In a further embodiment, the major component of the extract of momordica charantia fruit is the momordica charantia protein MAP 30.

In a second aspect, the present invention provides a process for preparing a composition according to the first aspect, characterized in that: the method comprises the following steps:
step (1): weighing water according to the weight part of the first aspect, and adding the water into a reaction kettle;
step (2): weighing hyaluronic acid according to the weight part of the first aspect, adding into a reaction kettle while stirring, stirring for 30 minutes, and homogenizing for 15 minutes;
and (3): weighing trehalose, mannitol and sodium chloride according to the weight parts of the first aspect, adding into a reaction kettle while stirring, and stirring for 30 minutes;
and (4): weighing 1, 2-hexanediol according to the weight part of the first aspect, adding the 1, 2-hexanediol into a reaction kettle under stirring, and stirring for 15 minutes to fully stir;
and (5): under the condition that the temperature of a reaction kettle is lower than 40 ℃, weighing the oligopeptide-1, the oligopeptide-2, the oligopeptide-5 and the bitter gourd fruit extract according to the weight parts in the first aspect, adding the materials into the reaction kettle under stirring, and stirring for 30 minutes; and
and (6): and filtering the material by a pneumatic pump filtering device to obtain a transparent and clear final product.
Preferably, the stirring speed of step (2), step (3) and step (4) is 30 rpm, the homogenizing speed of step (2) is 1500 rpm, and the stirring speed of step (5) is 10 rpm.
In a third aspect, the present invention provides the use of a composition according to the first aspect above for the preparation of a cosmetic product having anti-allergy, soothing and anti-inflammatory effects.
Preferably, the cosmetic is a toner, lotion, essence, cream or mask.
It is to be understood that within the scope of the present invention, the above-described features of the present invention and those specifically described below (e.g., in the examples) may be combined with each other to form new or preferred embodiments. For reasons of space, they will not be described in detail.
Compared with the prior art, the invention has the following beneficial effects:
(1) the invention breaks through the traditional antiallergic therapy idea, combines the functions of resisting allergy, relieving inflammation and repairing skin barriers, and provides a composition which contains natural plant balsam pear fruit extract and trehalose and has strong synergistic anti-allergy, relieving and inflammation-diminishing effects.
(2) The composition of the invention can not generate drug resistance after long-term use, has no side effect, and is very safe and reliable.
Detailed Description
In the intensive research on the anti-allergy, relieving and inflammation-diminishing effects of the momordica charantia fruit extract, the inventor unexpectedly discovers that the effects of the momordica charantia fruit extract can be enhanced by adding a proper amount of trehalose through a large amount of screening, and develops a safe and efficient composition containing the momordica charantia fruit extract and the trehalose and having the anti-allergy, relieving and inflammation-diminishing effects. The present invention has been completed based on this finding.
For a better understanding of the present invention, the effects of the main raw and auxiliary materials in the composition of the present invention are further explained below.
"extract of Momordica charantia fruit" is a natural plant extract that can be used in cosmetics, as specified in the cosmetic specification list, and has anti-aging, anti-free radical, hair care, anti-inflammatory, and anti-bacterial effects. The product is mainly used as an antioxidant, an anti-inflammatory agent and a softening agent, is used in products for hair care, skin care and the like, and can effectively protect skin and hair and enable the skin and the hair to be moistened and glossy.
The present inventors have previously searched that "momordica charantia fruit extract" has been used for sedation and reduction of irritation, inflammation, edema and erythema in some korean and japanese cosmetic field patent applications. For example, KR10-2010-0048969 discloses a cosmetic product containing extract of the fruit of momordica charantia capable of packaging skin sedation and reducing irritation. JP Kokai 2002-212050 also discloses a cosmetic for skin or hair by formulating an extract from the fruit of Momordica charantia or a squeezed liquid of Momordica charantia with an alkali. However, the formulation of these cosmetics is quite different from the present invention, and the synergistic effect of the trehalose on the anti-allergy, soothing and anti-inflammatory effects of the momordica charantia fruit extract is discovered by the inventor for the first time, and no relevant report is reported before.
The balsam pear protein MAP30 is a protein extracted from balsam pear fruit, and has effect of resisting Human Immunodeficiency Virus (HIV). Human lung fibroblast cells are cultured in a culture solution containing MAP30, and after the herpes simplex virus is inoculated for 24-48 hours, virus antigens are checked, and the virus antigens are found that only 0.1-0.2 micromole/liter of MAP30 can have a useful pressing effect on the virus, while acyclovir (a common Sichuan anti-herpes simplex virus drug) has the requirements of 0.2 micromole/liter and 1.6 micromole/liter on the pressing concentration for different types of herpes simplex virus, which shows that MAP30 has a high-efficiency antiviral function and is very useful for treating herpes infection. However, the anti-allergy, soothing and anti-inflammatory effects of the momordica charantia protein MAP30 are discovered by the inventor for the first time, and no relevant report is reported before.
The "extract of Momordica charantia fruit" used in the cosmetic compositions of the invention may be a common "extract of Momordica charantia fruit" product commercially available. Preferably, the "extract of Momordica charantia fruit" which is prepared by a specific preparation method (for example, CN112760310A, or other methods) and contains "Momordica charantia protein MAP 30" as the main active ingredient is also possible. Most preferably, the "extract of Momordica charantia fruit" is pure "Momordica charantia protein MAP 30". Thus, more choices of different prices can be provided for different consumer populations by selecting to add different costs of "extract of Momordica charantia fruit" and "protein MAP 30" to the composition.
Trehalose is used in the pharmaceutical industry as a stabilizer for reagent and diagnostic agents. At present, various applications are discussed on the basis of functions and characteristics of trehalose such as non-reducibility, stability, protection of biomacromolecules, energy supply and the like. The trehalose is used for drying antibody such as vaccine, hemoglobin, virus and other bioactive substances, and the activity can be recovered after rehydration without freezing. The trehalose replaces blood plasma to serve as a biological product and a stabilizer, so that the biological product can be stored at normal temperature, and pollution can be prevented, thereby ensuring the storage, transportation and safety of the biological product. In the field of cosmetics, trehalose has a very strong moisturizing effect and physiological effects in various aspects such as sun protection, ultraviolet protection and the like, can be used as a humectant, a protective agent and the like to be added into emulsion, facial mask, essence and facial cleanser, and can also be used as a sweetening agent and a quality improver of lipstick, oral cleanser, oral aromatic and the like. The anhydrous trehalose can also be used in cosmetics as a dehydrating agent for phospholipids and enzymes, and the fatty acid derivatives thereof are also excellent surfactants. The inventors believe that trehalose has two main functions in the composition of the invention, moisturizing and stabilizing the activity of the extract of bitter melon fruits.
The invention is further illustrated with reference to specific examples. It should be understood that the specific embodiments described herein are illustrative only and are not limiting upon the scope of the invention.
The examples do not show the specific techniques or conditions, according to the technical or conditions described in the literature in the field, or according to the product specifications. The reagents or instruments used are conventional products which are not known to manufacturers and are available from normal sources.
The experimental procedures in the following examples are conventional unless otherwise specified. The test materials used in the following examples are all commercially available products unless otherwise specified. The "extract of Momordica charantia fruit" used in the examples is a common "extract of Momordica charantia fruit" product that is commercially available.
Preparation examples:
example 1
The composition is prepared from the following raw materials in parts by weight: 60 parts of glycerol, 27.3 parts of water, 5 parts of trehalose, 0.4 part of hyaluronic acid, 0.3 part of mannitol, 0.01 part of sodium chloride, 0.5 part of bitter gourd fruit extract, 11 parts of oligopeptide, 22 parts of oligopeptide, 52 parts of oligopeptide and 1.5 parts of 1, 2-hexanediol.
The preparation method of the composition comprises the following steps:
step (1): weighing water according to the weight parts, and adding the water into a reaction kettle;
step (2): weighing hyaluronic acid according to the weight parts, adding the hyaluronic acid into a reaction kettle under stirring at 30 revolutions per minute, stirring for 30 minutes, and homogenizing at 1500 revolutions per minute for 15 minutes;
and (3): weighing trehalose, mannitol and sodium chloride according to the weight parts, adding the weighed trehalose, mannitol and sodium chloride into a reaction kettle under the stirring of 30 revolutions per minute, and stirring for 30 minutes;
and (4): weighing 1, 2-hexanediol according to the weight parts, adding the 1, 2-hexanediol into the reaction kettle under stirring at 30 revolutions per minute, and stirring for 15 minutes to fully stir;
and (5): under the condition that the temperature of the reaction kettle is lower than 40 ℃, weighing oligopeptide-1, oligopeptide-2, oligopeptide-5 and bitter gourd fruit extract according to the weight parts, adding the materials into the reaction kettle under the stirring of 1O revolution/minute, and stirring for 30 minutes; and
and (6): the material is filtered by a pneumatic pump filter unit, and the filter bag is a 1 mu m PEG filter bag, so that a transparent and clear final product is obtained.
Example 2
The composition is prepared from the following raw materials in parts by weight: 58 parts of glycerol, 29.3 parts of water, 5 parts of trehalose, 0.8 part of hyaluronic acid, 0.6 part of mannitol, 0.01 part of sodium chloride, O.5 parts of balsam pear fruit extract, 11 parts of oligopeptide, 22 parts of oligopeptide, 52 parts of oligopeptide and 0.8 part of 1, 2-hexanediol.
The preparation method is the same as example 1.
Example 3
The composition is prepared from the following raw materials in parts by weight: 55 parts of glycerol, 32.3 parts of water, 5 parts of trehalose, 0.4 part of hyaluronic acid, 0.4 part of mannitol, 0.01 part of sodium chloride, 0.5 part of bitter gourd fruit extract, 11 parts of oligopeptide, 22 parts of oligopeptide, 52 parts of oligopeptide and 1.4 parts of 1, 2-hexanediol.
The preparation method is the same as example 1.
Example 4
The composition is prepared from the following raw materials in parts by weight: 60 parts of glycerol, 27.3 parts of water, 5 parts of trehalose, 0.4 part of hyaluronic acid, 0.3 part of mannitol, 0.01 part of sodium chloride, 300.5 parts of balsam pear protein MAP, 11 parts of oligopeptide, 22 parts of oligopeptide, 52 parts of oligopeptide-and 1.5 parts of 1, 2-hexanediol.
The preparation method is the same as example 1.
Comparative example 1
The composition is prepared from the following raw materials in parts by weight: 60 parts of glycerol, 32.8 parts of water, 0.4 part of hyaluronic acid, 0.3 part of mannitol, 0.01 part of sodium chloride, 11 parts of oligopeptide, 22 parts of oligopeptide, 52 parts of oligopeptide and 1.5 parts of 1, 2-hexanediol.
The procedure was as in example 1 except that the procedure involving no added components was omitted.
Comparative example 2
The composition is prepared from the following raw materials in parts by weight: 60 parts of glycerol, 27.8 parts of water, 5 parts of trehalose, 0.4 part of hyaluronic acid, 0.3 part of mannitol, 0.01 part of sodium chloride, 11 parts of oligopeptide, 22 parts of oligopeptide, 52 parts of oligopeptide and 1.5 parts of 1, 2-hexanediol.
The procedure was as in example 1 except that the procedure involving no added components was omitted.
Comparative example 3
The composition is prepared from the following raw materials in parts by weight: 60 parts of glycerol, 32.3 parts of water, 0.4 part of hyaluronic acid, 0.3 part of mannitol, 0.01 part of sodium chloride, 0.5 part of balsam pear fruit extract, 11 parts of oligopeptide, 22 parts of oligopeptide, 52 parts of oligopeptide and 1.5 parts of 1, 2-hexanediol.
The procedure was as in example 1 except that the procedure involving no added components was omitted.
Comparative example 4
The composition is prepared from the following raw materials in parts by weight: 60 parts of glycerol, 32.3 parts of water, 0.4 part of hyaluronic acid, 0.3 part of mannitol, 0.01 part of sodium chloride, 300.5 parts of balsam pear protein MAP, 11 parts of oligopeptide, 22 parts of oligopeptide, 52 parts of oligopeptide and 1.5 parts of 1, 2-hexanediol.
The procedure was as in example 1 except that the procedure involving no added components was omitted.
Effect embodiment:
skin barrier repair function test
The effects of the compositions described in examples 1, 4 and comparative examples 1 to 4 on epidermal cell proliferation, epithelial cell proliferation and fibroblast cell proliferation were examined, respectively, according to the method described in CN 105708724A.
(1) Effect on epidermal cell proliferation: skin cells were cultured ex vivo using cell culture techniques and the number of cells was determined after 48 hours incubation with the compositions of the invention. The experimental results show that the compositions described in examples 1, 4 and comparative examples 1-4 all improve cell proliferation.
(2) Effect on epithelial cell proliferation: epithelial cells were cultured in vitro using cell culture techniques and the number of cells was determined after 48 hours incubation using the compositions of the invention. The experimental results show that the compositions of examples 1 and 4 and comparative examples 1 to 4 are effective in stimulating epithelial cell proliferation.
(3) Effect on fibroblast proliferation: fibroblasts were cultured in vitro using cell culture techniques and the number of cells was determined after 48 hours of incubation with the compositions of the invention. The experimental results show that the compositions described in examples 1 and 4 and comparative examples 1 to 4 can effectively inhibit fibroblast proliferation.
Therefore, the compositions in the examples 1 and 4 and the comparative examples 1 to 4 can promote the rapid repair of skin damage parts, regulate fibroblast disorder, effectively inhibit scar formation, promote the skin to recover and smooth, rebuild the skin barrier, repair the epidermis and increase the blood circulation of skin capillaries. Furthermore, it can be seen from the above experimental results that the skin barrier repair function of the composition of the present invention may be related to the bioactive peptides oligopeptide-1, oligopeptide-2 and oligopeptide-5 contained in the composition.
Anti-allergy, soothing and anti-inflammatory action test
1. Test subjects:
210 skin allergic subjects, between 18 and 60 years of age, randomized into 7 groups (30 subjects per group): blank control, example 1, example 4, comparative 1, comparative 2, comparative 3 and comparative 4.
2. The treatment method comprises the following steps:
test emulsions were prepared by adding 10% by weight of the compositions described in example 1, example 4 and comparative examples 1-4 to a conventional emulsion base. The blank used only the emulsion base. The corresponding test emulsions were used for example 1, example 4, comparative example 1, comparative example 2, comparative example 3 and comparative example 4, respectively.
The composition is applied once in the morning and evening every day, and the allergic region of skin is cleaned before application. The first re-diagnosis is carried out after 1 day of continuous use, the second re-diagnosis is carried out after 3 days of continuous use, and the third re-diagnosis is carried out after 7 days of continuous use.
3. And (4) judging the standard:
(1) the effect is shown: the skin allergy such as red and swelling, pruritus and skin damage can be eliminated.
(2) The method has the following advantages: the skin allergy such as red swelling, pruritus and skin damage is reduced.
(3) And (4) invalidation: the skin is red and swollen, pruritus, skin damage and other allergic phenomena are not obviously changed.
(4) Weighting: the skin is red and swollen, itching, skin damage and other allergic phenomena.
4. The treatment results are as follows:
as can be seen from the results in table 1, the subjects in the blank control group had gradually increased allergic symptoms during the study period due to the lack of timely anti-allergic treatment. The group of comparative example 1 contained no trehalose and extract of Momordica charantia fruits, only the basic ingredient for repairing the skin barrier function, and therefore, the subjects of the group of comparative example 1 also exhibited a tendency of gradual increase in allergy during the study, with the symptoms of allergy being only slightly lighter than those of the blank control group. Comparative example 2 group contained trehalose in addition to the basic ingredients for skin barrier repair, but did not contain momordica charantia fruit extract. The group of subjects had slightly less allergic symptoms during the study than the blank and comparative example 1 groups, with a few subjects even appearing to be effective on days 1 and 3, but the composition of comparative example 2 was still ineffective for the majority of subjects.
Comparative example 3 and comparative example 4 groups contained bitter melon fruit extract or bitter melon protein MAP30 in addition to the basic ingredients for repairing skin barrier function, but did not contain trehalose. It is evident that the anti-allergic, soothing and anti-inflammatory effects of the composition are significantly enhanced after the addition of momordica charantia fruit extract or momordica charantia protein MAP30 to the composition, and during the study period, significantly more subjects appeared to be active than the group of comparative example 2, and it is seen that the anti-allergic effect of momordica charantia protein MAP30 is superior to that of momordica charantia fruit extract, but the compositions of comparative example 2 and comparative example 3 are still not effective for many subjects. Thus, the anti-allergy, soothing and anti-inflammatory effects of these two compositions are still less than ideal.
The groups of example 1 and example 4 contain bitter melon fruit extract or bitter melon protein MAP30 and trehalose in addition to the basic ingredients for repairing skin barrier function. As shown in Table 1, after adding a proper amount of trehalose, the anti-allergy relieving effect of the composition is remarkably enhanced, most of the subjects showed significant effect after continuous use for 1 day, and the subjects in the example 4 group showed significant effect from day 3. The results show that the anti-allergy, soothing and anti-inflammation effects of the bitter melon fruit extract can be synergistically enhanced by adding a proper amount of trehalose into the composition.

Therefore, the composition disclosed by the invention has the functions of promoting the rapid repair of skin damage parts, regulating fibroblast disorder, effectively inhibiting scar formation, promoting the skin to restore to be flat, rebuilding the skin barrier, keeping moisture for a long time, resisting allergy, relieving and diminishing inflammation, and is mild and non-irritant to the skin. In addition, more choices of different prices can be provided for different consumer populations by selecting to add different costs of "extract of Momordica charantia fruit" and "protein of Momordica charantia MAP 30" to the composition.
It will be apparent to those skilled in the art that various changes and modifications may be made in the present invention without departing from the spirit and scope of the invention. Thus, if such modifications and variations of the present invention fall within the scope of the claims of the present invention and their equivalents, the present invention is also intended to include such modifications and variations.
Claims (4)
1. A composition with anti-allergy, relieving and anti-inflammation effects is characterized in that: the composition is prepared from the following raw materials in parts by weight: 60 parts of glycerol, 27.3 parts of water, 5 parts of trehalose, 0.4 part of hyaluronic acid, 0.3 part of mannitol, 0.01 part of sodium chloride, 0.5 part of momordica charantia fruit extract, 11 parts of oligopeptide, 22 parts of oligopeptide, 52 parts of oligopeptide and 1.5 parts of 1, 2-hexanediol, wherein the momordica charantia fruit extract is momordica charantia protein MAP 30.
2. A process for preparing the composition of claim 1, wherein: the method comprises the following steps:
step (1): weighing water according to the parts by weight of the components in the claim 1, and adding the water into a reaction kettle;
step (2): weighing hyaluronic acid according to the weight portion of claim 1, adding into a reaction kettle while stirring, stirring for 30 minutes, and homogenizing for 15 minutes;
and (3): weighing trehalose, mannitol and sodium chloride according to the weight parts of claim 1, adding into a reaction kettle while stirring, and stirring for 30 minutes;
and (4): weighing 1, 2-hexanediol according to the weight part of claim 1, adding into a reaction kettle while stirring, and stirring for 15 minutes to fully stir;
and (5): weighing the oligopeptide-1, the oligopeptide-2, the oligopeptide-5 and the balsam pear fruit extract according to the weight part of the claim 1 under the condition that the temperature of the reaction kettle is lower than 40 ℃, adding the materials into the reaction kettle under stirring, and stirring for 30 minutes; and
and (6): filtering the material by a pneumatic pump filtering device to obtain a transparent and clear final product,
wherein, the stirring speed of the step (2), the step (3) and the step (4) is 30 r/min, the homogenizing speed of the step (2) is 1500 r/min, and the stirring speed of the step (5) is 10 r/min.
3. Use of the composition of claim 1 for the preparation of a cosmetic having anti-allergic, soothing and anti-inflammatory effects.
4. Use according to claim 3, characterized in that: the cosmetic is a toner, lotion, essence, cream or mask.
