Abstract
本发明属于脂肪酸提取制备技术领域,具体公开一种高纯度不饱和脂肪酸的提取工艺,是将油脂与醇在碱的催化下通过酯交换反应使油脂中的甘油三酯转换为单链的甲酯或乙酯型产物,再经分离纯化得到不饱和脂肪酸。本发明提供的高纯度不饱和脂肪酸的提取工艺,不但可有效提高不饱和脂肪酸的纯度,而且成本低、适合工业化生产,为工业化制备高纯度不饱和脂肪酸提供了一种新的工艺技术路线选择。
Description
技术领域
本发明属于脂肪酸提取制备技术领域,具体公开一种高纯度不饱和脂肪酸的提取工艺。
背景技术
不饱和脂肪酸是构成体内脂肪的一种脂肪酸,是人体不可缺少的脂肪酸。不饱和脂肪酸根据双键个数的不同,分为单不饱和脂肪酸和多不饱和脂肪酸二种。食物脂肪中,单不饱和脂肪酸有油酸等,多不饱和脂肪酸有亚油酸、亚麻酸、花生四烯酸等。人体不能合成亚油酸和亚麻酸,必须从膳食中补充。
不饱和脂肪酸在营养、医药和工业上具有重要的应用价值。不饱和脂肪酸能够通过减少炎症降低心脏疾病、中风风险;增加细胞膜的流动性,降低血液中低密度脂蛋白胆固醇(LDL)含量,减少血管中粥样硬化斑块形成而造成的堵塞血管,从而防止心律失常和减少高血压等。另外,部分不饱和脂肪酸已被证明能提高人体对胰岛素的敏感性,对糖尿病、代谢综合征有效,且没有明显的副作用。
现有的不饱和脂肪酸的提取工艺主要有两种:一种是把植物油脂通过加碱皂化使甘油三酯型不饱和脂肪酸转变为游离脂肪酸,然后通过尿素包埋、柱层析、分子蒸馏等技术进一步纯化得到游离的不饱和脂肪酸,该种方法加工成本高,且得到的产品质量不稳定,如易氧化变色;另外一种是采用超临界CO2萃取技术萃取甘油三酯型不饱和脂肪酸,然后通过冬化结晶使饱和脂肪酸更多的结晶被过滤掉,从而进一步提高不饱和脂肪酸含量,该种方法提取得到的不饱和脂肪酸的纯度不高,且加工成本较高。因此,本发明在于以较低成本提供一种高纯度不饱和脂肪酸的提取工艺。
发明内容
本发明提供一种高纯度不饱和脂肪酸的提取工艺,不但可以有效提高不饱和脂肪酸的纯度,而且成本低、适合工业化生产,为工业化制备高纯度不饱和脂肪酸提供了一种新的工艺技术路线选择。
本发明提供的高纯度不饱和脂肪酸的提取工艺,是将油脂与醇在碱的催化下通过酯交换反应使油脂中的甘油三酯转换为单链的甲酯或乙酯型产物,再经分离纯化得到不饱和脂肪酸。
优选地,具体包括以下步骤:
将无水乙醇、油脂及氢氧化钾按质量比为0.8-1.2:1:0.01-0.05混合,加热回流进行酯交换反应,收集得到的回流液回收乙醇后依次用热盐水水洗、脱水,得到乙酯型产物;
将所述乙酯型产物依次经模拟移动床色谱、减压精馏处理,即得不饱和脂肪酸。
优选地,使用无水甲醇代替无水乙醇,得到甲酯型产物。
进一步优选地,所述加热回流是在95℃下回流3-5h。
进一步优选地,所述热盐水的质量分数为3-5%。
进一步优选地,所述模拟移动床色谱处理是以粒径30-50μm的硅胶/丙烯酸酯聚合物为填料,以质量分数为95%乙醇为洗脱剂,在50-60℃下进行洗脱。
进一步优选地,所述精馏温度为150-200℃,真空度≦100Pa。
优选地,所述油脂选自澳洲坚果油、猫儿屎籽油、植物猫爪油、毛莱油、沙棘果油、蓝藻油、昆士兰果油、鱼油中的任一种。
优选地,所述不饱和脂肪酸为棕榈油酸乙酯、棕榈油酸甲酯、棕榈酸甲酯、棕榈酸乙酯、油酸甲酯、油酸乙酯、亚油酸甲酯、亚油酸甲酯乙酯、共轭亚油酸甲酯、共轭亚油酸乙酯、亚麻酸甲酯或亚麻酸乙酯中的任一种。
本发明还提供一种根据上述提取工艺得到的乙酯型棕榈油酸,所述乙酯型棕榈油酸的纯度≥95%。
对比现有技术,本发明的有益效果为:
1、本发明提供的高纯度不饱和脂肪酸的提取工艺,采用酯交换反应使油脂中甘油三酯转换为单链的甲酯或乙酯型产物,再通过模拟移动床色谱技术初步提高产物的纯度,最后通过高真空精馏技术使不饱和脂肪酸的纯度进一步提高;
2、以甲酯或乙酯型不饱和脂肪酸作为不饱和脂肪酸的载体,相较于游离型的不饱和脂肪酸,加工和储存过程中更稳定,而且产品的刺激性气味更低,便于后续产品的复配应用;且相较于甘油三酯型载体,更容易通过加工提高纯度;
3、生产成本低,易于大规模推广和应用。

具体实施方式
以下结合具体实施例对本发明做进一步详细描述,这些实施例用于理解而不是限制本发明的保护范围。
实施例1
一种高纯度不饱和脂肪酸的提取工艺,具体方法如下:
将澳洲坚果油100kg、无水乙醇92kg、KOH 1kg混合,在95℃下加热回流2h,得到的回流液经真空回收乙醇后,用质量分数为3%的热盐水水洗,在100℃下脱水,得到棕榈油酸粗品;
将上述棕榈油酸粗品经模拟移动床色谱分离纯化,且该模拟移动床色谱的填料为粒径30μm的硅胶/丙烯酸酯聚合物,洗脱剂为质量分数95%乙醇,洗脱温度设置为50℃;经模拟移动床分离后回收乙醇,在150℃、真空度为80Pa下减压精馏,得棕榈油酸产物,得率为6.86%,棕榈油酸含量为98.59%、酸价为0.5mgKOH/g、色泽为R0.1,Y3.0。
实施例2
一种高纯度不饱和脂肪酸的提取工艺,具体方法如下:
将鱼油100kg、无水乙醇120kg、KOH 5kg混合,在95℃下加热回流2h,得到的回流液经真空回收乙醇后,用质量分数为5%的热盐水水洗,在100℃下脱水,得到棕榈油酸粗品;
将上述棕榈油酸粗品经模拟移动床色谱分离纯化,且该模拟移动床色谱的填料为粒径40μm的硅胶/丙烯酸酯聚合物,洗脱剂为质量分数95%乙醇,洗脱温度设置为55℃;经模拟移动床分离后回收乙醇,在170℃、真空度为100Pa下减压精馏,得棕榈油酸产物,得率为5.06%,棕榈油酸含量为97.40%、酸价为0.4mgKOH/g、色泽为R0.1,Y5.0。
实施例3
一种高纯度不饱和脂肪酸的提取工艺,具体方法如下:
将沙棘果油100kg、无水乙醇80kg、KOH 1kg混合,在95℃下加热回流2h,得到的回流液经真空回收乙醇后,用质量分数为5%的热盐水水洗,在100℃下脱水,得到棕榈油酸粗品;
将上述棕榈油酸粗品经模拟移动床色谱分离纯化,且该模拟移动床色谱的填料为粒径50μm的硅胶/丙烯酸酯聚合物,洗脱剂为质量分数95%乙醇,洗脱温度设置为60℃;经模拟移动床分离后回收乙醇,在200℃、真空度为50Pa下减压精馏,得棕榈油酸产物,得率为3.98%,棕榈油酸含量为96.17%、酸价为0.4mgKOH/g、色泽为R0.1,Y4.0。
上述实施例1、2、3制备得到的产物的气相色谱图分别如图2、3、4所示,37种混合脂肪酸甲酯标准品气相色谱图如图1所示。检测方法依照GB5009.168-2016食品中脂肪酸的测定,检测仪器为日本岛津气相色谱仪Nexis GC-2030。
需要说明的是,本发明权利要求书中采用的步骤方法与上述实施例相同,为了防止赘述,本发明的描述了优选的实施例,但本领域内的技术人员一旦得知了基本创造性概念,则可对这些实施例作出另外的变更和修改。所以,所附权利要求意欲解释为包括优选实施例以及落入本发明范围的所有变更和修改。
显然,本领域的技术人员可以对本发明进行各种改动和变型而不脱离本发明的精神和范围。这样,倘若本发明的这些修改和变型属于本发明权利要求及其等同技术的范围之内,则本发明也意图包含这些改动和变型在内。
Claims (10)
1.一种高纯度不饱和脂肪酸的提取工艺,其特征在于,是将油脂与醇在碱的催化下通过酯交换反应使油脂中的甘油三酯转换为单链的甲酯或乙酯型产物,再经分离纯化得到不饱和脂肪酸。
2.根据权利要求1所述的高纯度不饱和脂肪酸的提取工艺,其特征在于,具体包括以下步骤:
将无水乙醇、油脂及氢氧化钾按质量比0.8-1.2:1:0.01-0.05混合,加热回流进行酯交换反应,收集得到的回流液回收乙醇后依次用热盐水水洗、脱水,得到乙酯型产物;
将所述乙酯型产物依次经模拟移动床色谱、减压精馏处理,即得不饱和脂肪酸。
3.根据权利要求2所述的高纯度不饱和脂肪酸的提取工艺,其特征在于,使用无水甲醇代替无水乙醇,得到甲酯型产物。
4.根据权利要求2或3所述的高纯度不饱和脂肪酸的提取工艺,其特征在于,所述加热回流是在95℃下回流3-5h。
5.根据权利要求2或3所述的高纯度不饱和脂肪酸的提取工艺,其特征在于,所述热盐水的质量分数为3-5%。
6.根据权利要求2或3所述的高纯度不饱和脂肪酸的提取工艺,其特征在于,所述模拟移动床色谱处理是以粒径30-50μm的硅胶/丙烯酸酯聚合物为填料,以质量分数为95%乙醇为洗脱剂,在50-60℃下进行洗脱。
7.根据权利要求2或3所述的高纯度不饱和脂肪酸的提取工艺,其特征在于,所述精馏温度为150-200℃,真空度≦100Pa。
8.根据权利要求1-3任一项所述的高纯度不饱和脂肪酸的提取工艺,其特征在于,所述油脂选自澳洲坚果油、猫儿屎籽油、植物猫爪油、毛莱油、沙棘果油、蓝藻油、昆士兰果油、鱼油中的任一种。
9.根据权利要求1-3任一项所述的高纯度不饱和脂肪酸的提取工艺,其特征在于,所述不饱和脂肪酸为棕榈油酸乙酯、棕榈油酸甲酯、棕榈酸甲酯、棕榈酸乙酯、油酸甲酯、油酸乙酯、亚油酸甲酯、亚油酸甲酯乙酯、共轭亚油酸甲酯、共轭亚油酸乙酯、亚麻酸甲酯或亚麻酸乙酯中的任一种。
10.一种根据权利要求1所述的提取工艺得到的乙酯型棕榈油酸,所述乙酯型棕榈油酸的纯度≥95%。
